目的研究补光对灰仓鼠(Cricetulus migratorus)越冬时脏器器官的影响。方法室温下将其分为自然光照组和补充光照组,分别在第0、30、48和150天时测定动物的体重和脏器器官的重量,应用涂片法检查精子的数量。结果自然光照时,精子数量在冬至时变化不大,但冬至后第18天,灰仓鼠的睾丸系数降到最低值,精子数量仅为422枚,第150天时恢复到正常繁殖状态,精子数量达到7440枚。其他脏器指数在冬至时最高,第150天时最低。补充光照后,雌雄鼠体重高峰值时间均比自然光照组提前100 d,各器官指数变化与体重变化情况相类似,但睾丸和子宫系数维持稳定,精子数量未发现减少。结论在自然光照时越冬灰仓鼠的脏器指数逐渐下降,第150天时脏器指数达至最高值,睾丸指数在冬至后第18天下降到最低值,处于非繁殖状态。补充光照后,越冬灰仓鼠的精子数量和子宫指数变化不大,一直维持繁殖状态。 相似文献

Many membrane proteins exist and function as oligomers or protein complexes. Routine analytical methods involve extraction and solubilization of the proteins with detergents, which could disturb their actual oligomeric state. AcrB is a trimeric inner membrane multidrug transporter in E. coli. In previous studies, we created a mutant AcrBP223G, which behaves like a monomer when extracted from the cell membrane. However, the actual oligomeric state of AcrBP223G in cell membranes remained unclear, which complicated the interpretation of the mechanism by which the mutation affects function. Here we used several complementary methods to determine the oligomeric state of AcrBP223G in E. coli cell membranes. Two sets of quantitative fluorescent techniques were exploited. For these, we created fluorescent tagged AcrB, AcrB-CFP and AcrB-YPet. Fluorescence resonance energy transfer (FRET) and fluorescence recovery after photobleaching (FRAP) were employed to characterize independently the efficiency of energy transfer between co-expressed AcrB-CFP and AcrB-YPet, and the diffusion coefficient of AcrB-YPet and AcrBP223G-YPet in live E. coli cells. Second, we introduced Cys pairs at the inter-subunit interface and used controlled oxidation to probe inter-subunit distances. The results from all studies converge on the conclusion that AcrBP223G exists as a trimer in cell membranes, which dissociates during the purification steps. The small change in trimer affinity and structure leads to a significant loss of AcrB activity. In addition, throughout this study we developed protocols and established benchmark values, useful for further studies on membrane protein associations in cell membranes. 相似文献

The present study scrutinised how far temperature would affect the velocity of the insecticidal activity of Bacillus thuringiensis, as the rapidity of pest control achievements is of a great concern. Third instar Spodoptera littoralis larvae were treated with Bt at three concentration levels under five different temperatures (15°C, 20°C, 25°C, 30°C and 35°C). LT50s were evaluated in each case. The LT50 values showed various levels of reductions as temperature and/or Bt concentration increased, indicating that the velocity of mortality (1/LT50) and/or the rapidity of Bt activity was almost temperature dependant. However, relatively high and low reduction percentages in the LT50 values on the elevation of 5°C were obtained at lower and higher temperature ranges, respectively. The temperature coefficient, Q 10 values, determined within narrow ranges (5°C) showed great reductions when temperature increased from 15°C to 20°C at all Bt concentrations. Raising temperature by 5°C above 20°C or 25°C almost caused similar Q 10 values indicating constant increase in the response of Bt activity within 20–30°C temperature range. Q 10 values over 30°C were comparatively very low. This proved that decrease in Q 10 values due to the rise of temperature was dependant on the starting temperature. 相似文献

This paper evaluates the relative performance and competitive ability of seven legumes from the Iberian Peninsula, Spain (one already invasive in Australia), grown singly or pairwise with seven legumes from south‐western Australia in their own soils. Indices of growth used were root, shoot and total mass and shoot dimensions. Water content, xylem water potential during drying and nodule production were also measured. The index of competition used was the intensity of competition coefficient (ICC), which is suitable for additive designs. The Australian species usually grew larger than the Spanish species, in their presence or absence, although the results were sensitive to the index of growth or resource capture used. The Australian legumes usually possessed greater total water content than the co‐occurring Spanish legumes. At least five Australian species were less reduced or not affected at all by inter‐regional competition compared with growth in isolation. Two Australian legumes performed relatively poorly in the presence of at least four Spanish legumes. The ICC identified many instances of growth promotion (facilitation) of the Australian species, which we attribute to extra nitrogen obtained through enhanced nodulation of both cohabiting species. Five of the seven Spanish species were outcompeted by the Australian species, whereas the growth of two was sometimes facilitated. Although the Spanish legumes had greater competitive ability in the more fertile Australian soils, they remained competitively inferior to the Australian legumes in their first season of growth and none is likely to displace the native legumes at this stage of growth. 在大陆青冈分布区东缘的大金山岛种群中按不同坡向取样,旨在揭示相对均一条件下青冈种子的形态变异及变异的来源,以从不同侧面研究环境对形态变异的作用。大金山岛种群青冈种子的长度、直径和重量分别为1.557±0.128cm、1.175±0.112cm、1.321±0.310g,处于青冈种子正常分布范围内。单颗种子的长度、直径符合正态分布,而重量则明显偏离正态分布。不同坡向青冈种子间差异很小,表明微地形的影响较小;种群变异的来源株内和株间约各占一半,说明种子表型具有一定的可塑性,主要由于种子处于植株不同部位的缘故。 相似文献

A simple method for calculating the productivity of chromatography processes was proposed based on the iso-resolution curve concept. The model separation system was polyphenol separations by polystyrene divinylbenzene resins with the ethanol–water mixture mobile phase. The distribution coefficient K was determined as a function of ethanol concentration I by linear gradient elution experiments. The HETP-mobile phase velocity u curves were determined as a function of I. Using K and HETP, the iso-resolution curve was calculated, from which the productivity was determined as a function of I. It was found that there is an optimum I, where the highest productivity with the minimum amount of mobile phase consumption is obtained. 相似文献

以云南亚热带南部沿温度带分布的35个表土孢粉样品和7个植被样方调查为基础,计算了代表性孢粉类型的百分比和孢粉浓度,及常见孢粉的R值,并分析了影响R值的因素;同时,研究了植物群落和样方内孢粉组合的相似度。结果表明,云南亚热带南部表土样品孢粉产量丰富,蕨类植物孢子产量较高,具有典型的亚热带特征,且山地垂直分异显著;木本、草本植物花粉基本代表了区域内乔木和草本植物特征,蕨类植物孢子则具有超代表性;表土孢粉组合与植物群落间的相似系数绝大部分都在70%以上,表土孢粉组合基本上可以反映植物群落面貌;表明云南亚热带地区表土孢粉与现代植被之间具有较好的对应关系,这对在该区利用化石孢粉资料定量恢复古植被和重建古气候有重要意义。 相似文献
