Abstract Sulfate-reducing bacteria were enumerated in soils and water samples from Senegal ricefields using lactate and sulfate as substrates. When rice plants were severely injured by sulfides, maximum densities ranged from 107 to 109 bacteria g−1 of dry spermosphere or rhizosphere soil. Seven non-sporulating, mesophilic strains were isolated. The strains had motile curved cells and stained Gram-negative. Lactate, pyruvate, H2+ CO2, malate, fumarate, or ethanol could serve as electron donors. Organic acids were incompletely oxidized to acetate. Alcohols were degraded to the corresponding fatty acids. Sulfate, sulfite, or thiosulfate could serve as electron acceptors and were reduced to sulfide. Vitamins, yeast extract, Biotrypcase, or additional NaCl were not required for growth. On the basis of morphological and physiological properties, and the G + C mol % of the DNA, six isolates were identified as Desulfovibrio vulgaris and one as Desulfovibrio desulfuricans . The comparison of their main physiological properties with the physico-chemical properties of sampling sites indicated that they were better adapted to conditions prevailing in the rice rhizosphere than to those prevailing in the bulk of soil.  相似文献

Pseudomonas fluorescens and Rhizobium showed similar responses to Al in defined medium. Evidence that DNA is a site of action of Al in Rhizobium, and for mutagenic effects of Al in laboratory media, is reviewed. Preliminary data are presented which indicate that Al tolerance is not a plasmid-borne trait in these bacteria. The possible role of Al in the ecology of bacteria in soil is discussed in relation to the neutralisation of Al brought about by changes in soil pH by bacterial metabolism.  相似文献

The objective of the present work was to determine what impact extremely high nitrogen dosages would have on proline metabolism in order to use this amino acid as a bioindicator of N status of green bean plants (Phaseolus vulgaris L. cv. Strike). In this effort, we identified the most favourable pathway of proline synthesis under our experimental conditions. The N was applied to the nutrient solution in the form of NH4NO3 at 5.4 mmol/L (N1, optimal level), 11.6 mmol/L (N2), 17.4 mmol/L (N3), and 23.2 mmol/L (N4). Our results indicate that the application of high N dosages inPhaseolus is characterized by the accumulation of NO3, NH4+ and proline in root and foliar organs. However, although the enzymes in charge of proline biosynthesis, ornithine-δ-aminotransferase (OAT, EC 2.6.1.13) and Δ1-pyrroline-5-carboxylate synthetase (P5CS, EC 2.7.2.11/1.2.2.41) vary in behaviour depending on the N status, in our experiment, this amino acid appears to be synthesized mainly by the enzyme ornithine-δ-aminotransferase. This suggests predominance of the ornithine pathway over the glutamine pathway. Finally, under our experimental conditions, proline can be defined as a good indicator of N excess of green bean plants.  相似文献

In vertebrate species, cytotoxic H2O2 and other lipid or organic hydroperoxides (ROOH) formed in aerobic metabolism are removed by a selenoprotein, glutathione peroxidase (GPOX). The GPOX activity in most rat tissues ranges from 100 to 1,000 units (1 unit = 1 nmol NADPH oxidized·mg protein?1·min?1), except for muscles (20–30 units). In contrast, GPOX activities of two strains of the housefly (Musca domestica), cabbage looper (Trichoplusia ni), southern armyworm (Spodoptera eridania), and black swallowtail butterfly (Papilio polyxenes), were found to be in the range 2–12 units. Trivial GPOX activity was detected in the confused flour beetle (Tribolium confusum). In the earthworm (Lumbricus terrestris), banana slug (Ariolimax columbianus), and market squid (Loligo opalescens), the GPOX activity ranged from 1 to 5 units. Tissue selenium concentrations were about 500–1,000 ppb for adult M. domestica, 600 ppb in T. confusum, 32 ppb in T. ni, 17 ppb in S. eridania, and 31 ppb in P. polyxenes larvae. The form of selenium incorporated at such high levels in tissues of invertebrates such as M. domestica remains an unresolved issue. Peroxidase activity of non-selenium glutathione-S-transferase (GT) against ROOH may compensate for the low GPOX activity. Catalase (CAT) has high activity and wide subcellular distribution in insects. This may be an evolutionary adaptation to GT's inability to catalyze the reduction of H2O2. The GT's peroxidase and CAT activities were not assessed for other invertebrate species, and warrants an investigation due to their reported low GPOX levels.  相似文献

