The insect baculovirus AcMNPV (Autographa californica multiple nuclear polyhedrosis virus) enters many mammalian cell lines, prompting its application as a general eukaryotic gene delivery agent, but the basis of entry is poorly understood. For adherent mammalian cells, we show that entry is favoured by low pH and by increasing the available cell-surface area through a transient release from the substratum. Low pH also stimulated baculovirus entry into mammalian cells grown in suspension which, optimally, could reach 90% of the transduced population. The basic loop, residues 268–281, of the viral surface glycoprotein gp64 was required for entry and a tetra mutant with increasing basicity increased entry into a range of mammalian cells. The same mutant failed to plaque in Sf9 cells, instead showing individual cell entry and minimal cell-to-cell spread, consistent with an altered fusion phenotype. Viruses grown in different insect cells showed different mammalian cell entry efficiencies, suggesting that additional factors also govern entry.  相似文献

The relation between the rate of increase in nonprotein nitrogenous compounds (NPN) of rabbit muscle and muscle pH ranging from 5.9 to 7.2 was examined during the post-mortem storage. Muscle of a high ultimate pH was prepared by the injection of ICH2COOH into the vein. The more the muscle pH kept away from 6.3, the more NPN increased. Therefore, it has been suggested that the post-mortem proteolysis is mainly attributed to the acid proteolytic system comprising cathepsins in muscles at a pH lower than 6.3 and to the neutral proteolytic system in muscles at a pH higher than 6.3.

The ratio of the increment of ninhydrin positive materials to that of Cu-Folin phenol reagent positive materials among NPN was relatively large in muscles at a high pH. This result has suggested that the neutral proteolytic system was more abound in exopeptidase activity than acid proteolytic system.  相似文献

A rapid and specific method is described for the determination of nitrate in meat and fishery products.

Nitrate separated from foods by extraction with 1/50Ν sodium hydroxide and ultrafiltration was readily reduced to nitrite by the use of respiratory nitrate reductase (NR) from Escherichia coli K-12. The nitrite so obtained can be determined by the specific diazotation-coupling reaction method.

The use of an enzymatic reaction resulted in quantitative reduction of nitrate, and the method was relatively free of interferences. Recoveries of 10 and 100 ppm of nitrate from 5 samples of meat and fishery products ranged from 92.8 to 97.8% for 10 ppm and 97.8 to 99.4% for 100 ppm with a detection limit of 0.5 ppm.  相似文献

The flavonoid constituents of defatted soybeans were studied. Two kinds of new natural isoflavones and four kinds of isoflavones were isolated by rechromatography on silica gel and Sephadex LH–20 columns of ethyl acetate extracts.

One of the new isoflavones was identified as 6″-O-acetyl daidzin, C23H22O10, by UV, IR, PMR and CMR, and the four kinds of isoflavones were also identified as daidzein, daidzin, genistein and genistin.

High performance liquid chromatography of the ethyl acetate and acetone extracts were carried out, and six main peaks corresponding to the six isoflavones were recognized.

Glycitein and glycitein-7-O-glucoside were not detected in the soybeans.  相似文献

A methanolic extract of Agrimonia eupatoria (Rosaceae) significantly attenuated glutamate-induced oxidative stress in HT22 hippocampal cells. A new flavonoid, characterized as kaempferol 3-O-β-D-(2″-O-acetyl-6″-(E)-p-coumaroyl)-glucopyranoside (2″-acetyl-tiliroside (1), was isolated from the methanolic extract of A. eupatoria stems together with nine known flavonoids. Compounds 4, 7, 8 and 9 all showed a neuroprotective effect on glutamate-induced toxicity in HT22 cells.  相似文献

Theoretical and experimental investigations were conducted on growth kinetics and substrate consumption of hydrogen bacterium Alcaligenes hydrogenophilus. The kinetic models proposed were evaluated by continuous cultures of A. hydrogenophilus under various conditions. A methanol-utilizing phototrophic bacterium, strain M402, was isolated from surface water of an acidic hot spring. The isolated strain was identified as Rhodopseudomonas acidophila from its morphological and physiological characters. Profiles of the utilization of non-aromatic compounds as carbon sources by this strain were in good agreement with those of some strains of R. acidophila reported by Pfennig [J. Bacteriol., 99, 597 (1969)]. However, strain M402 was found to be capable of utilizing vanillic acid, vanillin, vanillyl alcohol, ferulic acid, veratric acid, syringic acid, syringal-dehyde and benzyl alcohol as carbon sources under anaerobic-light conditions. Although Pfennig did not refer to these abilities of his strains, these notable characters of strain M402 seem to be additional new characters of R. acidophila.  相似文献

Changes in gene expression levels of c-myc and CD38 were examined during the differentiation of HL-60 cells to granulocytes due to three nicotinic acid-related compounds. CD38 expression was increased by isonicotinic acid and all-trans-retinoic acid (ATRA). Nicotinamide and nicotinamide N-oxide drastically decreased c-myc expression, but isonicotinic acid had no effect, suggesting that these compounds differentiate HL-60 to granulocytes through different pathways. These results should provide useful information as to the mechanisms of cell differentiation.  相似文献
