Lack of knowledge of the exact chemical structure of cephalosporin antigenic determinants has hindered clinical interpretation of adverse reactions to these drugs and delayed understanding of the mechanisms involved in the specific recognition and binding of IgE molecules to these antigenic determinants. We further resolve the relationship between structure and activity of proposed antigenic chemicals, including the rational design and synthesis of these haptenic structures. Comparative RAST inhibition studies of the synthesized molecules revealed that they were recognized by IgE antibodies induced by cephalosporin antibiotics. Thus, these data indicate that recognition is mainly directed to the acyl side chain and to the beta-lactam fragment that remains linked to the carrier protein in the cephalosporin conjugation course.  相似文献

Preparation and lipase-catalyzed enantiotope selective acetylation of the prochiral 2-acyloxypropane-1,3-diols (1a–h) including sulfonic ester (1a–c) and carboxylic ester (1d–h) series is described. A strong influence of the acyl moiety in these diols on the enantiotope selectivity of the porcine pancreatic lipase (PPL)-catalyzed reaction with vinyl acetate was observed. The best results were achieved with the 2-(4-methylbenzoyl)oxy- and cyclohexanecarbonyloxypropane-1,3-diols (1g and 1h) resulting in acetylated products (2g) of ≥98% e.e. in 77% yield and (2h) of 95% e.e. in 66% yield, respectively.  相似文献

Polyelectrolyte-coated microcapsules can be prepared by adsorption of polyions onto microcapsule surfaces in aqueous solutions under appropriate pH and ionic conditions. The resulting polyelectrolyte-coated microcapsules provide a promising tool for studying pH-induced configurational changes in polyions adsorbed onto hydrophobic membranes (capsule walls). An interesting application of polyelectrolyte-coated microcapsules is the pH-sensitive on/off control of microencapsulated enzyme reactions through alterations in the substrate permeability of the capsule wall by pH-conditioned configurational changes in the adsorbed polyion layer. This paper presents an overview of pH-induced conformational changes of polyelectrolytes in solutions, preparation of polyelectrolyte-coated microcapsules with an immobilized enzyme, and on/off control of the respective enzyme reactions by pH adjustment. Perovskite oxide ceramics attracts significant attention as a strong candidate of bifunctional oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) electrocatalyst for the metal‐air batteries. Numerous approaches to the viability of bifunctional perovskite electrocatalyst represent that the electro­chemical performance is highly correlated with defect chemistry, surface structure, and overall polycrystalline perovskite structure. By making use of the intrinsic flexibility of internal structure and high nonstoichiometry in perovskite oxide, the heat treatment effect of the complex Ba0.5Sr0.5CoxFe1‐xO3‐δ (x = 0.2 and 0.8) perovskites in argon atmosphere at 950 °C (Ar‐BSCF5582 and Ar‐BSCF5528) on the surface structure/defect chemistry and electrocatalytic performance is intensively investigated. Upon heat‐treatment in argon atmosphere, the amorphous thickness layer increases from ≈20 to 180–200 nm in BSCF5582, while there is little change in BSCF5528 with ≈20 nm. The electrocatalytic performance of BSCF5582 catalyst both in ORR and OER deteriorates seriously, while Ar‐BSCF5528 demonstrates a significant increase of electro­chemical performance in ORR. This study demonstrates that the electrochemical performances of a perovskite catalyst can be significantly determined by the simultaneous modification of both surface structure and internal defect chemistry, which are explained with transmission electron microscopy and atomic‐selective X‐ray absorption fine structure analyses, respectively.  相似文献

目的:探究培美曲塞+顺铂方案诱导化疗联合同期放化疗对局部晚期非小细胞肺癌的近期疗效及安全性。方法:收集我院肿瘤科2009年7月到2011年7月住院治疗的局部晚期非鳞癌非小细胞癌患者54例,按照随机数字表法分为研究组和对照组,每组27例,研究组给予培美曲塞+顺铂方案诱导化疗联合同期放化疗,对照组仅给予培美曲塞+顺铂方案化疗联合同期放疗,治疗2个周期,随访2年,对比两组患者的近期疗效及安全性。结果:研究组的近期有效率和控制率分别为55.56%、77.78%,高于对照组的50.33%和59.26%,有效率和控制率差异具有统计学意义(P0.05)。随访2年,研究组的中位生存期为12.77个月,1年生存率为48.15%,高于对照组中位生存期11.28个月,1年生存率40.74%,但差异无统计学意义(P0.05),研究组2年生存率为43.46%,高于对照组的25.38%,差异具有统计学意义(P0.05);研究组白细胞减少以及脱发发生率分别为37.0%、25.9%,低于对照组的81.5%和55.5%,差异具有统计学意义(均P0.05);研究组中血小板减少、血红蛋白减少、呕吐、肝功能损害、肾功能损耗、皮疹等发生率分别为40.7%、22.2%、59.2%、11.1%、7.4%、14.8%,高于对照组中各不良反应发生率分别为33.3%、18.5%、55.5%、14.8%、11.1%、7.4%,但两组间差异无统计学意义(P0.05)。结论:培美曲塞+顺铂方案诱导化疗联合同期放化疗对局部晚期非小细胞肺癌治疗效果明显,且不良反应少,可作为临床上晚期非小细胞肺癌化疗一线药物。  相似文献

