Human urinary erythropoietin was adsorbed to phytohaemagglutinin coupled to agarose or porous glass and quantitatively eluted by a saturated solution of MgCl2. This method provides a means of separating erythropoietin from several of its contaminants, presumably on the basis of its carbohydrate side chains. Erythropoietin which had been purified by chromatography on insolubilized phytohaemagglutinin was sufficiently free of toxicity to be assayable in tissue culture even when crude urine was used as a starting material.  相似文献

We analyze data on the height and weight of mothers and newborn babies between 1980 and 2005 in St. Petersburg, Russia. We find that women's living standards, as measured by their height, improved steadily from the end of World War II through those born in 1972, hence reached adulthood in 1990. Thereafter, heights declined. Evidence on both the length and weight of babies corroborates this pattern. Their values trace a "U" shaped curve with troughs near the mid-1990s. Thus, the anthropometric results on newborn as well as for their mother point to the strains and challenges to living standards experienced during the restructuring of the post-Soviet economy. This is a general result that has become a recurring pattern: economic transitions are almost always accompanied by biological strains.  相似文献

The acid unfolding of staphylococcal nuclease (SNase) is very cooperative (Whitten and García-Moreno, Biochemistry 2000;39:14292-14304). As many as seven hydrogen ions (H+) are bound preferentially by the acid-unfolded state relative to the native (N) state in the pH range 3.2-3.9. To investigate the mechanism of acid unfolding, structure-based pKa calculations were performed with a variety of continuum electrostatic methods. The calculations reproduced successfully the H+ binding properties of the N state between pH 5 and 9, but they systematically overestimated the number of H+ bound upon acid unfolding. The calculated pKa values of all carboxylic residues in the N state were more depressed than they should be. The discrepancy between the observed and the calculated H+ uptake upon acid unfolding was not improved by using high protein dielectric constants, structures relaxed with molecular dynamics, or other empirical modifications implemented previously by others to maximize agreement between measured and calculated pKa values. This suggests an important role for conformational fluctuations of the backbone as important determinants of pKa values of carboxylic groups. Because no global or subglobal conformational changes have been observed previously for SNase under acidic conditions above the acid-unfolding region, these fluctuations must be local. The acid unfolding of SNase does not seem to involve the disruption of the N state by accruement of intramolecular repulsive interactions, nor the protonation of key ion paired carboxylic residues. It is more consistent with modest contributions from many H+ binding groups, with an important role for local conformational fluctuations in the coupling between H+ binding and the global structural transition.  相似文献

Nutrients in exotic species and invaded communities play a key role in determining the dynamics of invaders and the invasibility of a receipt community. This study focused on the effects of the native holoparasite Cuscuta campestris (for short Cuscuta) on nutrients in the exotic invasive Mikania micrantha (for short Mikania) and stands invaded by Mikania. We conducted a set of field investigations on Mikania with Cuscuta parasitism for 1–4 years, and measured soil properties, community composition, and the growth and nutrient content of Mikania and Cuscuta in two types of sub-communities (i.e. with Mikania only, or with Mikania and Cuscuta). Cuscuta dramatically reduced the cover, biomass, and nutrients (i.e. N, P, and K content) of Mikania, significantly enhanced soil water, pH and nutrient content (i.e. organic matter, total N and P, available P and K), and greatly increased the cover and species richness of native plants. In addition, N and K of Cuscuta were positively correlated with N of Mikania, which was negatively associated with soil total N, available P and K. These findings suggest that Cuscuta may be an effective measure against Mikania and be beneficial to the restoration of invaded communities.  相似文献

Latitudinal variation in avian life history strategies is well documented. Clutch size and nest success tend to increase with latitude, whereas longevity and developmental periods have been argued to decrease with latitude. However, these patterns are largely based on interspecific comparisons of species breeding at tropical and temperate latitudes. We compared the life history of Yellow Warblers Setophaga petechia breeding in arctic habitat at the northern extent of their range, in Inuvik, NWT (68°N), Canada, with those breeding in temperate habitat in Revelstoke, BC (50°N), and use data from 21 populations spanning 0–68°N to evaluate latitudinal trends in life history traits from tropical to arctic habitats. Females breeding in Inuvik laid first clutches that were slightly (although not significantly) larger and had higher nest success, which resulted in higher annual productivity compared with their low- latitude counterparts. Apparent adult survival rates were only marginally lower in Inuvik than in Revelstoke, whereas incubation and nestling periods in the arctic were similar to our temperate site. When comparing life history traits across the Yellow Warbler breeding range, we observed increases in clutch sizes and nest success with increasing latitude that appeared to be associated with declines in adult survival, though this relationship was weakened by the addition of our arctic site. We detected more moderate declines in incubation and nestling periods with increasing latitude. As we observed latitudinal variation in some life history traits, but not a consistent transition of traits associated with a shift from a slow to fast life history from tropical to arctic latitudes, our study suggests that the expectation for a general shift in life history traits may be over-simplified.  相似文献

