Micropropagation of Fraxinus angustifolia Vahl has been successfully achieved both from mature and juvenile plant material using shoot tip and nodal explants. Several basal media supplemented with benzyladenine (BA) and indolebutyric acid (IBA) were tested for shoot proliferation. The most new explants per mature explant (5.3) was obtained on DKW medium plus 4.4 M BA+0.98 M IBA. The most new explants per juvenile explant (5.6) was produced on QL medium plus 8.9 M BA+0.49 M IBA. Rooting was achieved on WPM supplemented with 0.98–4.9 M IBA. Three-day-old cotyledonary explants of Pinus canariensis were subjected to 30 induction treatments using half-strength Bornman's medium containing various combinations of N6- benzyladenine, zeatin, kinetin and 2-isopentenyl-adenine. The highest numbers of buds were obtained with 10 M 6-benzyladenine, but both kinetin and zeatin influenced shoot elongation. Shoots were maintained on half-strength Schenk and Hildebrandt medium with 2% sucrose and 0.05% activated charcoal. For rooting, shoots were pulsed for 4 h in a 100 M indole-3-butyric acid aqueous solution (pH 4.2–4.5), and planted in peat:vermiculite:perlite (1:1:1). After 8 weeks, the numbers of rooted shoots were similar for most treatments. Therefore, the bud induction treatments did not significantly influence rooting of adventitious shoots of Canary Island pine.  相似文献

Flower development can be divided into four major steps: phase transition from vegetative to reproductive growth, formation of inflorescence meristem, formation and identity determination of floral organs, and growth and maturation of floral organs. Intercellular and intracellular signalling mechanisms must have important roles in each step of flower development, because it requires cell division, cell growth, and cell differentiation in a concerted fashion. Molecular genetic analysis of the process has started by isolation of a series of mutants with unusual flowering time, with aberrant structure in inflorescence and in flowers, and with no self-fertilization. At present more than 60 genes are identified from Arabidopsis thaliana and some of them have cloned. Although the information is still limited, several types of signalling systems are revealed. In this review, we summarize the present genetic aspects of the signalling network underlying the processes of flower development.  相似文献

The effects of the acylcyclohexanedione-type growth retardant prohexadione calcium on seedling growth and endogenous levels of immunoreactive phytohormone-like substances in shoots of wheat ( Triticum aestivum L. cv. Kanzler) and oilseed rape ( Brassica napus L. ssp. napus cv. Lirajet) were studied. After treatment of seedlings with increasing retardant concentrations in hydroponics, plant height and fresh weight of shoots were reduced by up to 40%. Concomitantly, the amount of immunoreactive gibberellins decreased, on a fresh weight basis, when compared with levels in the shoots of control plants. In contrast, the levels of abscisic acid and dihydrozeatin riboside and isopentenyladenosine-type cytokinins were considerably elevated by the growth retardant. The content of 3-indoleacetic acid decreased slightly. These results suggest that, in addition to its effect on gibberellin content, prohexadione calcium also influences the levels of endogenous abscisic acid and cytokinins.  相似文献

在昆明地区,纵坑切梢小蠹(Tomicus piniperda)成虫蛀梢多集中在蛀干木附近。 种群密度以蛀干木为中心向周围呈指数递减,散布半径约30m。在蛀梢过程中,该种群逐渐向新区扩张。在树冠内,纵坑切梢小蠹主要分布在4-10轮枝上。第7轮枝虫口百分率最高。6-7轮枝受害率最大。 树冠上层受害较其下层严重。从树冠水平层次考察,树冠外层虫量相对集中,约为树冠中、内层虫量之和。 树冠内层虫量最少。纵坑切梢小蠹在树冠内的种群分布系由梢径、种群密度、蛀梢行为、降落方式、光照等因素综合影响的结果。  相似文献

对7个生食葡萄品种的试管苗热处理结合茎尖剥离培养脱除病毒的方法做了研究,结果表明,从热处理试管苗剥离的茎尖培养成活率为61.2%,其中有18.1%的成苗。各品种热处理试管苗剥离的茎尖分化再生能力不同:先形成愈伤组织的成苗率较低,而先分化芽的茎尖成苗率较高。这一方法对扇叶病毒和卷叶病毒的脱除率达92%以上。  相似文献

Pot experiments were conducted with a calcareous soil (Inceptisol) to elucidate the effects of bicarbonate (0 and 20 mM) and root zone temperature (15° and 25°C) on the uptake of Zn, Fe, Mn and Cu by "Zn-efficient" and "Zn-inefficient" rice cultivars. Bicarbonate decreased concentrations and total uptake of Zn in shoots of "Zn-inefficient" cultivars, especially of IR 26 at 25°C, but not in Zn-efficient cultivars. Bicarbonate decreased concentrations and uptake of Fe in shoots of Zn inefficient cultivars, particularly in IR 26. Concentrations and total uptake of Mn were lower in bicarbonate treatment in the Zn-inefficient cultivars at 15°C, and in all cultivars at 25°C. However, concentration and uptake of Cu were not affected by bicarbonate in all cultivars. Compared to the 25°C root zone temperature, the concentrations and total uptake of both Zn and Cu in shoots at 15°C were lower in Zn-inefficient than in the Zn-efficient cultivars. The results indicate that Zn-efficiency in rice is causally related to high tolerance of plant to elavated bicarbonate concentrations in soil solution.  相似文献

Small birch plants were grown for up to 80 d in a climate chamber at varied relative addition rates of nitrogen in culture solution, and at ambient (350 μmol mol-1) or elevated (700 μmol mol-1) concentrations of CO2. The relative addition rate of nitrogen controlled relative growth rate accurately and independently of CO2 concentration at sub-optimum levels. During free access to nutrients, relative growth rate was higher at elevated CO2. Higher values of relative growth rate and net assimilation rate were associated with higher values of plant N-concentration. At all N-supply rates, elevated CO2 resulted in higher values of net assimilation rate, whereas leaf weight ratio was independent of CO2. Specific leaf area (and leaf area ratio) was less at higher CO2 and at lower rates of N-supply. Lower values of specific leaf area were partly because of starch accumulation. Nitrogen productivity (growth rate per unit plant nitrogen) was higher at elevated CO2. At sub-optimal N-supply, the higher net assimilation rate at elevated CO2 was offset by a lower leaf area ratio. Carbon dioxide did not affect root/shoot ratio, but a higher fraction of plant dry weight was found in roots at lower N-supply. In the treatment with lowest N-supply, five times as much root length was produced per amount of plant nitrogen in comparison with optimum plants. The specific fine root length at all N-supplies was greater at elevated CO2. These responses of the root system to lower N-supply and elevated CO2 may have a considerable bearing on the acquisition of nutrients in depleted soils at elevated CO2. The advantage of maintaining steady-state nutrition in small plants while investigating the effects of elevated CO2 on growth is emphasized.  相似文献
