Mutated polyadenylation signals for controlling expression levels of multiple genes in mammalian cells 总被引:1,自引:0,他引:1
A set of mutated SV40 early polyadenylation signals (SV40pA) with varying strengths is generated by mutating the AATAAA sequence in the wild-type SV40pA. They are shown to control the expression level of a gene over a 10-fold range using luciferase reporter genes in transient transfection assays. The relative strength of these SV40pA variants remains similar under three commonly used mammalian promoters and in five mammalian cell lines. Application of SV40pA variants for controlling expression level of multiple genes is demonstrated in a study of monoclonal antibody (mAb) synthesis in mammalian cells. By using SV40pA variants of different strengths, the expression of light chain (LC) and heavy chain (HC) genes encoded in a single vector is independently altered which results in different ratios of LC to HC expression spanning a range from 0.24 to 16.42. The changes in gene expression are determined by measuring mRNA levels and intracellular LC and HC polypeptides. It is found that a substantial decrease of HC expression, which increases the LC/HC mRNA ratio, only slightly reduces mAb production. However, reducing the LC expression by a similar magnitude, which decreases the LC/HC mRNA ratio results in a sharp decline of mAb production to trace amounts. This set of SV40pA variants offers a new tool for accurate control of the relative expression levels of multiple genes. It will have wide-ranging applications in fields related to the study of biosynthesis of multi-subunit proteins, proteomic research on protein interactions, and multi-gene metabolic engineering. 相似文献

Cell population balance (CPB) models can account for the phenotypic heterogeneity that characterizes isogenic cell populations. To utilize the predictive power of these models, however, we must determine the single-cell reaction and division rates as well as the partition probability density function of the cell population. These functions can be obtained through the Collins-Richmond inverse CPB modeling methodology, if we know the phenotypic distributions of (a) the overall cell population, (b) the dividing cell subpopulation, and (c) the newborn cell subpopulation. This study presents the development of a novel assay that combines fluorescence microscopy and image processing to determine these distributions. The method is generally applicable to rod-shaped cells dividing through the formation of a characteristic constriction. Morphological criteria were developed for the automatic identification of dividing cells and validated through direct comparison with manually obtained measurements. The newborn cell subpopulation was obtained from the corresponding dividing cell subpopulation by collecting information from the two compartments separated by the constriction. The method was applied to E. coli cells carrying the genetic toggle network with a green fluorescent marker. Our measurements for the overall cell population were in excellent agreement with the distributions obtained via flow cytometry. The new assay constitutes a powerful tool that can be used in conjunction with inverse CPB modeling to rigorously quantify single-cell behavior from data collected from highly heterogeneous cell populations. 相似文献

Ghrelin, an acylated peptide produced in the stomach, increases food intake and growth hormone secretion, suppresses inflammation and oxidative stress, and promotes cell survival and proliferation. We investigated the pharmacological potential of ghrelin in the treatment of polyneuropathy in uncontrolled streptozotocin (STZ)-induced diabetes in mice. Ghrelin or desacyl-ghrelin was administered daily for 4 weeks after STZ-induced diabetic polyneuropathy had developed. Ghrelin administration did not alter food intake, body weight gain, blood glucose levels, or plasma insulin levels when compared with mice given saline or desacyl-ghrelin administration. Ghrelin administration ameliorated reductions in motor and sensory nerve conduction velocities in diabetic mice and normalized their temperature sensation and plasma concentrations of 8-isoprostaglandin α, an oxidative stress marker. Desacyl-ghrelin failed to have any effect. Ghrelin administration in a mouse model of diabetes ameliorated polyneuropathy. Thus, ghrelin’s effects represent a novel therapeutic paradigm for the treatment of this otherwise intractable disorder. 相似文献

The review presents data on some peripheral and central structures in the system of perception of chemical stimuli in crayfish and other Decapoda. The hair receptors on chelipeds, antennas, and antennules are innervated by mechano- and chemoreceptor neurons. Antennules are crayfish specialized chemoreceptor organs whose surface contains groups of exteroceptors. On claws of ambulatory feet (AF), antennas, antennules, and other mobile appendages there is a regular disposition of exteroceptor receptive fields in the form of receptor hair bushes. Behavioral experiments have shown sensitivity of crayfish to odor of individuals of their gender and sex partners as well as the presence in crayfish of pheromones providing connection of female with offspring at the initial stages of their life cycle. Individual chemosensory cells innervating hair bushes on the crayfish AF respond to amino acids, amines, nucleotides, and sugars. Minimal thresholds of reaction of the studied Decapoda chemoreceptors in response to some chemical compounds correspond to 0.1–1.0 μM. For some chemoreceptors, dose-dependent effects have been shown. Alongside with monomodal chemoreceptors, the crayfish have bimodal receptors perceiving mechanical and chemical stimuli. The efficient response of crayfish chemoreceptors can be obtained to the substance that has amino group with hydrogen bridge to carboxyl group, contains no more that 3 carbon atoms in the chain, is characterized by a certain stereoform. Among chemoreceptors there are fast and slow adapting cells. Efficiency of response of individual chemoreceptors depends on temperature of medium. In crayfish, chemoreceptors responding to ecdysterons have been revealed. Ecdysterons play a great role in intra- and inter-species communications in Crustacea. Based on the study of efferent responses of interneurons of the first and higher orders in the first thoracic crayfish ganglion to stimulation of the own receptive fields, a concept has been put forward of the structural-functional organization of afferent projections at the segmental level. Peculiarities of afferent projections from antennule chemoreceptors are considered. The data are presented on connections of these chemoreceptors with antennular, olfactory, and accessory lobules, various cell groups, interneurons of the first, second, and third orders located in various brain parts. An attention has been drawn to connections of serotonin neurons in glomeruli with endings of chemoreceptor neurons and projections of interneurons of the higher orders, which are located in the internal medulla of the crayfish eyestalks. Several principles of integration of the chemoreceptor information in central parts of the crayfish nervous system are discussed. The giant serotonin neurons revealed in crayfish glomeruli most likely participate in formation of memory to certain chemical actions. Polymodal receptor signals in the central chain of the perception system activate autonomic centers, and the changes of the animal functional state can be evaluated from the heart responses. These responses recorded by novel noninvasive methods allow detection of the initial and other phases of the stress state at changes of the medium chemical quality. Progress of the current biochemical and electrophysiological methods of study chemoreceptors allow hoping for learning of fine chemoperception mechanisms in invertebrate and vertebrate animals. 相似文献