Toxicity of two plumbaginoids viz., plumbagin and juglone to the eggs of the cotton stainer,Dysdercus koenigii was studied by a residual film technique. Of these two, plumbagin showed the higher toxicity against different-age eggs with LC50 ranging from 0.0044 to 0.0066%. Eggs showed low susceptibility in the middle of embryogenesis. The toxicity of plumbaginoids, especially plumbagin, is discussed in relation to mode of action and prospects of its use as an ovicide in control of the insect.  相似文献

Mangrove ecosystems generate the major biodiversity hotspots of actinobacteria. Among the actinobacteria, Streptomyces species are the prolific producers of bioactive natural products. In this study, with research efforts to discover biopotential compounds from marine actinobacteria, 41 actinobacterial strains were isolated from sediment soil sample of Indian mangrove regions. The phylogeny prediction using the 16S rRNA gene sequences revealed that the isolates were related to Streptomyces. Isolates were further screened based on a two-step process wherein the first step, around nine strains, unveiled the presence of type 1 polyketide synthase gene and dTDP-glucose 4,6-dehydratase gene through polymerase chain reaction. As the second step of the screening process, cell viability assay was performed in RAW264.7 cells to assess the toxicity of extracts. Among all the isolates, Streptomyces rochei strain VITGAP173 was subjected to further analysis. To explore the bioactivities, the organic solvent extraction method was utilized to extract the broth culture of VITGAP173. Inhibition of nitric oxide and cyclooxygenase enzymes upon lipopolysaccharide-induced inflammation were utilized to evaluate the anti-inflammatory efficacy, and the results showed the potency of VITGAP173 in a dose-dependent manner. The extract significantly suppressed the messenger RNA levels of the inflammatory mediators such as tumor necrosis factor-α and interleukin-6 induced by lipopolysaccharide in RAW264.7 macrophages. The presence of several chemical constituents was identified through gas chromatography-mass spectrometry analysis of VITGAP173 extract. To achieve the toxicity analysis, oral administration of VITGAP173 extract in Wistar albino rats was carried out to investigate the biochemical parameters, histopathology which revealed its nontoxic nature.  相似文献

Laboratory experiments were conducted to determine the effects of M-One (Bacillus thuringiensis var.san diego) on larval instars ofColeomegilla maculata lengi Timberlake. Coccinellid larval development (from egg hatch to adult), completed on pollen treated with suspensions of M-One at 20 ml/litre (5.6×108 CPBIU/litre) and 200 ml/litre, took respectively 29.3 and 38.5 days compared with 21.9 days for the control (water). M-One did not cause larval mortality.C. maculata third instars did not show any preference between eggs ofLeptinotarsa decemlineata (Say) treated with water or with M-One at 20 ml/litre. However, at 200 ml M-One/litre, the number of eggs attacked was 34.7% lower than the eggs treated with water only, 48 h after the beginning of the test. These results indicate that the use of M-One, at the manufacturer's recommended field rate of 20 ml/litre, does not cause a major threat to larvalC. maculata populations.
Résumé Des bioessais en laboratoire ont été effectués afin de déterminer si les larves de la coccinelle maculée,Coleomegilla maculata lengi Timberlake (Col.: Coccinellidae), sont affectées par M-One, un insecticide biologique préparé à partir de la bactérieBacillus thuringiensis var.san diego Berliner et utilisé dans la lutte contre le doryphore de la pomme de terre,Leptinotarsa decemlineata (Say) (Col.: Chrysomelidae). Le développement larvaire, effectué sur du pollen traité avec des concentrations de 20 ml M-One/litre (5,6×108 unités internationales de doryphore/litre) et 200 ml M-One/litre, a nécessité 29,3 et 38,5 jours respectivement, comparativement à 21,9 jours pour le témoin (eau). M-One n'a pas causé de mortalité chez les larves. Au cours d'un test de 48 h, les larves de stade III n'ont montré aucune préférence entre des œufs traités avec 20 ml M-One/litre et des œufs traités avec de l'eau. Par contre avec 200 ml M-One/litre, le nombre d'œufs attaqués a diminué significativement de 34,7% par rapport au témoin, 48 h après le début du test. Ces résultats indiquent que l'utilisation de M-One à la concentration recommandée de 20 ml/litre ne représente pas une menace pour les populations larvaires de la coccinelle maculée.

