In the treatment of the soft tissue defect of the elbow, flap reconstruction is necessitated in many cases because of thinness of soft tissue at this region. In addition, reacquirement of tactile sensation is desirable because of the anatomical and specific functions of the elbow. Of three cases treated for elbow defects, one was reconstructed with a pedicled island forearm flap containing the lateral cutaneous nerve of the forearm, another was reconstructed with a venoneuro-accompanying artery fasciocutaneous flap (VNAF flap) containing the basilic vein, and the third with the VNAF flap containing the cephalic vein. The three cases demonstrated a sudden change of sensory territory 4 to 6 months after surgery, which was confirmed by touching the reconstructed region with patients'' eye-closed: from its original territory to the elbow in a “switching”-like action. Here we describe and discuss the concept of “sensory switching.”  相似文献

Channelrhodopsin-1 from Chlamydomonas augustae (CaChR1) is a light-activated cation channel, which is a promising optogenetic tool. We show by resonance Raman spectroscopy and retinal extraction followed by high pressure liquid chromatography (HPLC) that the isomeric ratio of all-trans to 13-cis of solubilized channelrhodopsin-1 is with 70:30 identical to channelrhodopsin-2 from Chlamydomonas reinhardtii (CrChR2). Critical frequency shifts in the retinal vibrations are identified in the Raman spectrum upon transition to the open (conductive P2380) state. Fourier transform infrared spectroscopy (FTIR) spectra indicate different structures of the open states in the two channelrhodopsins as reflected by the amide I bands and the protonation pattern of acidic amino acids.  相似文献

The new and vibrant field of optogenetics was founded by the seminal discovery of channelrhodopsin, the first light-gated cation channel. Despite the numerous applications that have revolutionised neurophysiology, the functional mechanism is far from understood on the molecular level. An arsenal of biophysical techniques has been established in the last decades of research on microbial rhodopsins. However, application of these techniques is hampered by the duration and the complexity of the photoreaction of channelrhodopsin compared with other microbial rhodopsins. A particular interest in resolving the molecular mechanism lies in the structural changes that lead to channel opening and closure. Here, we review the current structural and mechanistic knowledge that has been accomplished by integrating the static structure provided by X-ray crystallography and electron microscopy with time-resolved spectroscopic and electrophysiological techniques. The dynamical reactions of the chromophore are effectively coupled to structural changes of the protein, as shown by ultrafast spectroscopy. The hierarchical sequence of structural changes in the protein backbone that spans the time range from 10− 12 s to 10− 3 s prepares the channel to open and, consequently, cations can pass. Proton transfer reactions that are associated with channel gating have been resolved. In particular, glutamate 253 and aspartic acid 156 were identified as proton acceptor and donor to the retinal Schiff base. The reprotonation of the latter is the critical determinant for channel closure. The proton pathway that eventually leads to proton pumping is also discussed. Animal locomotion arises from complex interactions among sensory systems, processing of sensory information into patterns of motor output, the musculo-skeletal dynamics that follow motor stimulation, and the interaction of appendages and body parts with the environment. These processes conspire to produce motions and forces that permit stunning manoeuvres with important ecological and evolutionary consequences. Thus, the habitats that animals may exploit, their ability to escape predators or attack prey, their capacity to manoeuvre and turn, or the use of their available energy all depend upon the processes that determine locomotion. Here, we summarize a series of 10 papers focused on this integrative research topic.  相似文献

Understanding what determines feeding behaviour in herbivores is essential to optimise the use of forages in breeding systems. Herbivores can evaluate foods by associative learning of their pre-ingestive characteristics (taste, odour, etc.) and their post-ingestive consequences. Post-ingestive consequences are acknowledged as influencing intake and food choices, but the role of pre-ingestive characteristics is still being debated. Our experiment was designed to test their separate effects on daily dry matter intake (DMI), intake patterns and short-term choices in sheep by crossing the nature of the hay orally consumed (o) ad libitum, lucerne (L) or grass (G), with the nature of the hay introduced into the rumen (r), L or G, at a rate of half the total amount of hay received the day before. We applied four treatments, Go/Gr, Go/Lr, Lo/Gr and Lo/Lr, to test the effects of (i) post-ingestive consequences with similar pre-ingestive characteristics (Go/Gr v. Go/Lr; Lo/Gr v. Lo/Lr) and (ii) pre-ingestive characteristics with similar post-ingestive consequences at the end of the feeding period (Go/Lr v. Lo/Gr). Six rumen-fistulated sheep underwent all the treatments over 11-day periods in a latin square design. Eating time was restricted to 6 h/day, intraruminal introductions were performed just before food offer and choice tests were conducted after food removal. For similar pre-ingestive characteristics, DMI increased when L hay was introduced into the rumen rather than G (P < 0.05), possibly owing to a lower fill effect of L due to its lower NDF content and higher rumen degradability. The increased DMI resulted from longer eating time when G was orally consumed (149 v. 192 min, P < 0.05), whereas it resulted from higher intake rate with L (4.8 v. 6.1 g/min, P < 0.05). For similar post-ingestive consequences at the end of the feeding period (Go/Lr and Lo/Gr), DMI were similar (P > 0.05). Pre-ingestive characteristics or palatability per se did not therefore influence daily intake, although they influenced eating patterns. Pre-ingestive characteristics also greatly influenced short-term choices in favour of the hay that was not previously consumed, independently of any post-ingestive influence. This study confirms the effects of post-ingestive consequences on daily intake, but demonstrates that these variations are obtained by different behavioural adjustments under the influence of pre-ingestive characteristics. Preference for novelty, regardless of post-ingestive consequences, thus suggests that sheep may seek a diverse diet more for pleasure than for functional purposes, with implications for animal welfare.  相似文献
