The effect of surface adsorption on the structure and stability of proteins is a matter of increasing interest in biotechnology. Therefore, we have examined the effect of adsorption to silica on the thermal stability of 7 proteins employing differential scanning calorimetry (DSC) and front surface fluorescence (FSF) spectroscopy. In general, it was found that surface adsorption decreased the thermal stability of the bound protein. Using lysozyme for further studies, DSC, FSF, and FTIR spectroscopies, as well as enzymatic activity measurements, were used to explore the effect of decreasing surface apolarity on stability. It was observed that increasing surface apolarity produced decreasing stability and increasing structural alteration of the adsorbed protein. 相似文献

During the summer of 1983, cryptophytes, diatoms, cyanophytes, and the dinoflagellate, Ceratium hirundinella were most prominant among the phytoplankton of Eau Galle Reservoir. In the open water, cryptophytes and diatoms peaked in the spring, cyanophytes were most successful in the early summer, and Ceratium was dominant from mid-July until early August. In general, the sequence of events corresponded quite closely to the model of seasonal succession developed by the Plankton Ecology Group of the International Society of Limnology. To a large extent, the same pattern held in four experimental water columns. Departures from the model involved the roles of specific nutrients in diatom and cyanophyte periodicity. Diatoms began to yield to cyanophytes in late spring despite intermittent mixing and silica enrichment. Although capable of buoyancy regulation and thus well adapted to stable water columns, cyanophytes had greater increases in biomass in mixed columns, and in those columns, were most successful during a period of intermittent mixing. Cyanophyte success varied inversely with TN : TP ratios during the period of intermittent mixing, but not subsequently. By mid-July, Ceratium dominated the phytoplankton of every column except that of a mixed column in which conditions favored cyanophytes and large diatom species. 相似文献

The- distribution of stegmata in Orchidales is given for 130 species representing 105 genera. The stegmata occur in leaves and stems as longitudinal files of silica cells. lining fibre bundles or vascular bundle sheaths. The silica bodies contained in orchid stegmata arc of two major kinds, spherical and conical. The species investigated are characterized either by a single kind of silica body or by lack of silica cells. Absence of stegmata and presence of stegmata with conical bodies are the two most common character states in the order. The stegmata containing spherical bodies characterize Vandeae and Eriinae, Podochilinae, and Dendrobiinae within Epidendreae

An assay capable of detecting tens-of-picomole quantities of choline and acetylcholine in milliliter volumes of a physiological salt solution has been developed. Silica column chromatography was used to bind and separate 10–3000 pmol [14C]choline and [14C]acetylcholine standards made up in 3 ml of a bicarbonate-buffered Krebs-Ringer solution. The silica columns bound 95–98% of both choline and acetylcholine. Of the bound choline 84–87% was eluted in 1.5 ml of 0.075 n HCl, whereas 95–98% of the bound acetylcholine was eluted in a subsequent wash with 1.5 ml of 0.030 n HCl in 10% 2-butanone. Vacuum centrifugation of the eluants yielded small white pellets with losses of choline and acetylcholine of only 1%. Dried pellets of unlabeled choline and acetylcholine standards were assayed radioenzymatically using [γ-32P]ATP, choline kinase, and acetylcholinesterase. The net disintegrations per minute of choline[32P]phosphate product was proportional to both the acetylcholine (10–3000 pmol) and choline (30–3000 pmol) standards. The “limit sensitivity” was 8.5 pmol for acetylcholine and 11.4 pmol for choline. Cross-contamination of the choline assay by acetylcholine averaged 1.3%, whereas contamination of the acetylcholine assay by choline averaged 3.1%. 相似文献

The synthesis and chromatographic evaluation of two novel chiral stationary phases (CSPs) deriving from (S)-tyrosine are reported. The chiral graft has been designed in order to bear both π-acid and π-basic sites, each one being connected to a distinct asymmetric centre. An intramolecular π-π interaction may take place within these CSPs, leading to an energetically favoured conformation of the chiral selector (CS). The enantiorecognition ability of these CSPs was investigated for various classes of either π-acid or π-basic racemates. It is shown that these CSPs are able to separate simultaneously π-acid and π-basic racemates. Finally, chiral recognition mechanisms and mobile phase optimization are discussed. 相似文献