Enzymatic preparation of wheat bran xylooligosaccharides and their stability during pasteurization and autoclave sterilization at low pH 总被引:2,自引:0,他引:2
Xylooligosaccharides (XOS) were prepared from wheat bran insoluble dietary fiber (WBIDF) by treatment with commercial xylanase preparation Sunzymes. XOS, with a purity of 95% (w/w) and degree of polymerization of 2-7 and the ratio of arabinose to xylose of 0.27, was obtained with a yield of approximately 31.2% of WBIDF. Their stability was evaluated by comparing with that of commercial fructooligosaccharides (FOS) during pasteurization (60–100 °C, 30 min) and autoclave sterilization (121 °C, 1 kg/cm2, 10–50 min) at pH 2.0–4.0. XOS was characterized by a high thermal stability during pasteurization at pH 2.5–4.0 and sterilization at pH 3.0–4.0. Even at pH 2.0, the remaining XOS reached 97.2% (w/w) and 84.2% (w/w) during pasteurization (100 °C, 30 min) and sterilization (50 min), respectively. Compared with FOS, XOS was strongly resistant to lower acidic conditions. The results revealed that XOS was considered to be more suitable for use as functional food ingredients. 相似文献

A male zebra finch begins to learn to sing by memorizing a tutor's song during a sensitive period in juvenile development. Tutor song memorization requires molecular signaling within the auditory forebrain. Using microarray and in situ hybridizations, we tested whether the auditory forebrain at an age just before tutoring expresses a different set of genes compared with later life after song learning has ceased. Microarray analysis revealed differences in expression of thousands of genes in the male auditory forebrain at posthatch day 20 (P20) compared with adulthood. Furthermore, song playbacks had essentially no impact on gene expression in P20 auditory forebrain, but altered expression of hundreds of genes in adults. Most genes that were song‐responsive in adults were expressed at constitutively high levels at P20. Using in situ hybridization with a representative sample of 44 probes, we confirmed these effects and found that birds at P20 and P45 were similar in their gene expression patterns. Additionally, eight of the probes showed male–female differences in expression. We conclude that the developing auditory forebrain is in a very different molecular state from the adult, despite its relatively mature gross morphology and electrophysiological responsiveness to song stimuli. Under the network environment, the trading volume and asset price of a financial commodity or instrument are affected by various complicated factors. Machine learning and sentiment analysis provide powerful tools to collect a great deal of data from the website and retrieve useful information for effectively forecasting financial risk of associated companies. This article studies trading volume and asset price risk when sentimental financial information data are available using both sentiment analysis and popular machine learning approaches: artificial neural network (ANN) and support vector machine (SVM). Nonlinear GARCH-based mining models are developed by integrating GARCH (generalized autoregressive conditional heteroskedasticity) theory and ANN and SVM. Empirical studies in the U.S. stock market show that the proposed approach achieves favorable forecast performances. GARCH-based SVM outperforms GARCH-based ANN for volatility forecast, whereas GARCH-based ANN achieves a better forecast result for the volatility trend. Results also indicate a strong correlation between information sentiment and both trading volume and asset price volatility. 相似文献

A new member of the FHIT protein family, designated HIT-45, has been identified in the African trypanosome Trypanosoma brucei. Recombinant HIT-45 proteins were purified from trypanosomal and bacterial protein expression systems and analyzed for substrate specificity using various dinucleoside polyphosphates, including those that contain the 5′-mRNA cap, i.e., m7GMP. This enzyme exhibited typical dinucleoside triphosphatase activity (EC 3.6.1.29), having its highest specificity for diadenosine triphosphate (ApppA). However, the trypanosome enzyme contains a unique amino-terminal extension, and hydrolysis of cap dinucleotides with monomethylated guanosine or dimethylated guanosine always yielded m7GMP (or m2,7GMP) as one of the reaction products. Interestingly, m7Gpppm3N6, N6, 2′OA was preferred among the methylated substrates. This hypermethylated dinucleotide is unique to trypanosomes and may be an intermediate in the decay of cap 4, i.e., m7Gpppm3N6, N6, 2′OApm2′OApm2′OCpm2N3, 2′OU, that occurs in these organisms. 相似文献
