The presence of collagen and charged macromolecules like glycosaminoglycans (GAGs) in the interstitial space limits the space available for plasma proteins and other macromolecules. This phenomenon, known as interstitial exclusion, is of importance for interstitial fluid volume regulation. Physical/mathematical models are presented for calculating the exclusion of electrically charged and neutral macromolecules that equilibrate in the interstitium under various degrees of hydration. Here, a central hypothesis is that the swelling of highly electrically charged GAGs with increased hydration shields parts of the neutral collagen of the interstitial matrix from interacting with electrically charged macromolecules, such that exclusion of charged macromolecules exhibits change due to steric and charge effects. GAGs are also thought to allow relatively small neutral, but also charged macromolecules neutralized by a very high ionic strength, diffuse into the interior of GAGs, whereas larger macromolecules may not. Thus, in the model, relatively small electrically charged macromolecules, such as human serum albumin, and larger neutral macromolecules such as IgG, will have quite similar total volume exclusion properties in the interstitium. Our results are in agreement with ex vivo and in vivo experiments, and suggest that the charge of GAGs or macromolecular drugs may be targeted to increase the tissue uptake of macromolecular therapeutic agents.  相似文献

Terrestrial isopods were collected in 13 forest fragments differing in area (within the range of 0.1 and 254.5 ha), shape and composition of forest vegetation (thermophilous oak, mesophilous oak-hornbeam, thermophilous oak-hornbeam, acidophilous oak, basiphilous oak, beech oak-hornbeam, moist mixed deciduous forest, plantations of deciduous and coniferous trees), all situated in the Český kras Protected Landscape Area, Czech Republic, Central Europe. Number of sites sampled in each fragment of forest depended on its size and ranged from 1 to 7. Altogether 30 sites were sampled. Soil samples (5 per site collected twice a year) and pitfall trapping (5 traps per site in continuous operation throughout a year) during 2008–2009 yielded a total of 14 species of terrestrial isopods. The highest densities and highest epigeic activities of terrestrial isopods were recorded in the smallest fragments of woodland. Although a wider range of habitats were sampled in the larger fragments of woodland there was not a greater diversity of species there and the population densities and epigeic activities recorded there were lower. Porcellium collicola was most abundant in small fragments of woodland regardless the vegetation there. Armadillidium vulgare and Protracheoniscus politus were statistically more abundant in the larger fragments of woodland. The results indicate that forest fragmentation does not necessarily result in a decrease in the species richness of the isopod assemblages in such habitats.  相似文献

A comparative study of species assemblages and population densities was conducted on Amazonian monkey communities in 16 areas, ranging from 3°S latitude in northern Peru to 18°S latitude in southern Bolivia. The habitats ranged from several types of tropical rain forest in the more northern latitudes to dry, deciduous forest in the southernmost study area. The monkey populations of three of the study areas have historically received light hunting pressure; the rest have been moderately to heavily hunted. A transect census technique was used to estimate the relative and absolute densities of all diurnal monkey species except Cebuella pygmaea. The number of coexisting monkey species ranged from 4–6 in the southern areas to 12–14 in the northern areas. The reduction in species richness in central and southern areas of Bolivia is probably attributable to several inimical habitat factors. Predation by humans was found to be the single most important factor affecting monkey densities. Monkey densities, and especially biomasses, were much lower in areas not protected from hunting than in protected areas. Hunting did not affect all species equally. Larger-sized species are hunted more and have severely reduced numbers in unprotected areas, whereas the densities of smaller species are not noticeably diminished in unprotected areas. Large, herbivorous monkey species contributed the major proportion of the total monkey biomass in protected areas. The strong influence of hunting has largely obscured the effects of other factors on population densities.  相似文献

The glutamate transporter (GLT1) regulates glutamate concentrations in glutamatergic synapses and it is expressed in at least two isoforms, GLT1a and GLT1b. In this work, we show that the C-terminus of GLT1b is able to interact with the PDZ domains of a number of proteins. Notably, one of them might be the scaffold protein post-synaptic density (PSD-95). GLT1b formed co-immunoprecipitable complexes with PSD-95 in solubilizated rat brain extracts, complexes that also contained NMDA receptors. Co-transfection of GLT1b, PSD-95, and NMDA receptor subunits in heterologous expression systems recapitulated in vitro the interactions among these proteins that had been observed in the rat brain extracts and revealed the importance of the GLT1b C-terminal PDZ binding motif in tethering this transporter to PSD-95. Significantly, co-expression of GLT1b and PSD-95 increased the V max of the transporter by decreasing the rate of GLT1b endocytosis. Moreover, GLT1b transfected into primary cultured neurons or glia formed protein clusters that co-localized with co-transfected PSD-95, clusters that in these neurons accumulated preferentially in dendritic spines. We hypothesize that the GLT1b/PSD-95 interaction, characterized here in vitro , might anchor this transporter close to the post-synaptic glutamate receptors, thereby permitting the fine regulation of glutamate concentrations in this microenvironment. This tight association might also facilitate the regulation of GLT1b through the signaling pathways initiated by the activation of glutamate receptors.  相似文献

