We have previously reported that the isolated frog corneal epithelium (a Cl–-secreting epithelium) has a large diffusional water permeability (Pdw 1.8×10–4 cm/s). We now report that the presence of Cl– in the apical-side bathing solution increases the diffusional water flux, Jdw (in both directions) by 63% from 11.3 to 18.4 l min–1 · cm–2 with 60 mm [Cl] exerting the maximum effect. The presence of Cl– in the basolateral-side bathing solution had no effect on the water flux. In Cl–-free solutions amphotericin B increased Jdw by 29% but only by 3% in Cl–-rich apical-side bathing solution, suggesting that in Cl–-rich apical side bathing solution, the apical barrier is no longer rate limiting. Apical Br– (75 mm) also increased Jdw by 68%. The effect of Cl– on Jdw was observed within 1 min after its addition to the apicalside bathing solution. HgCl2 (0.5 mm) reduced the Cl–-increased Pdw by 31%. The osmotic permeability (Pf) was also measured under an osmotic gradient yielding values of 0.34 and 2.88 (x 10–3 cm/s) in Cl–-free and Cl–-rich apical-side bathing solutions respectively. It seems that apical Cl–, or Cl– secretion into the apical bath could activate normally present but inactive water channels. In the absence of Cl–, water permeability of the apical membrane seems to be limited to the permeability of the lipid bilayer.This work was supported by National Eye Institute grants EY-00160 and EY-01867. 相似文献

Enantioselective HPLC methods have been developed for the resolution of (RS)-2-phenylcyclohexanone (compound 1) and (RS)-2-phenyltetrahydropyran-4-one (compound 4) and the diastereoselective and enantioselective separations of their respective cis- and trans-alcohols; reduction of compound 1 yields trans- and cis-2-phenyl-1-cyclohexanol (compounds 2 and 3, respectively) and reduction of compound 4 yields trans- and cis-2-phenyl-tetrahydropyran-4-ol (compounds 5 and 6, respectively). Compounds 1, 2, and 3 were stereochemically resolved using a chiral stationary phase (CSP) based upon amylose tris(3,5-dimethylphenyl carbamate) coated on 10 μm silica-gel (Chiralpak AD-CSP). Compounds 4, 5, and 6 were stereochemically resolved on a coupled column system where a column containing a CSP based upon cellulose tris(3,5-dimethylphenyl carbamate) coated on 5 μm silica (Chiralcel OD-H-CSP) was coupled in series to the AD-CSP. This study focused on effects from Monoporeia affinis reworking and ventilation activities on benthic fluxes and mineralization processes during a simulated bloom event. The importance of M. affinis density for benthic solute (O2, ΣNO2− + NO3−, NH4+ and HPO42−) fluxes and sediment reactivity (mobilization of NH4+ and HPO42−) following additions of organic material to the sediment surface was experimentally investigated using sediment-water and closed sediment (jar) incubations. Three different densities of M. affinis were used to resemble a low, medium and high density situation (1300, 2500 and 6400 ind. m− 2, respectively) of a natural amphipod community. The degradation of phytodetritus (Tetraselmis sp., 5 g C m− 2) added to the sediment surface was followed over a period of 20 days. Benthic solute fluxes of O2, ΣNO2− + NO3− and NH4+ were generally progressively stimulated with increasing number of M. affinis, while no such correlation was found for HPO42−. Solute fluxes were initially enhanced 1 to 2 days after the addition of phytodetritius, caused by mineralization of the most labile organic material and a food-stimulated irrigation by the amphipods. There was no effect from the activity of M. affinis on total denitrification (Dtot = Dn + Dw) or denitrification utilizing nitrate from coupled nitrification/denitrification (Dn) for any of the densities examined. Denitrification utilizing overlying water nitrate (Dw) was only about 10% of Dtot. Dw was significantly enhanced for the highest M. affinis density investigated. The reactivity of the sediment decreased progressively with increasing density of M. affinis and with time of the experiment. However, enhanced ammonium production at least 6 days after the organic addition indicated excretion of N-containing organic compounds by M. affinis. In conclusion, large spatial and temporal variations in density of M. affinis may be of significant importance for benthic solute fluxes and overall mineralization of organic material in Baltic Sea sediments. 相似文献

Soil and atmospheric droughts increasingly threaten plant survival and productivity around the world. Yet, conceptual gaps constrain our ability to predict ecosystem-scale drought impacts under climate change. Here, we introduce the ecosystem wilting point (ΨEWP), a property that integrates the drought response of an ecosystem's plant community across the soil–plant–atmosphere continuum. Specifically, ΨEWP defines a threshold below which the capacity of the root system to extract soil water and the ability of the leaves to maintain stomatal function are strongly diminished. We combined ecosystem flux and leaf water potential measurements to derive the ΨEWP of a Quercus-Carya forest from an “ecosystem pressure–volume (PV) curve,” which is analogous to the tissue-level technique. When community predawn leaf water potential (Ψpd) was above ΨEWP (=−2.0 MPa), the forest was highly responsive to environmental dynamics. When Ψpd fell below ΨEWP, the forest became insensitive to environmental variation and was a net source of carbon dioxide for nearly 2 months. Thus, ΨEWP is a threshold defining marked shifts in ecosystem functional state. Though there was rainfall-induced recovery of ecosystem gas exchange following soaking rains, a legacy of structural and physiological damage inhibited canopy photosynthetic capacity. Although over 16 growing seasons, only 10% of Ψpd observations fell below ΨEWP, the forest is commonly only 2–4 weeks of intense drought away from reaching ΨEWP, and thus highly reliant on frequent rainfall to replenish the soil water supply. We propose, based on a bottom-up analysis of root density profiles and soil moisture characteristic curves, that soil water acquisition capacity is the major determinant of ΨEWP, and species in an ecosystem require compatible leaf-level traits such as turgor loss point so that leaf wilting is coordinated with the inability to extract further water from the soil. 相似文献

