对一蚕豆病遗传家系的G6PD基因突变进行分析,检测突变后G6PD酶活变化,并对先证者家系进行X染色体失活(XCI)偏移模式检测,从而预测G6PD突变女性携带者患蚕豆病的风险。取家系成员的外周血样,并提取基因组DNA,用聚合酶链式反应(PCR)和DNA测序法进行序列分析,确定先证者突变位点和突变类型及家庭成员遗传情况,若先证者的母亲和姐姐为G6PD突变携带者,则对先证者母亲和姐姐进行X染色体偏移检测以及酶活检测分析,以评估携带者患蚕豆病的风险,同时对研究对象进行随访。结果患者X染色体上G6PD基因发生点突变c.1376G>T;酶活性检测结果显示该突变使G6PD酶活性下降大约25%,导致蚕豆病发生。该家系的两位女性携带者X染色体失活偏移<80%,未来发生蚕豆病的可能性低。 相似文献

Temperature fluctuation is one of the most important factors in climate change, and research into its effects on organisms is increasing. Nonetheless, few studies have addressed its effects on insects and particularly on the parental generation. We therefore simulated temperature amplitudes (± 0, 4, 6, 8, 10°C) for a mean environmental temperature of 25°C and studied their immediate and maternal effects on a global pest, the diamondback moth Plutella xylostella (Lepidoptera: Plutellidae), focusing on adult longevity and fecundity and egg development and survivorship in the next generation. Relative to constant temperature, moderate amplitudes (4 and 6°C) did not affect the adults, whereas 8 and 10°C amplitudes hindered adult longevity and fecundity, but significantly increased the proportional of early fecundity after each treatment. Temperature amplitude 10°C inhibited development of Day 2 and Day 3 laid eggs. These findings show that temperature amplitude affects both the maternal and offspring phenotype and indicate that the influence of temperature amplitude on diamondback moth population models requires further research. 相似文献

Background and Aims
In heterostylous plant species, skewed morph ratios are not uncommon and may arise from a range of factors. Despite the recognized importance of skewed morph ratios on overall reproductive success within populations, little is known about the impact of skewed morph ratios on population genetic diversity and differentiation in heterostylous species. This study specifically aimed to clarify the effect of population size and morph bias on population genetic diversity and differentiation in the temperate forest herb Pulmonaria officinalis. This species is characterized by a distylous breeding system and shows morph-specific differences in reproductive success.Methods
Genetic diversity was determined for 27 P. officinalis populations in northern Belgium by using eight recently developed microsatellite markers. Multiple regressions were used to assess the relationship between genetic diversity, morph bias and population size, and FST-values were calculated for short- and long-styled morphs separately to study genetic differentiation as a function of morph type.Key Results
For all genetic measures used, morph bias was more important in explaining patterns of genetic diversity than population size, and in all cases patterns of population genetic diversity followed a quadratic function, which showed a symmetrical decrease in genetic diversity with increasing morph bias. However, probably due to the reproductive advantage of L-morphs relative to S-morphs, maximum genetic diversity was found in populations showing an excess of L-morphs (60·7 % L-morph). On the other hand, no significant difference in pairwise genetic distances between populations was observed between L- (0·107) and S-morphs (0·106).Conclusions
Our results indicate that significant deviations from equal morph ratios not only affect plant reproductive success but also population genetic diversity of heterostylous plant species. Hence, when defining conservation measures for populations of heterostylous plant species, morph ratios should be considered as an important trait affecting their long-term population viability. 相似文献74.

