1. Glyceraldehyde-3-phosphate dehydrogenase (G3PD) is a glycolytic enzyme that has also been implicated in a wide variety of functions within neurons. Because of the well-documented role of G3PD as an actin-binding protein, we sought evidence for a G3PD–actin complex in synaptosomes and postsynaptic densities (PSDs).2. We have shown G3PD association with 0.5-m synaptosomal particles by immunofluorescence as similarly demonstrated for actin (Toh et al., Nature 264:648–650, 1976). An immunoblot analysis also showed G3PD and actin to be enriched in synaptosomes. Further analysis of subcellular fractions from synaptosomes showed the PSD but not the synaptosomal plasma membranes to be enriched in G3PD and actin.3. Highest levels of G3PD catalytic activity were found in synaptosomes and PSDs. Although synaptosomes showed significant activity for phosphoglyceratekinase (PGK), an enzyme in sequence with G3PD for ATP production in the glycolytic pathway, no such activity was detected in the PSD fraction.4. Our studies indicate that a G3PD–actin complex may exist at the synapse. A physical association of G3PD with endogenous F-actin in synaptosomes and PSDs was demonstrated by combined phalloidin shift velocity sedimentation/immunoblot studies. By this approach, synaptosomal G3PD–actin complexes were also found to be significantly less dense than the PSD G3PD–actin complexes.5. G3PD and PGK catalytic activity in synaptosomes suggests a role in glycolysis, as well as actin binding, in the presynaptic terminals. On the other hand, the high levels of G3PD activity in PSDs but lack of PGK activity suggests that G3PD is involved in nonglycolytic functions, such as actin binding and actin filament network organization.  相似文献

Till date, fabrication of piezoelectric nanogenerator (PNG) with highly durable, high power density, and high energy conversion efficiency is of great concern. Here a flexible, sensitive, cost effective hybrid piezoelectric nanogenerator (HPNG) developed by integrating flexible steel woven fabric electrodes into poly(vinylidene fluoride) (PVDF)/aluminum oxides decorated reduced graphene oxide (AlO‐rGO) nanocomposite film is reported where AlO‐rGO acts as nucleating agent for electroactive β‐phase formation. The HPNG exhibits reliable energy harvesting performance with high output, fast charging capability, and high durability compared with previously reported PVDF based PNGs. This HPNG is capable for harvesting energy from a variety and easy accessible biomechanical and mechanical energy sources such as, body movements (e.g., hand folding, jogging, heel pressing, and foot striking, etc.) and machine vibration. The HPNG exhibits high output power density and energy conversion efficiency, facilitating direct light on different color of several commercial light‐emitting diodes instantly and powers up many portable electronic devices like wrist watch, calculator, speaker, and mobile liquid crystal display (LCD) screen through capacitor charging. More importantly, HPNG retains its performance after long compression cycles (≈158 400), demonstrating great promise as a piezoelectric energy harvester toward practical applications in harvesting biomechanical and mechanical energy for self‐powered systems.  相似文献

1 Predation and parasitism on litter‐buried cocoons of the common pine sawfly Diprion pini (L.) were compared in different forest types with endemic sawfly populations by field exposure of laboratory‐reared cocoons during three consecutive years (1993–1995). 2 The impact of cocoon predation was dependent on season and forest type. The highest predation (up to 95%) was found during autumn in forest stands with a dense understory vegetation. 3 Cocoon parasitism varied between year, season and forest type. The highest parasitoid attack was observed in pure pine forests with more or less barren soils, but did not exceed 24% of exposed cocoons. 4 Cocoons were exposed in small patches. Predators tended to exploit all cocoons of a patch, whereas parasitoids only attacked a few cocoons of a patch. Predation was similar on cocoons placed in the litter and those buried more deeply in the soil, whereas parasitism of soil‐buried cocoons was rare. 5 These results indicate that predators can have a remarkable potential for limiting endemic sawfly densities, if habitat conditions in a forest maintain their population and support their foraging behaviour. A notable effect of parasitoids on sawfly cocoons deposited in the litter is obviously restricted to typical pure and barren pine forests, but may play there a similar role as predation.  相似文献

We review the concepts and research associated with measuring fear and its consequences for foraging. When foraging, animals should and do demand hazardous duty pay. They assess a foraging cost of predation to compensate for the risk of predation or the risk of catastrophic injury. Similarly, in weighing foraging options, animals tradeoff food and safety. The foraging cost of predation can be modelled, and it can be quantitatively and qualitatively measured using risk titrations. Giving‐up densities (GUDs) in depletable food patches and the distribution of foragers across safe and risky feeding opportunities are two frequent experimental tools for titrating food and safety. A growing body of literature shows that: (i) the cost of predation can be big and comprise the forager's largest foraging cost, (ii) seemingly small changes in habitat or microhabitat characteristics can lead to large changes in the cost of predation, and (iii) a forager's cost of predation rises with risk of mortality, the forager's energy state and a decrease in its marginal value of energy. In titrating for the cost of predation, researchers have investigated spatial and temporal variation in risk, scale‐dependent variation in risk, and the role of predation risk in a forager's ecology. A risk titration from a feeding animal often provides a more accurate behavioural indicator of predation risk than direct observations of predator‐inflicted mortality. Titrating for fear responses in foragers has some well‐established applications and holds promise for novel methodologies, concepts and applications. Future directions for expanding conceptual and empirical tools include: what are the consequences of foraging costs arising from interference behaviours and other sources of catastrophic loss? Are there alternative routes by which organisms can respond to tradeoffs of food and safety? What does an animal's landscape of fear look like as a spatially explicit map, and how do various environmental factors affect it? Behavioural titrations will help to illuminate these issues and more.  相似文献

