Summary Natural senescence of the root cortex was assessed by nuclear staining, for cultivars and chromosome substitution lines of spring wheat known to differ in (1) susceptibility to common root rot, (2) total rhizosphere populations and (3) ability to support growth of a free-living nitrogen-fixing bacterium.Together, three root rot susceptible wheat lines showed significantly more cortical senescence than did three resistant lines; the susceptible lines also support larger rhizosphere populations. The wheat line that supports growth of a nitrogen-fixing bacterium showed significantly less cortical death than did any other line. Substitution of chromosome pairs 5B or 5D between the parent cultivars Rescue and Cadet substantially altered the amount of root cortex death, which is thus genetically determined. It is suggested thatCochliobolus sativus and other weak parasites benefit from early natural senescence of the root cortex, and that the degree of susceptibility or resistance of wheat lines to common root rot is at least partly determined by differences in cortical senescence.  相似文献

Pectobacterium and Dickeya spp. are soft rot Pectobacteriaceae that cause aggressive diseases on agricultural crops leading to substantial economic losses. The accurate, rapid and low‐cost detection of these pathogenic bacteria are very important for controlling their spread, reducing the consequent financial loss and for producing uninfected potato seed tubers for future generations. Currently used methods for the identification of these bacterial pathogens at the strain level are based mainly on molecular techniques, which are expensive. We used an alternative method, infrared spectroscopy, to measure 24 strains of five species of Pectobacterium and Dickeya. Measurements were then analyzed using machine learning methods to differentiate among them at the genus, species and strain levels. Our results show that it is possible to differentiate among different bacterial pathogens with a success rate of ~99% at the genus and species levels and with a success rate of over 94% at the strain level.  相似文献

Optical fiber sensors can offer robust and miniaturized detection of wideband ultrasound, yielding high sensitivity and immunity to electromagnetic interference. However, the lack of cost-effective manufacturing methods prevents the disseminated use of these sensors in biomedical applications. In this study, we developed and optimized a simple method to create optical cavities with high-quality mirrors for acoustic sensing based on micro-manipulation of UV-curable optical adhesives and electroless chemical silver deposition. This approach enables the manufacturing of ultrasound sensors based on Fabry-Pérot interferometers on optical fiber tips with minimal production costs. Characterization and high-resolution optoacoustic imaging experiments show that the manufacturing process yielded a fiber sensor with a small NEP () over a broad detection bandwidth (25 MHz), generally outperforming conventional piezoelectric based transducers. We discuss how the new manufacturing process leads to a high-performance acoustic detector that, due to low cost, can be used as a disposable sensor.  相似文献

Strawberry roots were sampled through the year to determine the populations and distribution of Pratylenchus penetrans and Meloidogyne hapla. Three strawberry root types were sampled—structural roots; feeder roots without secondary tissues; and suberized, black perennial roots. Both lesion and root-knot nematodes primarily infected feeder roots from structural roots or healthy perennial roots. Few nematodes were recovered from soil, diseased roots, or suberized roots. Lesion nematode recovery was correlated with healthy roots. In both 1997 and 1998, P. penetrans populations peaked about day 150 (end of May) and then declined. The decline in numbers corresponded to changes in total strawberry root weight and root type distribution. The loss of nematode habitat resulted from loss of roots due to disease and the transition from structural to suberized perennial roots. Meloidogyne hapla juvenile recovery peaked around 170 days (mid June) in 1997 and at 85, 147, 229, and 308 days (late March, late May, mid August, and early November, respectively) in 1998. There appear to be at least four generations per year of M. hapla in Connecticut. Diagnostic samples from an established strawberry bed may be most reliable and useful when they include feeder roots taken in late May.  相似文献

Disease progress of Stenocarpella macrospora (Earle) Sutton (syn. Diplodia macrospora Earle) was monitored on selected maize breeding lines over two seasons at three locations. Tagged plants were assessed at 10 day intervals for foliar lesions on a 1–9 scale and for ear rot on a 1–5 scale. The level of disease at a farm in Jos (West Africa Milk Company farm) was significantly greater than those at the other two locations (UTC Tenti and Saminaka), which had reduced or zero debris from the previous year's cultivation. There was a significant correlation between the leaf severity score and grain weight, where grain weight decreased with increasing leaf disease severity. However, no significant correlation was observed between ear rot and leaf severity. Varieties responded differently throughout the monitoring period. Late planting (1996) resulted in a significantly reduced disease incidence, but also resulted in lower grain weights. Spatial disease progress diagrams indicated that S. macrospora was initiated from random foci from which secondary spread occurred.  相似文献

