Somatic stem/progenitor cells actively proliferate and give rise to different types of mature cells (active state) in embryonic tissues while they are mostly dormant (quiescent state) in many adult tissues. Notch signaling is known to regulate both active and quiescent states of somatic stem cells, but how it regulates these different states is unknown. Recent studies revealed that the Notch effector Hes1 is expressed differently during the active and quiescent states during neurogenesis and myogenesis: high in the quiescent state and oscillatory in the active state. When the Hes1 expression level is high, both Ascl1 and MyoD expression are continuously suppressed. By contrast, when Hes1 expression oscillates, it periodically represses expression of the neurogenic factor Ascl1 and the myogenic factor MyoD, thereby driving Ascl1 and MyoD oscillations. High levels of Hes1 and the resultant Ascl1 suppression promote the quiescent state of neural stem cells, while Hes1 oscillation-dependent Ascl1 oscillations regulate their active state. Similarly, in satellite cells of muscles, known adult muscle stem cells, high levels of Hes1 and the resultant MyoD suppression seem to promote their quiescent state, while Hes1 oscillation-dependent MyoD oscillations activate their proliferation and differentiation. Therefore, the expression dynamics of Hes1 is a key regulatory mechanism of generating and maintaining active/quiescent stem cell states.  相似文献

Black soldier fly (BSF) larvae, Hermetia illucens L. (Diptera: Stratiomyidae), bio-convert organic side streams into high-quality biomass, the composition of which largely depends on the side stream used. In the present study, BSF larvae were reared on feed substrates composed of dried brewers’ spent grains, each supplemented with either water, waste brewer’s yeast, or a mixture of waste brewer’s yeast and cane molasses to obtain 12 different substrates: barley/water, barley/yeast, barley/yeast/molasses, malted barley/water, malted barley/yeast, malted barley/yeast/molasses, malted corn/water, malted corn/yeast, malted corn/yeast/molasses, sorghum-barley/water, sorghum-barley/yeast, and sorghum-barley/yeast/molasses. The crude protein, fat, ash, and mineral contents of the BSF larvae fed each feed substrate were quantified by chemical analyses. The effect of substrate, supplementation, and their interaction on crude protein, fat, and ash contents of BSF larval body composition was significant. Calcium, phosphorus, and potassium were the most abundant macrominerals in the larvae and their concentrations differed significantly among substrates. These findings provide important information to support the use of BSF larval meal as potential new source of nutrient-rich and sustainable animal feed ingredients to substitute expensive and scarce protein sources such as fishmeal and soya bean meal.  相似文献

Application of external organic inputs to soils can be considered as one of the most ancient strategies in agriculture, and it has been commonly used since the very beginning of human-based agricultural practices. During all this time, application of several organic matters to agricultural soils has demonstrated their benefit to plants and soils. Organic amendments have proved to be useful in recovering soil properties, improving soil quality and, in some cases, can be directly involved in providing beneficial effects to plants. All these obtained effects finally lead to an increase in crop protection and sustainability. One most expected effect caused by the application of organic amendments, is the suppression of a wide range of soilborne pathogens (mainly bacterial and fungal pathogens) due to the induction of physicochemical and biological changes in soils. In order to get insight into the nature of the induced soil suppression of soilborne plant pathogens, the analysis of the physical, chemical and the microbial changes, pointed to the key role of beneficial activities produced by soil microorganisms finally adapted to the environmental changes produced by the influence of organic amendments. As shown in the case studies reported here, participation of soil microbes specifically selected after organic amendment is crucial in the control of fungal soilborne diseases. Moreover, the development of “omics” approaches allowed these recent studies to go one step further, revealing the main actors involved in the induced soil suppressiveness and their activities. Thus “omics” techniques will help to understand the soil and its microbiome as a whole system, and to assign the important roles of its biological components.  相似文献

Dissolved carbon dioxide (dCO2) is a well-known critical parameter in bioprocesses due to its significant impact on cell metabolism and on product quality attributes. Processes run at small-scale faces many challenges due to limited options for modular sensors for online monitoring and control. Traditional sensors are bulky, costly, and invasive in nature and do not fit in small-scale systems. In this study, we present the implementation of a novel, rate-based technique for real-time monitoring of dCO2 in bioprocesses. A silicone sampling probe that allows the diffusion of CO2 through its wall was inserted inside a shake flask/bioreactor and then flushed with air to remove the CO2 that had diffused into the probe from the culture broth (sensor was calibrated using air as zero-point calibration). The gas inside the probe was then allowed to recirculate through gas-impermeable tubing to a CO2 monitor. We have shown that by measuring the initial diffusion rate of CO2 into the sampling probe we were able to determine the partial pressure of the dCO2 in the culture. This technique can be readily automated, and measurements can be made in minutes. Demonstration experiments conducted with baker's yeast and Yarrowia lipolytica yeast cells in both shake flasks and mini bioreactors showed that it can monitor dCO2 in real-time. Using the proposed sensor, we successfully implemented a dCO2-based control scheme, which resulted in significant improvement in process performance.  相似文献

