AmB is an antifungal drug of polyene. Although it is prone to nephrotoxicity, it is still the gold standard in the clinical treatment of fungal infection. Sterol plays a decisive role in the drug activity of AmB. The antifungal activity of AmB depends on ergosterol in fungal membranes, and its toxicity is related to cholesterol in mammalian membranes. At the same time, AmB interacts with biofilms, leading to a significant loss of potassium ions and affecting the transport of potassium ions across membranes. Meanwhile, metal cation may also affect AmB molecules’ aggregation on the membrane. This paper mainly studied the effects of different concentrations of potassium ions on the interactions between AmB and lipid monolayers containing cholesterol or ergosterol and explored the differences in the impact of varying potassium ions on the drug activity of AmB on monolayers rich in these two kinds of sterols. The results show that potassium ions caused the collapse of lipid monolayer and lipid-AmB monolayer to disappear. The limiting molecular area of these monolayers also increased due to potassium ions. The limiting molecular area of the monolayer in the presence of ergosterol has a great difference in the different concentration of potassium ions, which is different from that in the presence of cholesterol. The presence of potassium ions, regardless of the intensity of K+ ions, increased the maximum elastic modulus of the lipid/sterol monolayer with and without AmB. The presence of potassium ions reduced the influence of AmB on the stability of the lipid monolayer containing cholesterol. The impact of AmB on the stability of the lipid monolayer containing ergosterol was related to the concentration of potassium ions. The potassium ions increased the area of the ordered “island” region on the lipid-AmB monolayer containing cholesterol, and the boundary of the microregion produced different degrees of curvature. However, on the lipid/ergosterol monolayer, 5 mM and 10 mM potassium ions made the holes caused by AmB more denser, and the diameter of holes become larger. These results can help to improve the effect of potassium ions on the transmembrane transport of substances affected by AmB. The results will provide a basis for further exploration of the effect mechanism of metal ions on the antifungal activity of polyene drugs.  相似文献

A frequently desired outcome when rehabilitating Zn toxic sites in Australia is to establish a self-sustaining native ecosystem. Hence, it is important to understand the tolerance of Australian native plants to high concentrations of Zn. Very little is known about the responses of Australian native plants, and trees in particular, to toxic concentrations of Zn. Acacia holosericea, Eucalyptus camaldulensis and Melaleuca leucadendra plants were grown in dilute solution culture for 10 weeks. The seedlings (42 days old) were exposed to six Zn treatments viz., 0.5, 5, 10, 25, 50 and 100 M. The order of tolerance to toxic concentrations of Zn was E. camaldulensis> A. holosericea> M. leucadendra, the critical external concentrations being approximately 20, 12 and 1.5 M, respectively. Tissue Zn concentrations increased as solution Zn increased for all species. Root tissue concentrations were higher than shoot tissue concentrations at all solution Zn concentrations. The critical tissue Zn concentrations were approximately 85 and 110 g g–1 DM for M. leucadendra, 115 and 155 g g–1 DM for A. holosericea and 415 and 370 g g–1 DM for E. camaldulensis for the youngest fully expanded leaf and total shoots, respectively. The results from this paper provide the first comprehensive combination of growth responses, critical external concentrations, critical tissue concentrations and plant toxicity symptoms for three important Australian genera, viz., Eucalyptus, Acacia and Melaleuca, for use in the rehabilitation of potentially Zn toxic sites.  相似文献

Fluorescence photobleaching recovery methods reveal marked changes in lateral mobilities of rabbit lymphocyte membrane components during the course of stimulation with succinyl concanavalin A (S Con A). The diffusion constant of S Con A receptors on T lymphocytes falls from 1.6×10?10 cm2/sec to 6.5×10?11 cm2/sec within 4 hr after stimulation, remains constant for 14 hr, and returns to its former value. The mobility of B cell receptors similarly falls from 1.4×10?10 cm2/sec to 5.5×10?11 cm2/sec but regains its unstimulated value much more slowly. In contrast, a fluorescent phospholipid analog shows constant mobilities of 1.9×10?8 cm2/sec and 1.5×10?8 cm2/sec in T and B cells, respectively, throughout the experiment.  相似文献