We studied the effects of environmental factors on the ability to compensate for simulated herbivory in an annual (Urtica urens) and two perennial (U. dioica subspp. dioica and sondenii) nettle species in order to test the compensatory continuum hypothesis. Further, we studied the expression of costs of structural defence in different environmental conditions. Herbivory interacted significantly with density and fertilization for height growth, branching and reproductive traits. The negative effects of simulated herbivory on relative height growth were less detrimental for plants grown in low density and high fertilization level, which supports the compensatory continuum hypothesis. However, with respect to other traits measured, our results do not support the idea of compensatory continuum. The negative effects of apical excision on inflorescence biomass were relatively more prominent in plants growing at low density and receiving more fertilization than on those growing in worse conditions. In addition, branching was reduced by apical excision regardless of resource levels. The lack of compensation for herbivory is explained by the role which the damaged or removed tissue plays in plant development and function. In the perennial U. dioica, defensive responses to herbivory, measured as changes in trichome density, were stronger than in the annual U. urens. In the southern subspecies dioica, trichome density increased in newly emerged leaves after leaf clipping. In the northern subspecies sondenii, trichome density increased after apical excision. The differences in the defensive responses between the two subspecies of U. dioica may be due to differential natural herbivory pressures on subspecies inhabiting different geographical regions. We observed negative phenotypic correlations between structural defence and other plant traits, which suggests the existence of costs of defence. In addition to differences among the species studied, the expression of costs of defence was dependent on the resource levels.  相似文献

We surveyed the non-mycorrhizal model plant Arabidopsis thaliana microscopically for its ability to form dark septate endophyte (DSE) symbioses in field, greenhouse, and laboratory studies. The laboratory studies were also used to estimate host growth responses to 34 Periconia macrospinosa and four Microdochium sp. isolates. Consistent with broad host range observed in previous experiments, field-, greenhouse-, and laboratory-grown A. thaliana were colonized by melanized inter- and intracellular hyphae and microsclerotia or chlamydospores indicative of DSE symbiosis. Host responses to colonization were variable and depended on the host ecotype. On average, two A. thaliana accessions (Col-0 and Cvi-0) responded negatively, whereas one (Kin-1) was unresponsive, a conclusion consistent with our previous analyses with forbs native to the field site where the fungi originate. Despite the average negative responses, examples of positive responses were also observed, a conclusion also congruent with earlier studies. Our results suggest that A. thaliana has potential as a model for more detailed dissection of the DSE symbiosis. Furthermore, our data suggest that host responses are controlled by variability in the host and endophyte genotypes.  相似文献

Aim Aspects of connectivity and continuity operating in the River Meuse were analysed for their contribution to the biodiversity of the floodplain. From this analysis of the diversity and composition of the meadow communities, we aimed to derive effective biodiversity conservation strategies. Location The River Meuse is one of the larger rivers in the European Western Plains ecoregion. The alluvial plains of the river have a long history of cultivation, and for these plains the floodplain meadow vegetation is a highly appreciated and valuable nature conservation asset. Method We sampled floodplain meadows from 400 km of the six geomorphic reaches of the middle to lower course of the River Meuse. For each, 50 vascular plant relevés were recorded, representing the spectrum of floodplain meadow communities of that reach. Beta diversity was calculated to quantify similarity in species pools between the reaches. A dissimilarity formula was used to determine the turnover between the reaches, and these dissimilarities were compared with a Mantel test to detect whether species composition of the floodplain meadows exhibited connectivity and continuity between the reaches. Species richness for the floodplain vegetation data of the reaches was compared with data for riparian invertebrate communities. The vegetation data sets were ordinated using detrended correspondence analysis (DCA) to reveal patterns in the floodplain meadow species composition, and the DCA axes were related to plant functional groups and population strategies. The axis scores of the species and plots were linked to river and plant species traits. Results We did not observe an overall continuity trend in similarity, nor one in diversity in a downstream direction. Lateral connectivity was highlighted by the dissimilarity between the reaches and in the influxes of species from adjacent ecoregions. The DCA ordination showed statistically significant separations between reaches and between the plant functional groups. The second DCA axis related to the longitudinal gradient of the river, whereas the first axis showed stronger correlations with river and plant species traits. We termed this axis the ‘disorder axis’. Plant invaders and avoiders are located at the left side of the disorder axis, whereas the true river‐adapted categories of resisters and endurers are at the right extremity. Contributions to the disorder were identified in terms of connectivity with adjacent ecoregions and the physical disturbance regime of natural and anthropogenic perturbations, resulting in community changes between the reaches. Main conclusions We concluded that a single overall strategy for biodiversity conservation of the river or its floodplains is not feasible. Strategies can, however, be derived for separate river reaches based on functional groups in the communities, the disorder characteristics of the reach, and the influence of surrounding ecoregions.  相似文献

Behavior in a confined group of mandrills (Mandrillus sphinx) was compared before and after their move from a traditional, all indoor exhibit to an indoor/outdoor facility. The outdoor exhibit was designed to represent as well as is possible important features of the mandrills' forest habitat. We predicted that activity budgets in the outdoor exhibit would more closely resemble those of wild African Cercopithecines, specifically that time spent in forage/feed, locomotion, and social behavior would increase and time spent stationary would decrease. Time spent in forage/feed, locomotion, and stationary did change, as expected. However, there was a small significant decrease in some social behaviors. Differences in proximate environmental contingencies may explain the behavioral changes observed: feeding/foraging opportunities throughout the time on exhibit; increased social refuges; modest increase in facility size; and significantly increased environmental complexity and novelty. Our results are discussed within the context of data obtained from mandrills and baboons observed along a theoretical continuum between confined and free‐ranging environments. This analysis supports what others have suggested: that primate well‐being is not so much a function of confinement as of the presence of relevant incentives to engage in species‐appropriate behavior.