In vivo redox activities in the apoplast of axenically cultured intact seedling roots (superoxide anion generation, and superoxide dismutase and peroxidase activities) in contact with the compatible arbuscular mycorrhizal fungus (AMF) were clearly attenuated in comparison with those in contact with the pathogenic fungus (PF) or treated with MeJA, even at the early stages of treatment. Contact of roots with the AMF did not enhance the biosynthesis of phenolic compounds (total phenolics, flavonoids, and phenylpropanoid glycosides), while contact with the PF significantly enhanced the biosynthesis of all phenolic fractions. Reactive oxygen and nitrogen species both seemed to be involved in these responses from the first moments of contact, but the fluorescence imaging of roots showed that ROS were mainly accumulated in the apoplast while NO was mainly stored in the cytosol. In conclusion, intact olive seedling roots clearly differentiated between AMF and PF.  相似文献

Administration of antivenoms to treat snakebite envenomings has the potential risk of inducing early adverse reactions. The mechanisms involved in these reactions are unclear. In this study, polyspecific antivenom consisting of whole IgG purified from equine plasma by caprylic acid precipitation was administered intravenously to non-envenomed horses (n = 47) and cows (n = 20) at a dose of 0.4 mL/kg. It has been reported that, in humans, this formulation (administered at a dose of 0.4 mL/kg) induces mild noticeable early adverse reactions, such as fever, vomiting, diarrhea, urticaria, generalized rash, tachypnea or tachycardia, in about 15–20% of the patients. Unexpectedly, none of the animals receiving antivenom in our study showed any evidence of early adverse reaction. Moreover, no late adverse reactions, i.e. serum sickness, were observed during 40 days after antivenom administration. Unlike studies performed in envenomed humans, our present results were obtained in a group of non-envenomed individuals. It is concluded that, in addition to the physicochemical characteristics of the formulation, other unknown factors must determine the occurrence of adverse reactions in snakebite envenomed humans treated with equine-derived antivenoms.  相似文献

In antioxidant competition assays, an antioxidant (A) and a detector compound (D) compete for a reactive species (R). In the evaluation of these assays, it is tacitly assumed that all of R is captured by either D or A. Due to the - by definition - high reactivity of R, unspecific reactions of R are likely to occur and neglecting these reactions will result in a systematic underestimation of antioxidant activity. It was shown that in the standard hydroxyl radical scavenging assay this was indeed the case; the inaccurate mathematical evaluation resulted in an underestimation of antioxidant activity of 25% in this competition assay. The systematic underestimation of antioxidant activity can be prevented by using an adjusted Stern-Volmer equation that takes into account that only part of R is captured by D or A.  相似文献

Stereoselective carbon–carbon bond formation with aldolases has become an indispensable tool in preparative synthetic chemistry. In particular, the dihydroxyacetone phosphate (DHAP)-dependent aldolases are attractive because four different types are available that allow access to a complete set of diastereomers of vicinal diols from achiral aldehyde acceptors and the DHAP donor substrate. While the substrate specificity for the acceptor is rather relaxed, these enzymes show only very limited tolerance for substituting the donor. Therefore, access to DHAP is instrumental for the preparative exploitation of these enzymes, and several routes for its synthesis have become available. DHAP is unstable, so chemical synthetic routes have concentrated on producing a storable precursor that can easily be converted to DHAP immediately before its use. Enzymatic routes have concentrated on integrating the DHAP formation with upstream or downstream catalytic steps, leading to multi-enzyme arrangements with up to seven enzymes operating simultaneously. While the various chemical routes suffer from either low yields, complicated work-up, or toxic reagents or catalysts, the enzymatic routes suffer from complex product mixtures and the need to assemble multiple enzymes into one reaction scheme. Both types of routes will require further improvement to serve as a basis for a scalable route to DHAP.  相似文献

Eicosanoids are a group of oxygenated fatty acid derivatives formed from C20 polyunsaturated fatty acids, including arachidonic and eicosapentaenoic acids. The potential of the coelomocytes of the starfish, Asterias rubens, to generate eicosanoids through the cyclooxygenase (COX) and lipoxygenase (LOX) pathways was investigated using reverse-phase high performance liquid chromatography, enzyme immunoassay and gas chromatography–mass spectrometry. The principal LOX product was identified as 8-hydroxyeicosatetraenoic acid (8-HETE) with 8-hydroxyeicosapentaenoic acid (8-HEPE) synthesised at significantly lower levels. No classical prostaglandins (PG), such as PGE2 or PGD2, were found to be generated by ionophore-challenged coelomocytes. Incubation of coelomocytes with lipopolysaccharides from either Escherichia coli or Salmonella abortus failed to induce an increase in generation of LOX products and the presence of 8-HETE (0–25 μM) had no significant effect on the in vitro phagocytic activity of Asterias coelomocytes. Neither indomethacin (a COX inhibitor) or esculetin (a LOX inhibitor) had any effect on the clearance of the bacterium, Vibrio splendidus, from the coelomic cavity of starfish suggesting that products of these enzymes are not involved in such coelomocyte responses to foreign particles.  相似文献