利用分子标记图谱对溶液培养与旱作培养(纸培养)下的水稻(Oryza sativa L.)幼苗的种子根与最长不定根长,不定根数,总根干重,根冠比等性状进行了基因定位与遗传分析。4种参数共检测到6个数量性状位点(quantitative trait loci,QTLs)与22对上位性互作位点,其中溶液培养中的最长不定根长,总根干重和旱作培养中的总根干重检测到的QTLs位点对总变异的贡献率分别为20%,23%和13%左右;旱作培养中的最长不定根长,不定根数,根冠比和溶液培养中的根冠比仅检测到上位性位点,对表型变异的贡献率在12%-61%之间,溶液培养与旱作条件下没有一个或一对检测到的QTL或互作位点完全相同,提示溶液培养和旱作条件下影响幼苗根系生长的遗传机制差异,上位性作用对旱作培养条件下的根生长具重要影响。  相似文献

Introduction

Type I cGMP-dependent protein kinase (PKGIα) belongs to the family of cyclic nucleotide-dependent protein kinases and is one of the main effectors of cGMP. PKGIα is involved in regulation of cardiac contractility, vasorelaxation, and blood pressure; hence, the development of potent modulators of PKGIα would lead to advances in the treatment of a variety of cardiovascular diseases. Aim: Representatives of ARC-type compounds previously characterized as potent inhibitors and high-affinity fluorescent probes of PKA catalytic subunit (PKAc) were tested towards PKGIα to determine that ARCs could serve as activity regulators and sensors for the latter protein kinase both in vitro and in complex biological systems. Results: Structure–activity profiling of ARCs with PKGIα in vitro demonstrated both similarities as well as differences to corresponding profiling with PKAc, whereas ARC-903 and ARC-668 revealed low nanomolar displacement constants and inhibition IC50 values with both cyclic nucleotide-dependent kinases. The ability of ARC-based fluorescent probes to penetrate cell plasma membrane was demonstrated in the smooth muscle tissue of rat cerebellum isolated arteries, and the compound with the highest affinity in vitro (ARC-903) showed also potential for in vivo applications, fully abolishing the PKG1α-induced vasodilation.  相似文献

To determine how elevated night temperature interacts with carbon dioxide concentration ([CO2]) to affect methane (CH4) emission from rice paddy soil, we conducted a pot experiment using four controlled‐environment chambers and imposed a combination of two [CO2] levels (ambient: 380 ppm; elevated: 680 ppm) and two night temperatures (22 and 32 °C). The day temperature was maintained at 32 °C. Rice (cv. IR72) plants were grown outside until the early‐reproductive growth stage and then transferred to the chambers. After onset of the treatment, day and night CH4 fluxes were measured every week. The CH4 fluxes changed significantly with the growth stage, with the largest fluxes occurring around the heading stage in all treatments. The total CH4 emission during the treatment period was significantly increased by both elevated [CO2] (P=0.03) and elevated night temperature (P<0.01). Elevated [CO2] increased CH4 emission by 3.5% and 32.2% under high and low night temperature conditions, respectively. Elevated [CO2] increased the net dry weight of rice plants by 12.7% and 38.4% under high and low night temperature conditions, respectively. These results imply that increasing night temperature reduces the stimulatory effect of elevated [CO2] on both CH4 emission and rice growth. The CH4 emission during the day was larger than at night even under the high‐night‐temperature treatment (i.e. a constant temperature all day). This difference became larger after the heading stage. We observed significant correlations between the night respiration and daily CH4 flux (P<0.01). These results suggest that net plant photosynthesis contributes greatly to CH4 emission and that increasing night temperature reduces the stimulatory effect of elevated [CO2] on CH4 emission from rice paddy soil.  相似文献

