Proteomic profiling of the substantia nigra demonstrates CNDP2 overexpression in Parkinson's disease

Despite decades of intensive investigations, the precise sequence of molecular events and the specific proteins mediating the degenerative process underlying Parkinson's disease (PD) remain unraveled. Proteomic strategies may provide unbiased tools to identify novel candidates and explore original mechanisms involved in PD. Substantia nigra pars compacta (SN) tissue, whose degeneration is the hallmark of PD, was dissected from neuropathologically confirmed PD patients (n=3) and control subjects (n=3), before being submitted to a comparative 2-DE analysis. The present study revealed a subset of neuronal and/or glial proteins that appears to be deregulated in PD and likely to contribute to neurodegeneration. Observed alterations not only consolidate well accepted concepts surrounding PD pathogenesis such as oxidative stress and mitochondrial dysfunction but also point out to novel pathways. Among the latter, cytosolic non specific dipeptidase 2 (CNDP2), a relatively unknown protein not yet reported to be associated with PD pathogenesis, was shown to be increased in the SN of PD patients, as confirmed by Western blot. Immunohistochemical analyses demonstrated the presence of CNDP2 within the cytoplasm of SN dopaminergic neurons. Altogether, our findings support a key role of CNDP2 in PD neurodegeneration, by mechanisms that could involve oxidative stress, protein aggregation or inflammation. The multilevel society of hamadryas baboons, consisting of troops, bands, clans, and one-male units (OMUs), is commonly perceived to be an effective means of adapting to variable food availability while allowing spatial cohesion in response to predator pressure. The relationship between these variables, however, has never been tested quantitatively. The Filoha site in Awash National Park, Ethiopia is ideally suited to such an investigation as it contains nutrient-dense palm forests in addition to the Acacia scrublands typical of hamadryas distribution elsewhere, allowing comparisons of spatial cohesion across habitat types. Here, we use observations over a 1-year period to examine the relationship between resource availability, perceived predator pressure, and spatial cohesion in a band of wild hamadryas baboons at Filoha. Our results demonstrate that the band was more likely to break into OMUs when foraging in habitats with lower food availability, and that the band fissioned into independent clans more often when preferred resources were not available. Furthermore, the baboons remained in larger aggregations for longer periods of time (i.e., prior to embarking on their daily foraging route) on mornings after predators were heard in the vicinity, and increased cohesion in response to encounters with people who may have been perceived as predators. These results support the notion that hamadryas baboons change their social groupings in response to both food availability and predation risk and that the ability of hamadryas bands to cleave and coalesce in response to changes in these factors underlies the evolution of the hamadryas modular social structure. 相似文献

The lack of association between wealth and fertility in contemporary industrialized populations has often been used to question the value of an evolutionary perspective on human behaviour. Here, we first present the history of this debate, and the evolutionary explanations for why wealth and fertility (the number of children) are decoupled in modern industrial settings. We suggest that the nature of the relationship between wealth and fertility remains an open question because of the multi-faceted nature of wealth, and because existing cross-sectional studies are ambiguous with respect to how material wealth and fertility are linked. A literature review of longitudinal studies on wealth and fertility shows that the majority of these report positive effects of wealth, although levels of fertility seem to fall below those that would maximize fitness. We emphasize that reproductive decision-making reflects a complex interplay between individual and societal factors that resists simple evolutionary interpretation, and highlight the role of economic insecurity in fertility decisions. We conclude by discussing whether the wealth–fertility relationship can inform us about the adaptiveness of modern fertility behaviour, and argue against simplistic claims regarding maladaptive behaviour in humans. 相似文献

The topic of this paper is external versus internal explanations, first, of the genesis of evolutionary theory and, second, its reception. Victorian England was highly competitive and individualistic. So was the view of society promulgated by Malthus and the theory of evolution set out by Charles Darwin and A.R. Wallace. The fact that Darwin and Wallace independently produced a theory of evolution that was just as competitive and individualistic as the society in which they lived is taken as evidence for the impact that society has on science. The same conclusion is reached with respect to the reception of evolutionary theory. Because Darwins contemporaries lived in such a competitive and individualistic society, they were prone to accept a theory that exhibited these same characteristics. The trouble is that Darwin and Wallace did not live in anything like the same society and did not formulate the same theory. Although the character of Victorian society may have influenced the acceptance of evolutionary theory, it was not the competitive, individualistic theory that Darwin and Wallace set out but a warmer, more comforting theory. 相似文献

In political and biomedical discourses, "posttraumatic stress disorder" has become a set of organizing concepts for trauma and traumatic memory. These concepts, however, are predicated on an understanding of traumatic memory as a discrete etiological event that, when reexperienced, is productive of symptoms. In this essay, I explore alternative framings of trauma that arise out of historical changes in political economic language and from experiences of monetary, historical, and affective indebtedness in Santiago, Chile. This ethnographic research is based in an historically leftist población (poor urban sector) and follows the interwoven narratives of a formerly exiled communist militant and her adopted daughter. Throughout this essay, I describe the mother's attempts to inhabit an untimely language of socialist politics and the daughter's rejection of both this language and her mother's pain. I elaborate on how these attempts are products of and productive of monetary and intersubjective indebtedness in a neoliberal present. By describing the differing historical languages inhabited by these subjects, I attempt to evoke an understanding of trauma not as an individual possession or etiological event, but rather as a referential dissonance in the neoliberal context. This referential dissonance emerges from the gap between the historical languages that inform subjectivities. I explore how such a gap can create contexts in which the everyday itself both threatens the disarticulation of the subject and produces injurious affective relationships. In this way, I interrogate relationships between trauma, recovery, and the everyday. 相似文献

Drawing from an evening of fieldwork at an eastern German Protestant churchin the rapidly deindustrialized city of Leipzig, this article finalyzes a discussion among church leaders, former ‘dissidents,’ and unemployed workers on the subject of ‘community economy’ (Gemeinschaftsökonomie), a concept people interpret to represent a potential solution to the crisis of mass unemployment. Such local debates on unemployment offer a frame for understanding the re-negotiation of labor and social welfare in eastern Germany following socialism's end. finalyzing this discussion within the context of theoretical debates on ‘civil society,’ I follow Burawoy's and Gramsci's lead in fillustrating that the imagined economic sphere of ‘community economy’ is an ideological battle ground, where hegemonic discourse supporting economic neoliberalism may be either reinforced or subverted. 相似文献