Dichotomous noise detected with the help of various single-molecule techniques convincingly reveals the actual occurrence of a multitude of conformational substates composing the native state of proteins. The nature of the stochastic dynamics of transitions between these substates is determined by the particular statistical properties of the noise observed. These involve nonexponential and possibly oscillatory time decay of the second order autocorrelation function, its relation to the third order autocorrelation function, and a relationship to dwell-time distribution densities and their correlations. Processes gated by specific conformational substates are distinguished from those with fluctuating barriers. This study throws light on the intriguing matter of the possibility of multiple stepping of the myosin motor along the actin filament per ATP molecule hydrolyzed. Aim Climate‐driven changes affecting ecosystem primary production have been well documented for many vegetation types, while the effects of climate on plant populations remains unclear. Herein, we address the relationships between climatic variables and shoot density, reproductive allocation and shoot biomass in Leymus chinensis on a large‐scale climatic gradient in 2000. Location Nine sites experiencing similar light regimes, but differing in longitude, precipitation and altitude were selected on the North‐east China Transect (NECT) from 115° to 124°E, around a latitude of 43.5°N. Methods Densities of total, vegetative and reproductive shoots and of shoot biomass were measured twice over the growing season in each site. Climatic data were taken from the climate database of the Laboratory of Quantitative Vegetation Ecology, Institute of Botany, Chinese Academy of Sciences and from the local weather stations throughout the NECT. Results Densities of total, vegetative and reproductive shoots increased significantly from the west to the east and from dry to moist along the NECT, and were strongly correlated with annual precipitation (r2 = 0.934, 0.943 and 0.863, respectively) and an aridity index (r2 = 0.809, 0.816 and 0.744, respectively). The average total shoot density at the east end (470/m2) was about three times that at the west (160/m2). Reproductive allocation and shoot biomass for both vegetative and reproductive shoots increased with precipitation and declined with the aridity index along the NECT. There were positive correlations between shoot biomass and annual precipitation for vegetative shoots (P < 0.05, R2 = 0.604) and March precipitation for reproductive shoots (P < 0.05, R2 = 0.533), respectively. Main conclusions These findings suggest that L. chinensis adjusts to decreasing precipitation/increasing aridity by alterations in shoot density, reproductive allocation and shoot biomass along the drought gradient of the NECT.  相似文献

Abstract

Central to the calculation of useful interatomic potentials is a reliable determination of the electron densities of interacting ions. When the ions are embedded in a crystalline environment, it is reasonable to expect the effect of the crystal to be important in such calculations.

For certain ions (for example Mg2+ ions in MgO) the crystal has little effect on ion electron densities. Conversely, ions such as O2? can only exist as a result of the stabilizing influence of lattice potentials; consequently, they are significantly modified by different lattice environments.

This paper will discuss the relevance of lattice effects to ions in MgO, in particular, Mg2+, Mg+, O2? and F?. Of these examples, the defect ions Mg+ and F? are used to demonstrate how, in certain circumstances, the usual assumption that cations are not affected by lattice potentials but that anions are greatly influenced is not applicable. This work also emphasises the role of embedding electron-electron interactions in addition to the Madelung potential.  相似文献

Seasonal cycles of zooplankton from San Francisco Bay   总被引:5,自引:5,他引:0  
The two estuarine systems composing San Francisco Bay have distinct zooplankton communities and seasonal population dynamics. In the South Bay, a shallow lagoon-type estuary, the copepods Acartia spp. and Oithona davisae dominate. As in estuaries along the northeast coast of the U.S., there is a seasonal succession involving the replacement of a cold-season Acartia species (A. clausi s.l.) by a warm-season species (A. californiensis), presumably resulting from the differential production and hatching of dormant eggs. Oithona davisae is most abundant during the fall. Copepods of northern San Francisco Bay, a partially-mixed estuary of the Sacramento-San Joaquin Rivers, organize into discrete populations according to salinity distribution: Sinocalanus doerrii (a recently introduced species) at the riverine boundary, Eurytemora affinis in the oligohaline mixing zone, Acartia spp. in polyhaline waters (18–30\%), and neritic species (e.g., Paracalanus parvus) at the seaward boundary. Sinocalanus doerrii and E. affinis are present year-round. Acartia clausi s.l. is present almost year-round in the northern reach, and A. californiensis occurs only briefly there in summer-fall. The difference in succession of Acartia species between the two regions of San Francisco Bay may reflect differences in the seasonal temperature cycle (the South Bay warms earlier), and the perennial transport of A. clausi s.l. into the northern reach from the seaward boundary by nontidal advection.Large numbers (>106 m–3) of net microzooplankton (>64 µm), in cluding the rotifer Synchaeta sp. and three species of tintinnid ciliates, occur in the South Bay and in the seaward northern reach where salinity exceeds about 5–10 Maximum densities of these microzooplankton are associated with high concentrations of chlorophyll. Meroplankton (of gastropods, bivalves, barnacles, and polychaetes) constitute a large fraction of zooplankton biomass in the South Bay during winter-spring and in the northern reach during summer-fall.Seasonal cycles of zooplankton abundance appear to be constant among years (1978–1981) and are similar in the deep (>10 m) channels and lateral shoals (<3 m). The seasonal zooplankton community dynamics are discussed in relation to: (1) river discharge which alters salinity distribution and residence time of plankton; (2) temperature which induces production and hatching of dormant copepod eggs; (3) coastal hydrography which brings neritic copepods of different zoogeographic affinities into the bay; and (4) seasonal cycles of phytoplankton.  相似文献