Various fusion proteins from eukaryotes and viruses share structural similarities such as a coiled coil motif. However, compared with eukaryotic proteins, a viral fusion protein contains a fusion peptide (FP), which is an N-terminal hydrophobic fragment that is primarily involved in directing fusion via anchoring the protein to the target cell membrane. In various eukaryotic fusion proteins the membrane targeting domain is cysteine-rich and must undergo palmitoylation prior to the fusion process. Here we examined whether fatty acids can replace the FP of human immunodeficiency virus type 1 (HIV-1), thereby discerning between the contributions of the sequence versus hydrophobicity of the FP in the lipid-merging process. For that purpose, we structurally and functionally characterized peptides derived from the N terminus of HIV fusion protein - gp41 in which the FP is lacking or replaced by fatty acids. We found that fatty acid conjugation dramatically enhanced the capability of the peptides to induce lipid mixing and aggregation of zwitterionic phospholipids composing the outer leaflet of eukaryotic cell membranes. The enhanced effect of the acylated peptides on membranes was further supported by real-time atomic force microscopy (AFM) showing nanoscale holes in zwitterionic membranes. Membrane-binding experiments revealed that fatty acid conjugation did not increase the affinity of the peptides to the membrane significantly. Furthermore, all free and acylated peptides exhibited similar α-helical structures in solution and in zwitterionic membranes. Interestingly, the fusogenic active conformation of N36 in negatively charged membranes composing the inner leaflet of eukaryotic cells is β-sheet. Apparently, N-terminal heptad repeat (NHR) can change its conformation as a response to a change in the charge of the membrane head group. Overall, the data suggest an analogy between the eukaryotic cysteine-rich domains and the viral fusion peptide, and mark the hydrophobic nature of FP as an important characteristic for its role in lipid merging.  相似文献

Leukocyte cell surface sialyl Lewis x (sLex) and related epitopes play an important role in cell rolling and adhesion during diapedesis via interaction with E-selectin. Here, we present evidence that Mac-1 (CD11b/CD18, CR-3) is a major neutrophil glycoprotein decorated with sLex and ligation of these carbohydrate moieties by anti-sLex antibody significantly impairs neutrophil functions. First, Western blot analysis shows that both CD11b and CD18 subunit of purified Mac-1 are decorated with sLex moieties. A significant co-localization of CD11b and sLex moieties is observed at neutrophil secondary granules. With stimulation of formyl-Met-Leu-Phe (fMLP), neutrophil surface labeling with anti-sLex antibody follows an identical up-regulation pattern of Mac-1. Second, protein-binding assays indicate that sLex moieties on Mac-1 are critical for binding interaction of Mac-1 to E-selectin. Removal of sLex moieties completely abolishes Mac-1-E-selectin binding. Finally, ligation of Mac-1 sLex by anti-sLex antibody induces a significant degranulation of neutrophil secondary granules at the absence of chemoattractant stimulation. This “dysregulated” degranulation induced by anti-sLex antibody strongly inhibits neutrophil transmigration in response to fMLP. In summary, Mac-1 sLex moieties play a critical role in regulating β2 integrin functions during neutrophil transmigration and degranulation.  相似文献

This study was aimed at enhancing the physical stability of the drug clotrimazole (CT) and the polymer contained within hot-melt extrusion (HME) films using polymer blends of hydroxypropyl cellulose (HPC) and poly(ethylene oxide) (PEO). The HME films were investigated for solid-state characteristics, moisture sorption, bioadhesivity, mechanical properties, glass transition temperature, release characteristics, and physical and chemical stability of the drug and the polymer within the HME films. The solid-state characterization of the drug and the polymer was performed using differential scanning calorimetry, x-ray diffractometry, and dynamic mechanical analysis. A texture analyzer was used to study the bioadhesive and mechanical properties of the HME films. The physical and chemical stability of the films, stored at 25°C/60% relative humidity or in a desiccator, was studied for up to 12 months. CT was found to be in solid solution within all of the formulations extruded. The physical stability of the drug and PEO in the HME films increased with increasing HPC concentration, but the bioadhesivity and flexibility of the PEO films decreased with increasing HPC concentration. Films containing HPC: PEO∶CT in the ratio of 55∶35∶10 demonstrated optimum physical-mechanical, bioadhesive, and release properties. In conclusion, polymer blends of HPC and PEO were used successfully to tailor the drug release, mechanical and bio-adhesive properties, and stability of the HME films.
