We examined the modifying effect of hypokinetic stress on the duration of behavioral phenomena in rats under conditions of experimentally induced tonic somatic, visceral, acute thermal, and electrostimulation-evoked pain. Stress of the above type (hypokinetic) was found to modify the pain sensitivity in rats related to all tested types of pain stresses of different etiology. Changes in the pain sensitivity of the animals under conditions of experimental pain tests depended on the duration of mobility restriction and could demonstrate opposite directions. Alpha-chymotrypsin was made more hydrophilic by modifying 11 (out of 16) ε-amino groups with pyromellitic dianhydride. The hydrophilic preparation was precipitated with n-propanol. This preparation gave significantly higher initial rates at the optimum aw (127.51 nmol mg?1 min?1 in n-octane and 21.30 nmol mg?1 min?1 in acetonitrile at aw=0.33) compared with the lyophilized preparation (53.50 nmol mg?1 min?1 in n-octane and 0.26 nmol mg?1 min?1 in acetonitrile at aw=0.97). FT-IR showed that the precipitate of modified alpha-chymotrypsin has a higher content of alpha-helices and beta-sheets compared to the lyophilized powder.  相似文献

Increasing photosynthetic efficiency is crucial to increasing biomass production to meet the growing demands for food and energy. Previous theoretical arithmetic analysis suggests that the light reactions and dark reactions are imperfectly coupled due to shortage of ATP supply, or accumulation of NADPH. Here we hypothesized that solely increasing NADPH consumption might improve the coupling of light reactions and dark reactions, thereby increasing the photosynthetic efficiency and biomass production. To test this hypothesis, an NADPH consumption pathway was constructed in cyanobacterium Synechocystis sp. PCC 6803. The resulting extra NADPH-consuming mutant grew much faster and achieved a higher biomass concentration. Analyses of photosynthesis characteristics showed the activities of photosystem II and photosystem I and the light saturation point of the NADPH-consuming mutant all significantly increased. Thus, we demonstrated that introducing extra NADPH consumption ability is a promising strategy to increase photosynthetic efficiency and to enable utilization of high-intensity lights.  相似文献

The 5-methylcytosine (5mC) modification regulates multiple cellular processes and is faithfully maintained following DNA replication. In addition to DNA methyltransferase (DNMT) family proteins, ubiquitin-like PHD and ring finger domain-containing protein 1 (UHRF1) plays an important role in the maintenance of 5mC levels. Loss of UHRF1 abolishes 5mC in cells and leads to embryonic lethality in mice. Interestingly, UHRF1 has a paralog, UHRF2, that has similar sequence and domain architecture, but its biologic function is not clear. Here, we have generated Uhrf2 knockout mice and characterized the role of UHRF2 in vivo. Uhrf2 knockout mice are viable, but the adult mice develop frequent spontaneous seizures and display abnormal electrical activities in brain. Despite no global DNA methylation changes, 5mC levels are decreased at certain genomic loci in the brains of Uhrf2 knockout mice. Therefore, our study has revealed a unique role of UHRF2 in the maintenance of local 5mC levels in brain that is distinct from that of its paralog UHRF1.  相似文献

Dopamine and related compounds are known to be toxic to melanoma cells. Some of their toxicity may be related, in part, to the oxidation products generated from them upon their interaction with melanogenic enzymes. In this paper, we present our studies on the oxidation chemistry of 3,4-dihydroxybenzylamine, the lower homolog of dopamine. Mushroom tyrosinase catalyzed oxidation of 3,4-dihydroxybenzylamine rapidly generated the corresponding quinone. However, aminomethyl-o-benzoquinone thus formed did not accumulate in the reaction mixture, but readily transformed to another product that exhibited absorbance maxima at 280 and 310 nm. This compound was identified to be 3,4-dihydroxybenzaldehyde based on its HPLC elution profile, cochromatography with authentic sample and UV spectral properties. Possible mechanism for the formation 3,4-dihydroxybenzaldehyde from 3,4-dihydroxybenzylamine and the nature of cytotoxic quinonoid intermediates formed are discussed.  相似文献

The effect of 2-(3-chloro-4-trifluoromethyl)anilino-3,5-dinitrothiophene (ANT-2p), known to be the most powerful ADRY agent (Renger, G. (1972) Biochim. Biophys. Acta 256, 428–439), on thermoluminescence has been investigated. Two thermoluminescence bands were analyzed: (a) the emission peaking at about 20–30°C caused by warming up of untreated chloroplasts, illuminated with a single 5 μs flash at room temperature and frozen rapidly to 77 K; and (b) the band emitted in the range of ?10 up 10°C after warming of chloroplast suspensions containing 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) which were illuminated with a single 5 μs flash at ?15°C and frozen rapidly at 77 K. These bands were attributed to the recombination of the B ?S2(S3) and X-320 ?S2 states, respectively (Rutherford, A.W., Crofts, A.R. and Inoue, Y. (1982) Biochim. Biophys. Acta 682, 457–465). It was found that: (1) The B ?S2(S3) band is markedly diminished at very low ANT-2p concentrations of less than one molecule per 2000 chlorophylls. (2) The inhibition of the X-320 ?S2 band requires significantly higher concentrations of ANT-2p (50% peak reduction at one ANT-2p molecule per 100 chlorophylls). (3) Preflashing at room temperature before cooling to ?15°C diminishes the X-320 ?S2 band significantly in the presence of ANT-2p, while almost no effect is observed in its absence. (4) The state X-320 ?S2 decays monoexponentially with a half-lifetime of 2 min at ?15°C in the absence of ANT-2p. In the presence of one ANT-2p molecule per 800 chlorophylls the decay becomes biphasic with half-lifetimes of 0.5 and 2 min and an amplitude ratio of 2:3, respectively. The results obtained can be explained consistently by the function of ANT-2p as an ADRY agent acting as a mobile species within the thylakoid membrane at room temperature. At subzero temperatures, a ‘fixed-place’ mechanism appears to be operative. The implications for the ADRY effect and thermoluminescence are discussed.  相似文献