Slow growth rate restricts the development and growth of seedlings due to nutrients deficiency or nutrient imbalance. Exponential fertilization can enhance the internal nutrient reserves in seedlings at the nursery-stage and strengthen their resistance to adverse conditions. In this study, nitrogen requirements for producing Hydrangea macrophylla ‘Hanatemari’ that robust seedlings, nutrient dynamics, biomass and growth, was examined utilizing exponential fertilization. The potted seedlings were fertilized with urea under exponential regime at rates of 0.5, 1.5 and 2.0 g nitrogen/plant (EF1, EF2, and EF3), respectively. In addition, an unfertilized group treated with equal volume of deionized water was used as control. The results showed that seedlings under 1.5 g N/plant (EF2) had the highest plant growth index and total biomass. The nutrient concentrations of different organs varied in different fertilization treatments. Based on the results of current study, it is concluded that 1.5 g N/plant (EF2) is suitable exponential fertilization treatment for the culture of hydrangea seedlings. Our treatments results showed that 2.0 g N/plant is not suitable for seedling culturing, because of serious nutrient toxicity. These findings will help to improve seedling quality and strengthen the production of H. macrophylla for plantation.  相似文献

[目的]明确噻虫嗪与噻虫胺2种新烟碱农药对鸟类的风险。[方法]采用经口灌胃法测定98%噻虫嗪或噻虫胺原药、30%噻虫嗪或20%噻虫胺悬浮剂、42%噻虫嗪或46%噻虫胺种子处理悬浮剂对日本鹌鹑的毒性等级。根据供试农药对鸟类的毒性数据,结合田间用药信息,开展了2种农药对鸟类的初级风险评估。[结果]98%噻虫嗪原药、30%噻虫嗪悬浮剂和42%噻虫嗪种子处理悬浮剂对日本鹌鹑的半致死剂量(7 d-LD50)分别为1500、481和266 mg·kg-1,毒性等级分别为低毒、中毒、中毒,悬浮剂和种子处理悬浮剂的毒性高于原药。98%噻虫胺原药、20%噻虫胺悬浮剂和46%噻虫胺种子处理悬浮剂对日本鹌鹑的7 d-LD50分别为369、397和1569 mg·kg-1,毒性等级分别为中毒、中毒和低毒,种子处理悬浮剂的毒性最低。噻虫嗪和噻虫胺的悬浮剂在水稻田间使用时,对鸟类急性、短期和长期暴露的风险商值均小于1,风险可接受;噻虫嗪和噻虫胺的种子处理悬浮剂在玉米田应用时,对鸟类急性、短期和长期暴露的风险商值均大于1,风险不可接受。[结论]噻虫嗪和噻虫胺的悬浮剂在田间应用时对鸟类相对安全,2种农药的种子处理悬浮剂可能对鸟类产生危害。在种子处理场景中,播种后尽可能覆土或覆膜,减少种子直接暴露被鸟类取食;在鸟类保护区域,应避免使用噻虫嗪和噻虫胺的种子处理悬浮剂,以减轻农药施用对鸟类的危害。  相似文献