为了掌握中国特有裸子植物巴山榧树的种群现状,预测种群数量动态趋势,以分布于重庆、四川、陕西、湖北、安徽、河南和甘肃等省市的15个地点的残存巴山榧树种群为对象,建立巴山榧树种群径级结构和高度级结构图,编制种群静态生命表,绘制种群存活曲线、死亡率和消失率曲线,同时结合谱分析方法分析种群数量动态变化。结果表明:巴山榧树种群密度较低,平均为5.33株/100m~2,其中神农架种群(SN)密度最高,为9.58株/100m~2,城口种群(CK)密度最低,为2.75株/100m~2。径级结构中低龄级个体占有较高的比例,种群表现为增长型年龄结构;不同局域种群的径级结构差异明显,除宝兴种群(BX)外,其余种群的径级分布不完整。高度级结构中低于6m的个体占88.74%,绝大多数个体处于群落灌木层。种群的生命期望在不同龄级间波动明显,第3、7龄级的生命期望较低。巴山榧树种群存活曲线趋于Deevey-Ⅲ型;死亡率曲线和消失率曲线变化趋势一致,均在第3、7龄级出现峰值。巴山榧树种群的数量动态除受基波的影响外,在3、5、7龄级还显示出明显的小周期波动。巴山榧树为长寿命孑遗植物,呈岛屿化斑块分布,种群规模小,自然更新能力差,面临灭绝的危险,应及时加以保护。  相似文献

In two long-term field experiments the zinc (Zn)/cadmium (Cd) hyperaccumulator Sedum plumbizincicola (S. plumbizincicola) was examined to optimize the phytoextraction of metal contaminated soil by two agronomic strategies of intercropping with maize (Zea mays) and plant densities. Soil total Zn and Cd concentrations decreased markedly after long-term phytoextraction. But shoot biomass and Cd and Zn concentrations showed no significant difference with increasing remediation time. In the intercropping experiment the phytoremediation efficiency in the treatment “S. plumbizincicola intercropped with maize” was higher than in S. plumbizincicola monocropping, and Cd concentrations of corn were below the maximum national limit. In the plant density experiment the phytoremediation efficiency increased with increasing plant density and 440,000 plants ha?1 gave the maximum rate. These results indicated that S. plumbizincicola at an appropriate planting density and intercropped with maize can achieve high remediation efficiency to contaminated soil without affecting the cereal crop productivity. This cropping system combines adequate agricultural production with soil heavy metal phytoextraction.  相似文献

In Tanzania, where tourist hunting is employed as a conservation tool for habitat protection, information on population sizes and hunting offtake was used to assess the impact of tourist hunting on mammal densities. In general, tourist hunting pressure was unrelated to local population sizes, but for most species, animals were removed at a level of less than 10% of the local population size, suggesting that over-exploitation was unlikely. Eland, however, and perhaps small antelope, bushbuck, kudu and reedbuck were hunted at levels which may be unsustainable in the long term. Analyses also identified areas of Tanzania with high levels of tourist hunting pressure, showed that, in certain areas, species with small population sizes such as eland could be declining as a result of tourist hunting, and suggested that current levels of lion and leopard offtake are too high. These findings, although preliminary, allow recommendations to be put forward for changing hunting quotas for certain species in particular areas of Tanzania.  相似文献

Adenoviruses are promising vectors for gene therapy and vaccination protocols. Consequently, the market demands for adenovirus are increasing, driving the search for new methodologies for large-scale production of concentrated vectors with warranted purity and efficacy, in a cost-effective way. Nevertheless, the production of adenovirus is currently limited by the so-called ‘cell density effect’, i.e. a drop in cell specific productivity concomitant with increased cell concentration at infection. Of two different serum-free culture media (CD293 and EX-Cell), evaluated for their effect on human 293 cells growth and adenovirus production at cell densities higher than 1×106 cells/ml, EX-Cell proved the better medium for cell growth. Although adenovirus production was equivalent in both media when the infection was performed at 1×106 cells/ml, at 3×106 cells/ml CD293 was the better. This result related to the high ammonia content in EX-Cell medium at the highest cell concentration at infection. Besides this, the large-scale production of these vectors at high cell densities often requires re-feed strategies, which increase medium osmolality. While a negative effect on cell growth was observed with increasing osmolalities, adenovirus productivity was only affected for osmolalities higher than 430 mOsm.  在不同土壤水分条件下,研究了不同种植密度(1、10、100、1 000、10 000株·m-2)的春小麦 (Triticum aestivum L.) 叶片气孔密度以及气孔分布的变化。结果表明:不同水分和种植密度不仅影响了叶片的气孔密度,而且也影响了其分布。随着种植密度的升高,气孔密度增大,在1 000株·m-2时气孔密度达最大值;当种植密度从1 000株·m-2上升到10 000株·m-2时,气孔密度下降;叶基部气孔数占全部气孔数的比例最大,叶尖最小,但随着种植密度的增大,气孔在叶片上的分布趋向均匀。干旱条件下随种植密度的升高气孔密度及分布的变化趋势与正常水分条件下的一致,但趋势更为明显。说明种植密度增大导致的气孔密度和分布的变化很可能是通过影响水分的供应而造成的。  相似文献
