Nitrogen loss from deserts in the southwestern United States 总被引:19,自引:6,他引:13
A lower limit for nitrogen loss from desert ecosystems in the southwestern United States was estimated by comparing nitrogen inputs to the amount of nitrogen stored in desert soils and vegetation. Atmospheric input of nitrogen for the last 10 000 years was conservatively estimated to be 2.99 kg N/m2. The amount of nitrogen stored in desert soils was calculated to be 0.604 kg N/m3 using extant data from 212 profiles located in Arizona, California, Nevada, and Utah. The average amount of nitrogen stored in desert vegetation is approximately 0.036 kg N/m2.Desert conditions have existed in the southwestern United States throughout the last 10 000 years. Under such conditions, vertical leaching of nitrogen below a depth of 1 m is small (ca. 0.028 kg N/m2 over 10 000 years) and streamflow losses of nitrogen from the desert landscape are negligible. Thus, the discrepancy found between nitrogen input and storage represents the amount of nitrogen lost to the atmosphere during the last 10 000 years. Loss of nitrogen to the atmosphere was calculated to be 2.32 kg N/m2, which is 77% of the atmospheric inputs.Processes resulting in nitrogen loss to the atmosphere from desert ecosystems include wind erosion, ammonia volatilization, nitrification, and denitrification. Aim
To evaluate how the establishment risk of freshwater fish species from the aquarium trade will change under a climate change scenario forecast for the year 2050.Location
North America.Methods
In order to estimate changes in the magnitude of risk across geography and across different species in the aquarium pathway, we considered an integrated approach to modelling the probability of establishment, which simultaneously included proxies of propagule pressure, environmental variables, species traits and interactions between environment and traits. We then used the parameters of our model to predict how the risk of establishment will change under a scenario of climate change forecast for the year 2050.Results
Our joint model performed better than submodels, suggesting that combining all components is worthwhile. The most predictive factors were precipitation, maximum temperature tolerance, maximum fish length and minimum temperature. Our joint model forecasted a 40% increase in the average risk of establishment by 2050 in the United States. In contrast to our expectations, the absolute establishment risk associated with this pathway remained very low for the entire suite of species in the aquarium trade in northern regions, such as Quebec, Canada. Instead, Florida, which has one of the highest current risks of establishment, was also forecasted to have the greatest absolute risk increase.Main conclusions
Our methodology for risk assessment allows invasive species management strategies to consider entire suites of species at a time and to forecast establishment risk for each species and location. While the aquarium pathway is likely to become more important for the United States, the Quebec government should prioritize other pathways of introduction in its exotic invasive species strategy. Our approach can be extended to be applied to different sets of species pertaining to the same or different pathways.124.

Plasmid engineering and molecular cloning is a virtually ubiquitous tool in biology. Although various methods have been developed for ligating DNA molecules or targeted mutagenesis of plasmids, each has its limitations. Many of the commonly used laboratory strategies are inefficient, while commercially available kits are quite costly and often specialized for highly specific circumstances. Here, we describe the SapI/AarI incision mediated plasmid editing (SIMPLE) method, which allows users to perform site-directed mutagenesis, deletions, and even short insertions into any plasmid in a single PCR reaction, using just one restriction enzyme. In addition, the SIMPLE method can be adapted to insert any sized DNA fragment into a vector using a two-step PCR approach, and can be used to ligate any number of DNA fragments with non-compatible ends in the specific order desired. The SIMPLE method provides researches an efficient and powerful tool with a broad range of applications for molecular cloning. 相似文献

In Spanish Legacies, Portes, Aparicio, and Haller offer the results of their longitudinal study on the assimilation of the children of immigrants in Spanish society. Thanks to their study design, which parallels the earlier Children of Immigrants Longitudinal Study conducted by Portes and Rumbaut, the authors are able to compare assimilation trajectories in Spain with those of second-generation youth in the United States. This comparison raises important considerations about how immigration policy shapes assimilation processes. More centrally, the contrast between the cases invites a deeper consideration of normative questions that not only undergird immigration policy but also shape the assimilation experiences of the second generation. The juxtaposition of the two cases also elicits provocations about how the sociological theories about assimilation might have been different if they had been developed based on the Spanish, rather than the American, experience, and how those Spanish-inflected theories might support different directions of inquiry. 相似文献

