Characteristics of mitochondrial (mt) DNA such as gene content and arrangement, as well as mt tRNA secondary structure, are frequently used in comparative genomic analyses because they provide valuable phylogenetic information. However, most analyses do not characterize the relationship of tRNA genes from the same mt genome and, in some cases, analyses overlook possible novel open reading frames (ORFs) when the 13 expected protein-coding genes are already annotated. In this study, we describe the sequence and characterization of the complete mt genome of the silver-lip pearl oyster, Pinctada maxima. The 16,994-bp mt genome contains the same 13 protein-coding genes (PCGs) and two ribosomal RNA genes typical of metazoans. The gene arrangement, however, is completely distinct from that of all other available bivalve mt genomes, and a unique tRNA gene family is observed in this genome. The unique tRNA gene family includes two trnS− AGY and trnQ genes, a trnM isomerism, but it lacks trnS− CUN. We also report the first clear evidence of alloacceptor tRNA gene recruitment (trnP → trnS− AGY) in mollusks. In addition, a novel ORF (orfUR1) expressed at high levels is present in the mt genome of this pearl oyster. This gene contains a conserved domain, “Oxidored_q1_N”, which is a member of Complex I and thus may play an important role in key biological functions. Because orfUR1 has a very similar nucleotide composition and codon bias to that of other genes in this genome, we hypothesize that this gene may have been moved to the mt genome via gene transfer from the nuclear genome at an early stage of speciation of P. maxima, or it may have evolved as a result of gene duplication, followed by rapid sequence divergence. Lastly, a 319-bp region was identified as the possible control region (CR) even though it does not correspond to the longest non-coding region in the genome. Unlike other studies of mt genomes, this study compares the evolutionary patterns of all available bivalve mt tRNA and atp8 genes.  相似文献

小型温室棚作为农业生产的重要设施,可为农作物创造一个温度适宜的、受保护的生长空间,保证作物正常生长、促进作物早熟。以确保重庆地区春冬季烟苗正常发育、维持温室内适宜的土壤温度为导向,对温室棚内温度进行了试验和数值模拟,探究了温室棚内温度分布规律,并在此基础上分析生物质发酵产热对温室棚内土壤温度场的影响。结果表明:试验中埋设稻壳的温室棚内土壤温度平均提高3.5℃且温室棚中平均温度低于10℃的天数明显减少。埋设稻壳的温室棚中烟苗茎高和叶片数增长值最大,平均茎高从移栽时的7.8 cm增长到了12 cm,叶片数从平均5片增长到了11片,均大于未埋设稻壳试验组,这表明土壤中埋设稻壳有助于烟草的生长发育。数值研究表明温室棚内不同深度的土壤层温度变化状况一致,均呈现先降低后升高再降低的趋势。地温随土壤深度的增加而降低,地表下10 cm、20 cm的土壤层区域温度高于10.6℃、8℃,有利于烟苗在低温环境下正常生长。且土壤温度存在一定的昼夜温差,更利于烟苗发育。埋设稻壳的温室土壤层温度变化趋势与未埋设稻壳的温室一致,但其土壤层平均温度、最低温度至最高温度以及昼夜温差分别升高了2.3℃、1.1—5.5℃、...  相似文献

How plant‐feeding insects distribute themselves and utilize their host plant resources is still poorly understood. Several processes may be involved, and their relative roles may vary with the spatial scale considered. Herein, we investigate small‐scale patterns, namely how population density of a gall midge is affected by individual growth form, phenology, and microsite characteristics of its herb host. The long‐lived plant individuals vary much with regard to number of shoots, flower abundance, and flowering phenology. This variation is connected to site characteristics, primarily the degree of sun exposure. The monophagous insect galls the flowers of the host plant – an easily defined food resource. It is a poor disperser, but very long‐lived; diapausing larvae can stay in the soil for many years. Galls were censused on individual plants during 5 years; from a peak to a low in gall population density. Only a very small fraction of the flowers produced (<0.5%) were galled even in the peak year. Nevertheless, most plant individuals had galls at least 1 year. In a stepwise multiple regression, plant size (number of shoots) was found to be the most important predictor of gall density (galls/flower). However, gall density decreased more than one order of magnitude over the plant size range observed. There was also a weak effect of plant phenology. Early flowering plants had lower gall densities than those starting later. Sun exposure had no direct effect on gall density, but a path analysis revealed indirect effects via the timing of flowering. Gall population change was highly synchronous in different parts of the study area with no significant decrease in synchrony with distance.  相似文献