Continuous light can be used as a tool to understand the diurnal rhythm of plants and it can also be used to increase the plant production. In the present research, we aimed to investigate the photosynthetic performance of V. radiata under continuous light as compared with the plants grown under normal light duration. Chlorophyll a fluorescence transient (OJIP test) technique was used to understand the effect on various stages of photosynthesis and their consequences under continuous light condition. Various Chl a Fluorescence kinetic parameters such as Specific energy fluxes (per QA-reducing PSII reaction center (RC)) (ABS /RC; TR0/RC; ET0/RC; DI0/RC), phenomenological fluxes, leaf model, (ABS/CSm; TR/CSm; ETo/CSm), Quantum yields and efficiencies (φPo; φEo; Ψo) and Performance index (PIabs) was extracted and analysed in our investigation. Glycoproteins in human serum play fundamental roles in many biological processes, and also have clinical value as biomarkers for disease progression and treatment. In this study, we isolated glycoproteins from the sera of three healthy individuals and three lung adenocarcinoma patients using multilectin affinity chromatography. The recovered glycoproteins were subjected to treatment with peptide-N-glycosidase F (PNGase F) and in-gel digestion by trypsin. Tryptic peptides were analyzed by nano-LC coupled to ESI-MS/MS and the MS/MS spectra were processed by Bioworks 3.2 and an in-house bioinformatics tool, ProtAn. Approximately 90% of the proteins identified contained more than one potential glycosylation site. Comparison of the serum glycoproteome of healthy and adenocarcinoma individuals revealed 38 cancer-selective proteins. Among them, 60% have previously been reported as low abundance proteins in human sera. We identified several cancer-selective proteins that have been previously characterized as potential indicators of lung cancer in serum or plasma, including haptoglobin (HP), inter-alpha-trypsin inhibitor heavy chain 4 (ITI-H4), complement C3 precursor, and leucine-rich alpha-2-glycoprotein. In addition, plasma kallikrein (KLKB1) and inter-alpha-trypsin inhibitor heavy chain 3 (ITI-H3) were identified as being potentially elevated in the lung cancer group, and were validated by Western blot analysis. Furthermore, approximately 18 kDa plasma kallirein protein fragment was detected at high levels in 25 out of 28 adenocarcinoma patients, while one of the eight normal individuals showed moderate positive. BACKGROUND: Sepsis causes diaphragmatic dysfunction, which can lead to the development of respiratory failure. We previously reported that isoproterenol, non-selective beta-adrenergic agonist, improved contractility of the diaphragm in a septic rat model. Since beta(2)-adrenoceptor agonists are widely used in the treatment of chronic respiratory disease, we investigated the effect of terbutaline, a selective beta(2)-adrenergic agonist, on contractility of the septic rat diaphragm and the contribution of intracellular Ca(2+) to the effect of terbutaline in vitro. METHODS: Forty-eight rats were divided into a sham group (in which sham laparotomy was performed) and a CLP group (in which peritonitis was induced by cecal ligation and perforation). The left hemidiaphragm was removed at 16 h after the operation. The effect of terbutaline (10(-)(6) M) on contractility of the diaphragm was assessed by twitch characteristics (twitch tension, contraction time and contraction velocity) and force-frequency relationship. In addition, to investigate the role of calcium ions in the effect of terbutaline on contractility of the diaphragm, contractility of the diaphragm was assessed after the pre-incubation of the diaphragm with methoxy-verapamil (10(-)(5) M), Ca(2+)-free Krebs-Ringer's solution buffered with 2 mM of ethylene glycol tetra-acetic acid (EGTA), and ryanodine (10(-)(6) M). RESULTS: Terbutaline significantly improved twitch characteristics and force-frequency relationship of the diaphragm in the CLP group (P<0.01). Incubation with methoxy-verapamil or calcium-free solution with EGTA did not show any changes in the inotropic effect of terbutaline in the CLP group. However, incubation with ryanodine completely abolished the inotropic effect of terbutaline in the CLP group. CONCLUSIONS: The present study demonstrated that terbutaline increased contractility of the diaphragm in the septic rats. Since this inotropic effect was abolished by ryanodine administration, calcium release from the sarcoplasmic reticulum may contribute to the terbutaline-induced improvement in dysfunction of the septic diaphragm. 相似文献

Involvement of nitric oxide in elicitor-induced defense responses and secondary metabolism of Taxus chinensis cells 总被引:5,自引:0,他引:5
This work was to characterize the generation of nitric oxide (NO) in Taxus chinensis cells induced by a fungal elicitor extracted from Fusarium oxysporum mycelium and the signal role of NO in the elicitation of plant defense responses and secondary metabolite accumulation. The fungal elicitor at 10-100 microg/ml (carbohydrate equivalent) induced a rapid and dose-dependent NO production in the Taxus cell culture, which exhibited a biphasic time course, reaching the first plateau within 1 h and the second within 12 h of elicitor treatment. The NO donor sodium nitroprusside potentiated elicitor-induced H2O2 production and cell death but had little influence on elicitor-induced membrane K+ efflux and H+ influx (medium alkalinization). NO inhibitors Nomega-nitro-L-arginine and 2-phenyl-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide partially blocked the elicitor-induced H2O2 production and membrane ion fluxes. Moreover, the NO inhibitors suppressed elicitor-induced activation of phenylalanine ammonium-lyase and accumulation of diterpenoid taxanes (paclitaxel and baccatin III). These results suggest that NO plays a signal role in the elicitor-induced responses and secondary metabolism activities in the Taxus cells. 相似文献