The effect of different external salt concentrations, from 0 mM to 1030 mM NaCl, on photosynthetic complexes and chloroplast ultrastructure in the halophyte Arthrocnemum macrostachyum was studied. Photosystem II, but not Photosystem I or cytochrome b6/f, was affected by salt treatment. We found that the PsbQ protein was never expressed, whereas the amounts of PsbP and PsbO were influenced by salt in a complex way. Analyses of Photosystem II intrinsic proteins showed an uneven degradation of subunits with a loss of about 50% of centres in the 0 mM NaCl treated sample. Also the shape of chloroplasts, as well as the organization of thylakoid membranes were affected by NaCl concentration, with many grana containing few thylakoids at 1030 mM NaCl and thicker grana and numerous swollen thylakoids at 0 mM NaCl. Much of our understanding of protein folding mechanisms is derived from experiments using intrinsic fluorescence of natural or genetically inserted tryptophan (Trp) residues to monitor protein refolding and site-directed mutagenesis to determine the energetic role of amino acids in the native (N), intermediate (I) or transition (T) states. However, this strategy has limited use to study complex folding reactions because a single fluorescence probe may not detect all low-energy folding intermediates. To overcome this limitation, we suggest that protein refolding should be monitored with different solvent-exposed Trp probes. Here, we demonstrate the utility of this approach by investigating the controversial folding mechanism of ubiquitin (Ub) using Trp probes located at residue positions 1, 28, 45, 57, and 66. We first show that these Trp are structurally sensitive and minimally perturbing fluorescent probes for monitoring folding/unfolding of the protein. Using a conventional stopped-flow instrument, we show that ANS and Trp fluorescence detect two distinct transitions during the refolding of all five Trp mutants at low concentrations of denaturant: T1, a denaturant-dependent transition and T2, a slower transition, largely denaturant-independent. Surprisingly, some Trp mutants (UbM1W, UbS57W) display Trp fluorescence changes during T1 that are distinct from the expected U → N transition suggesting that the denaturant-dependent refolding transition of Ub is not a U → N transition but represents the formation of a structurally distinct I-state (U → I). Alternatively, this U → I transition could be also clearly distinguished by using a combination of two Trp mutations UbF45W-T66W for which the two Trp probes that display fluorescence changes of opposite sign during T1 and T2 (UbF45W-T66W). Global fitting of the folding/unfolding kinetic parameters and additional folding-unfolding double-jump experiments performed on UbM1W, a mutant with enhanced fluorescence in the I-state, demonstrate that the I-state is stable, compact, misfolded, and on-pathway. These results illustrate how transient low-energy I-states can be characterized efficiently in complex refolding reactions using multiple Trp probes. 相似文献

The precursor protein receptor at the chloroplast outer membrane atToc33 is a GTPase, which can be inactivated by phosphorylation in vitro, being arrested in the GDP loaded state. To assess the physiological function of phosphorylation, attoc33 knock out mutants were complemented with a mutated construct mimicking the constitutively phosphorylated state. Our data suggest that the reduced functionality of the mutant protein can be compensated by its upregulation. Chloroplast biogenesis and photosynthetic activity are impaired in the mutants during the early developmental stage, which is consistent with the requirement of atToc33 in young photosynthetic tissues. 相似文献

Some biological experiments report effects that depend on low frequency modulation of a radiofrequency (RF) carrier. Such effects require nonlinear responses in biological preparations, which we show could be observed with great generality by the unique frequency signatures that would appear in the scattered RF energy. Following Illinger [Illinger (1982): Bioelectromagnetics 3:9-16], we considered a two part physical system. The greater part, dominated by the properties of water, interacts linearly with the RF field and is described by equilibrium thermodynamics. However, another, much smaller part, e.g., certain biological molecules and molecular subgroups, supports nonlinear interactions and is described by nonequilibrium thermodynamics. For example, a nonlinear interaction might result from scattering of RF photons from oscillators located in a region of strong field gradients, such as at membrane surfaces. A second nonlinear mechanism could appear if stress (elastic) waves were launched within the confines of the exposure vessel by RF heating. Amplitude modulation at angular frequency Omega of a carrier wave with angular frequency omega (omega < omega) produces two peaks in elastic stress in the cell structure during each period; that is, there is "full-wave demodulation." As a result of coherent nonlinear charge motion, modulation products could appear at frequencies omega +/- 2 omega and, in general, at omega +/- n 2 omega (n = 1, 2, em leader ) if vibrational harmonics at 2 n omega also are excited. Although in principle microwaves can alter the stability of a thermodynamic system by pumping a chemical transition, the degree of nonlinear coupling required for an observable instability is so great that its probability is effectively zero, unless field intensity is extremely high. Male grass gobies show two alternative breeding tactics, territorial and sneaker, distinguished by body size and difference in ray elongation on the second dorsal fin. The larger males, with elongated fins, are territorial and emit sounds during courtship. Smaller males, without elongated fins, act as sneakers. Both large and small males produce sounds in the presence of a ripe female. Males produce a grunt, lasting about 300ms, made up of pulses repeated at a low rate (22–68pps). Pulse duration, number, and repetition rate, did not differ between the two male types, but dominant frequency and sound amplitude did. Dominant frequency had a strong, inverse relationship with body size, whereas sound amplitude showed a weak positive relation to body size. Male size, and not the particular reproductive male tactic employed, is the most important correlate of sound properties in this species. 相似文献

Using recording of visual evoked potentials (VEP), we examined 126 children (from 1 to 14 years old) suffering from spastic forms of cerebral palsy. In the overwhelming majority of cases (84.1%), we found in these patients significant modifications of the VEP related to dysfunctions of the visual analyzer system at different levels of the latter. We analyzed correlation of clinical manifestations of the disease with deviations of the VEP characteristics from the age norm. We conclude that an EEG examination, including VEP recording, allows one to obtain objective estimates of disorders of the visual function in the studied population of patients. 相似文献