The influence of a mammalian sterol cholesterol and a plant sterol β-sitosterol on the structural parameters and hydration of bilayers in unilamellar vesicles made of monounsaturated diacylphosphatidylcholines (diCn:1PC, n = 14-22 is the even number of acyl chain carbons) was studied at 30 °C using small-angle neutron scattering (SANS). Recently published advanced model of lipid bilayer as a three-strip structure was used with a triangular shape of polar head group probability distribution (Ku?erka et al., Models to analyze small-angle neutron scattering from unilamellar lipid vesicles, Physical Review E 69 (2004) Art. No. 051903). It was found that 33 mol% of both sterols increased the thickness of diCn:1PC bilayers with n = 18-22 similarly. β-sitosterol increased the thickness of diC14:1PC and diC16:1PC bilayers a little more than cholesterol. Both sterols increased the surface area per unit cell by cca 12 Å2 and the number of water molecules located in the head group region by cca 4 molecules, irrespective to the acyl chain length of diCn:1PC. The structural difference in the side chain between cholesterol and β-sitosterol plays a negligible role in influencing the structural parameters of bilayers studied.  相似文献

Background

The endoplasmic reticulum enzyme glucose-6-phosphatase catalyzes the hydrolysis of glucose-6-phosphate to glucose and inorganic phosphate. The enzyme is a part of a multicomponent system that includes several integral membrane proteins; the catalytic subunit (G6PC) and transporters for glucose-6-phosphate, inorganic phosphate and glucose. The G6PC gene family presently includes three members, termed as G6PC, G6PC2, and G6PC3. Although the three isoforms show a moderate amino acid sequence homology, their membrane topology and catalytic site are very similar. The isoforms are expressed differently in various tissues. Mutations in all three genes have been reported to be associated with human diseases.

Scope of review

The present review outlines the biochemical features of the G6PC gene family products, the regulation of their expression, their role in the human pathology and the possibilities for pharmacological interventions.

Major conclusions

G6PCs emerge as integrators of extra- and intracellular glucose homeostasis. Beside the well known key role in blood glucose homeostasis, the members of the G6PC family seem to play a role as sensors of intracellular glucose and of intraluminal glucose/glucose-6-phosphate in the endoplasmic reticulum.

General significance

Since mutations in the three G6PC genes can be linked to human pathophysiological conditions, the better understanding of their functioning in connection with genetic alterations, altered expression and tissue distribution has an eminent importance.  相似文献

A consequence of the Chernobyl disaster was the isolated increase of small air ions for a limited time period over Athens-Greece. Taking advantage of this unique condition we studied the relation of log daily mean concentrations of small positive air ions, small negative air ions, NO2 and SO2 with the total daily number of asthma attacks attendances at the region's largest chest diseases hospital for 35 days following the disaster and during a same time period in the preceding four years. No correlation was found between air ionization and asthma attacks, though the study was of a rather short duration and several possible influencing factors such as aeroallergen concentrations were not taken into consideration.  相似文献

A synthesis of available data on the morphological diversity of polykrikoid dinoflagellates allowed us to formulate a hypothesis of relationships that help explain character evolution within the group. Phylogenetic analyses of new SSU rDNA sequences from Pheopolykrikos beauchampii Chatton, Polykrikos kofoidii Chatton, and Polykrikos lebourae Herdman helped refine this hypothetical framework. Our results demonstrated that “pseudocolonies” in dinoflagellates evolved convergently at least three times independently from different Gymnodinium‐like ancestors: once in haplozoans; once in Ph. beauchampii; and at least once within a lineage containing Ph. hartmannii, P. kofoidii, and P. lebourae. The Gymnodiniales sensu stricto was strongly supported by the data, and the type species for the genus, namely Gymnodinium fuscum (Ehrenb.) F. Stein, formed the nearest sister lineage to a well‐supported Polykrikos clade. The best synapomorphy for the Polykrikos clade was the presence of two nuclei irrespective of zooid number. Two unidentified Gymnodinium species formed the nearest sister clade to Ph. beauchampii, which has four nuclei and four zooids per pseudocolony. The chain‐forming dinoflagellate G. catenatum L. W. Graham branched closely to the clade containing all members of Polykrikos and Pheopolykrikos, suggesting that an ancestral capacity toward chain formation existed before the evolution of pseudocolonies in this group. Our results also clarified the phylogenetic significance of nematocysts, ocelloids, and photosynthesis in reconstructing the evolution of polykrikoids and warnowiids. The molecular phylogenies exposed taxonomic problems associated with Polykrikos, Pheopolykrikos, and Gymnodinium, and suggested that a revision for some of these genera is warranted.  相似文献