  1. Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae) is a common polyphagous mite in tropical and subtropical areas and is considered as an important citrus pest.
  2. To understand the response of citrus to P. latus infestation, we have characterized the volatile profile and the molecular defence mechanisms of two citrus genotypes, namely sour orange (Citrus aurantium) and Cleopatra mandarin (Citrus reshni), to P. latus infestation. These two species are important rootstocks for the citrus industry and display differential resistance to Tetranychus urticae Koch (Acari: Tetranychidae), with sour orange showing elevated levels of constitutive and induced resistance associated with the jasmonic acid (JA) pathway compared with Cleopatra mandarin.
  3. P. latus infestation activated both the JA- and the salicylic acid-dependent pathways in sour orange but not in Cleopatra mandarin. However, this differential activation resulted in the production of similar volatile blends (a mixture of green leaf volatiles and aromatic compounds).
  4. Contrary to T. urticae infestation, sour orange supported larger densities of P. latus than Cleopatra mandarin with similar injury levels.
  5. Therefore, sour orange may be more tolerant to P. latus than Cleopatra mandarin.

In artificial inoculations of bean leaves (Phaseolus vulgaris), the minimum period of plant surface wetness (PSW) required for the start of infection was temperature dependent. Infection was most rapid at 20–25oC and no infection occurred at 5oC or 27oC. In experiments conducted in a glasshouse or controlled environment chamber, the influence of fluctuating temperatures was taken into account by calculating degree hours (oh) of PSW. A continuous period of PSW was not necessary for successful infection. Successive short periods of PSW were not strictly additive. A dry period following a wet period delayed the start of infection for longer than the duration of the dry period. The length of the delay depended on the preceding oh PSW and on the duration of the dry period. From this information, a model was constructed to predict when first infections could be expected. This model was tested in small plot field experiments in two seasons. The date on which infection was first noticed coincided closely with the date on which oh PSW first exceeded 1200. The use of this model in disease control is discussed.  相似文献

The cocoa tree, Theobroma cacao L., suffers large yield losses in Aceh Indonesia due to the disease black pod rot, caused by Phytophthora spp. Despite having the largest area under cacao production in Sumatra, farmers in the Aceh region have low overall production because of losses to insect pests and black pod rot. Trichoderma spp. were isolated from the roots and leaves of cacao trees and screened as potential biological control agents. Isolates used in the study were Trichoderma asperellum isolates T2 and T4, Trichoderma longibrachiatum isolates T15 and T16, and Trichoderma virens isolates T1 and Tv. T1, T2, T4, and Tv completely colonized and destroyed Phytophthora tropicalis and Phytophthora palmivora mycelium in precolonized plate assays. All six isolates reduced P. tropicalis, but none reduced the growth of P. palmivora in dual plate assays. Phytophthora growth was suppressed on MIN media amended with sterile heat inactivated Trichoderma culture filtrates, with Tv best suppressing growth of both Phytophthora spp. T. virens isolate Tv was the only isolate observed coiling around P. tropicalis mycelium and disrupted the formation of P. palmivora sporangia. Of all six isolates, only Tv reduced P. palmivora lesion expansion in a detached pod assay, reducing severity by 71%. Tv also reduced P. palmivora infection on seedlings when applied aerially at 1 × 106 and 1 × 108 conidia/ml, by 19% and 59%, respectively. T. virens isolate Tv is a mycoparasite, antagonizes Phytophthora in a dual plate assay, and shows antibiosis against Phytophthora spp., suggesting that multiple modes of action contribute to its ability to limit Phytophthora lesion expansion on cacao pods and seedlings.  相似文献

The present study investigated the potential of benzo-thiadiazole-7-carbothioic acid S -methyl ester (BTH) to protect postharvest melons var. 'Orange Flesh' from the fruit rot caused by Fusarium pallidoroseum . It was noticed that melon fruits immersed in BTH and postinoculated with the fungus presented the same pattern of disease incidence/severity and activity of the defence-related enzymes superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, phenylalanine ammonia-lyase, and β-1,3-glucanase of controls, indicating that BTH was ineffective in protecting melons from the fruit rot disease. However, the preflowering application of BTH in melon seedlings induced stunted growth, probably related to enhanced lignification which is related to the plant cell wall reinforcement and increase of resistance against invading pathogens, and alterations of the activity of the studied defence-related enzymes in comparison with controls, suggesting that this strategy could probably be effective for the control of the postharvest rot of melon fruits through activation of systemic resistance.  相似文献