Using the unique quantitative capabilities of hyperspectral confocal microscopy combined with multivariate curve resolution, a comparative approach was employed to gain a deeper understanding of the different types of interactions of antimicrobial peptides (AMPs) with biological membranes and cellular compartments. This approach allowed direct comparison of the dynamics and local effects of buforin II, magainin II, and arenicin with nanoporous silica bead supported bilayers and living E. coli. Correlating between experiments and comparing these responses have yielded several important discoveries for pursuing the underlying biophysics of bacteriocidal specificity and the connection between structure and function in various cellular environments. First, a novel fluorescence method for direct comparison of a model and living system is demonstrated by utilizing the membrane partitioning and environmental sensitivity of propidium iodide. Second, measurements are presented comparing the temporal dynamics and local equilibrium concentrations of the different antimicrobial agents in the membrane and internal matrix of the described systems. Finally, we discuss how the data lead to a deeper understanding of the roles of membrane penetration and permeabilization in the action of these AMPs. The demand for new materials and products is still growing and the interest in naturally formed biopolymers and biominerals, such as chitin, calcium precipitates and silica is increasing. Photosynthesizing microalgae of the family Bacillariophyceae (diatoms) produce silica exoskeletons with a potential to be used in specific industrial or technological processes, they also are an excellent model in studies of silicon biomineralization. In contrast to geologically aged diatomaceous earth, the freshly prepared silica of cultured or harvested natural diatoms has been characterized insufficiently with respect to the properties (e.g. purity, specific surface area, porosity) required for technological and industrial application. In this contribution we summarize aspects of cellular processes that are involved in silicon biomineralization of diatoms and the current knowledge of the characterization of diatomaceous silica, following methods used for synthetically derived silica-based materials. 相似文献

BACKGROUND AND AIMS: Silicon (Si) in plants provides structural support and improves tolerance to diseases, drought and metal toxicity. Shoot Si concentrations are generally considered to be greater in monocotyledonous than in non-monocot plant species. The phylogenetic variation in the shoot Si concentration of plants reported in the primary literature has been quantified. METHODS: Studies were identified which reported Si concentrations in leaf or non-woody shoot tissues from at least two plant species growing in the same environment. Each study contained at least one species in common with another study. KEY RESULTS: Meta-analysis of the data revealed that, in general, ferns, gymnosperms and angiosperms accumulated less Si in their shoots than non-vascular plant species and horsetails. Within angiosperms and ferns, differences in shoot Si concentration between species grouped by their higher-level phylogenetic position were identified. Within the angiosperms, species from the commelinoid monocot orders Poales and Arecales accumulated substantially more Si in their shoots than species from other monocot clades. CONCLUSIONS: A high shoot Si concentration is not a general feature of monocot species. Information on the phylogenetic variation in shoot Si concentration may provide useful palaeoecological and archaeological information, and inform studies of the biogeochemical cycling of Si and those of the molecular genetics of Si uptake and transport in plants. 相似文献

以闽江口潮滩湿地为研究对象,由岸及海方向对芦苇、短叶茳芏、互花米草(潮沟内)和互花米草(潮沟外)4种湿地土壤生物硅和土壤间隙水氮硅营养盐含量及其随深度变化的特征进行为期1a的季度观测。结果显示:互花米草(外)、短叶茳芏、芦苇和互花米草(内)带湿地土壤生物硅的年均含量依次降低,分别为14.33、10.40、9.98、7.50 mg/g;互花米草(外)、互花米草(内)、短叶茳芏和芦苇带湿地土壤间隙水活性硅酸盐年均含量依次降低,分别为407、359、344、323μmol/L;湿地各植被带土壤及间隙水含硅量均呈现夏秋季节高于冬春季节的趋势。统计分析表明:间隙水活性硅酸盐与土壤生物硅含量、距潮沟的距离之间的正相关性均比较显著(P0.05),温度对土壤中硅含量的影响也有一定的正相关性,说明湿地植被、温度和潮汐作用是影响闽江口湿地硅分布的重要因素。与土著种对比,互花米草入侵在一定程度上改变了闽江口潮滩湿地土壤硅分布的格局。 相似文献