Synechocystis sp. PCC 6803, a cyanobacterium widely used for basic research, is often cultivated in a synthetic medium, BG-11, in the presence of 4-(2-hydroxyethyl)-1-piperazine ethanesulfonic acid (HEPES) or 2-[[1,3-dihydroxy-2-(hydroxymethyl)propan-2-yl]amino]ethanesulfonic acid buffer. Owing to the high cost of HEPES buffer (96.9% of the total cost of BG-11 medium), the biotechnological application of BG-11 is limited. In this study, we cultured Synechocystis sp. PCC 6803 cells in BG-11 medium without HEPES buffer and examined the effects on the primary metabolism. Synechocystis sp. PCC 6803 cells could grow in BG-11 medium without HEPES buffer after adjusting for nitrogen sources and light intensity; the production rate reached 0.54 g cell dry weight·L−1·day−1, exceeding that of commercial cyanobacteria and Synechocystis sp. PCC 6803 cells cultivated under other conditions. The exclusion of HEPES buffer markedly altered the metabolites in the central carbon metabolism; particularly, the levels of compatible solutes, such as sucrose, glucosylglycerol, and glutamate were increased. Although the accumulation of sucrose and glucosylglycerol under high salt conditions is antagonistic to each other, these metabolites accumulated simultaneously in cells grown in the cost-effective medium. Because these metabolites are used in industrial feedstocks, our results reveal the importance of medium composition for the production of metabolites using cyanobacteria.  相似文献

Species inhabiting mountain ecosystems are expected to be particularly vulnerable to environmental change, yet information on their basic ecology is often lacking. Knowledge from field-based empirical studies remains essential to refine our understanding of the impact of current habitat alterations and for the consequential development of meaningful conservation management strategies. This study focuses on a poorly investigated and vulnerable mountain bird species in Europe, the Ring Ouzel Turdus torquatus. Our aim was to identify the species’ key ecological requirements during the crucial period of nestling provisioning in the context of environmental change. We radiotracked and observed Alpine Ring Ouzels in a high-density population, investigating their pattern of foraging habitat selection in 2015 and 2017, and evaluated the transferability of these results over a wider geographical range across the SW Swiss Alps. Foraging birds selected, consistently in space and time, short grass swards (< 10 cm) with interspersed patches of accessible and penetrable soils, at intermediate moisture levels (around 40–65% volumetric water content). In Alpine ecosystems, this microhabitat configuration is typically widespread during the spring snowmelt, but extremely seasonal, with a rapid decrease in its availability over the course of the breeding season. This underlines the high vulnerability of the Ring Ouzel to environmental change: an earlier snowmelt could generate a temporal mismatch between the peak of the breeding effort and optimal foraging conditions; however, abandoning grazing activities on semi-wooded Alpine pastures may further decrease foraging habitat suitability through taller and denser grass swards, and subsequent woody vegetation encroachment. This study provides a mechanistic appraisal of the challenges Ring Ouzels will face in the future, as well as initial guidelines for targeted habitat management within timberline ecotones.  相似文献

The photosynthetic CO2 fixing enzyme ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) forms dead-end inhibited complexes while binding multiple sugar phosphates, including its substrate ribulose 1,5-bisphosphate. Rubisco can be rescued from this inhibited form by molecular chaperones belonging to the ATPases associated with diverse cellular activities (AAA+ proteins) termed Rubisco activases (Rcas). The mechanism of green-type Rca found in higher plants has proved elusive, in part because until recently higher-plant Rubiscos could not be expressed recombinantly. Identifying the interaction sites between Rubisco and Rca is critical to formulate mechanistic hypotheses. Toward that end here we purify and characterize a suite of 33 Arabidopsis Rubisco mutants for their ability to be activated by Rca. Mutation of 17 surface-exposed large subunit residues did not yield variants that were perturbed in their interaction with Rca. In contrast, we find that Rca activity is highly sensitive to truncations and mutations in the conserved N terminus of the Rubisco large subunit. Large subunits lacking residues 1–4 are functional Rubiscos but cannot be activated. Both T5A and T7A substitutions result in functional carboxylases that are poorly activated by Rca, indicating the side chains of these residues form a critical interaction with the chaperone. Many other AAA+ proteins function by threading macromolecules through a central pore of a disc-shaped hexamer. Our results are consistent with a model in which Rca transiently threads the Rubisco large subunit N terminus through the axial pore of the AAA+ hexamer.  相似文献

ABSTRACT

This paper presents an active learning approach that focuses on practical investigation of the ecosystem of tidal flats using 3D modeling and printing for biology students in order to enhance understanding of natural selection. The learning approach for the study followed a 5-step procedure: i) learning about 3D modeling and printing, ii) exploration of the ecosystem of tidal flats, iii) 3D designing of a bird beak, iv) 3D printing of a constructed beak, and v) a natural selection simulation activity. The learning method presented in this study centered on active student exploration of the tidal flats ecosystem using 3D modeling and printing. The learning approach presented in this paper could be implicated at schools to aid in students’ understanding of natural selection as it allows students to firsthand examine simulation changes to a bird beak and benthic communities. This study suggested the active learning method for natural selection as it incorporates student-designed exploration and direct investigative appraisal of the selection process.  相似文献

黑土是有机质含量高且肥沃的土壤类型之一,气候变化会显著改变黑土中微生物群落的结构,同时影响群落间的潜在相互作用关系。[目的] 揭示水热增加对黑土中的细菌群落结构及潜在互作关系的影响。[方法] 基于土壤移置试验,采用16S rRNA高通量测序解析农田黑土(原位黑土、水热增加1和水热增加2)中的细菌群落结构对水热增加的响应;使用CoNet构建微生物群落共生网络,识别共生网络中的枢纽微生物;利用结构方程模型、相关性分析探究水热条件变化下土壤性质、微生物交互作用、多样性之间的直接、间接关系。[结果] 黑土中的微生物以疣微菌、变形杆菌、酸性杆菌和放线菌为主。水热增加下土壤微生物共生网络的拓扑性质发生显著变化,网络中表征微生物潜在竞争关系的负连线随着水热增加而显著增加。气候因素通过改变微生物潜在相互作用影响了群落水平分类多样性。物种竞争增强可能直接导致了土壤有机碳含量的降低。[结论] 水热增加会显著改变黑土中微生物之间的潜在交互作用,枢纽微生物的响应更加敏感。  相似文献