Isoxazolo[3,4-d] pyridazinones ([3,4-d]s) are selective positive modulators of the metabotropic glutamate receptors (mGluRs) subtypes 2 and 4, with no functional cross reactivity at mGluR1a, mGLuR5 or mGluR8. Modest binding for two of the [3,4-d]s is observed at the allosteric fenobam mGluR5 site, but not sufficient to translate into a functional effect. The structure activity relationship (SAR) for mGluR2 and mGluR4 are distinct: the compounds which select for mGluR2 all contain fluorine on the N-6 aryl group. Furthermore, the [3,4-d]s in this study showed no significant binding at inhibitory GABAA, nor excitatory NMDA receptors, and previously we had disclosed that they lack significant activity at the System Xc-Antiporter. A homology model based on Conn’s mGluR1 crystal structure was examined, and suggested explanations for a preference for allosteric over orthosteric binding, subtype selectivity, and suggested avenues for optimization of efficacy as a reasonable working hypothesis. 相似文献

Bone is a common site of metastasis for breast, prostate, lung, kidney and other cancers. Bone metastases are incurable, and substantially reduce patient quality of life. To date, there exists no small-molecule therapeutic agent that can reduce tumor burden in bone. This is partly attributed to the lack of suitable in vitro assays that are good models of tumor growth in bone. Here, we take advantage of a novel ex vivo model of bone colonization to report a series of pyrrolopyrazolone small molecules that inhibit cancer cell invasion and ex vivo tumor growth in bone at single-digit micromolar concentration. We find that the compounds modulated the expression levels of genes associated with bone-forming osteoblasts, bone-destroying osteoclasts, cancer cell viability and metastasis. Our compounds provide chemical tools to uncover novel targets and pathways associated with bone metastasis, as well as for the development of compounds to prevent and reverse bone tumor growth in vivo. 相似文献

In 1979 the US Fish and Wildlife Service published and adopted a classification of wetlands and deepwater habitats of the United States. The system was designed for use in a national inventory of wetlands. It was intended to be ecologically based, to furnish the mapping units needed for the inventory, and to provide national consistency in terminology and definition. We review the performance of the classification after 13 years of use. The definition of wetland is based on national lists of hydric soils and plants that occur in wetlands. Our experience suggests that wetland classifications must facilitate mapping and inventory because these data gathering functions are essential to management and preservation of the wetland resource, but the definitions and taxa must have ecological basis. The most serious problem faced in construction of the classification was lack of data for many of the diverse wetland types. Review of the performance of the classification suggests that, for the most part, it was successful in accomplishing its objectives, but that problem areas should be corrected and modification could strengthen its utility. The classification, at least in concept, could be applied outside the United States. Experience gained in use of the classification can furnish guidance as to pitfalls to be avoided in the wetland classification process.The US Government's right to retain a non-exclusive, royalty free licence in and to any copyright is acknowledged. 相似文献

Understanding the mechanisms underlying ecosystem resilience – why some systems have an irreversible response to disturbances while others recover – is critical for conserving biodiversity and ecosystem function in the face of global change. Despite the widespread acceptance of a positive relationship between biodiversity and resilience, empirical evidence for this relationship remains fairly limited in scope and localized in scale. Assessing resilience at the large landscape and regional scales most relevant to land management and conservation practices has been limited by the ability to measure both diversity and resilience over large spatial scales. Here, we combined tools used in large‐scale studies of biodiversity (remote sensing and trait databases) with theoretical advances developed from small‐scale experiments to ask whether the functional diversity within a range of woodland and forest ecosystems influences the recovery of productivity after wildfires across the four‐corner region of the United States. We additionally asked how environmental variation (topography, macroclimate) across this geographic region influences such resilience, either directly or indirectly via changes in functional diversity. Using path analysis, we found that functional diversity in regeneration traits (fire tolerance, fire resistance, resprout ability) was a stronger predictor of the recovery of productivity after wildfire than the functional diversity of seed mass or species richness. Moreover, slope, elevation, and aspect either directly or indirectly influenced the recovery of productivity, likely via their effect on microclimate, while macroclimate had no direct or indirect effects. Our study provides some of the first direct empirical evidence for functional diversity increasing resilience at large spatial scales. Our approach highlights the power of combining theory based on local‐scale studies with tools used in studies at large spatial scales and trait databases to understand pressing environmental issues. 相似文献
