Participatory research documented the hunting yields of 59 households in five neighboring indigenous villages in western Panama. These households captured 2,580 kg of game over 8 months, with 47% of the harvest coming from agricultural areas. The quantity of game captured in anthropogenic habitats is influenced by the hunting strategies employed. Only 25% of game captured during hunting trips was captured in agricultural areas, as opposed to 93% while “awaiting” and 65% using traps. Reliance on different strategies is in turn dependent on age, gender, and access to firearms. I argue that garden hunting is not a response to game depletion, but rather a productive activity that is complementary to broader cultural and economic patterns, and that simultaneously protects crops from animal predation. The creation of heterogeneous habitat mosaics through shifting cultivation has played a key role in the relationship between people and wildlife in the humid neotropics, leading to adjustments in both animal foraging patterns and indigenous hunting practices.  相似文献

This article examines transformations associated with changes in resource use and land cover dynamics in the community of São Manoel, Maranhão state, in the eastern Brazilian Amazon. The shifting cultivator peasants in São Manoel integrate swidden fields for annual cropping, the extraction of babassu palm products, and pastures for cattle ranching. Since the early twentieth century, predominant vegetative cover patterns have been altered from species-rich mature forests to secondary succession with babassu dominant to pasture or swidden fields containing palm stands of various densities. A grounded political ecology of resource use in the area suggests that management strategies and the resulting land cover dynamics integrate site-specific decisions of peasant producers. I discuss the trajectory of production strategies in São Manoel since the establishment of the community in the 1920s, and identify the multiple dimensions affecting resource use and environmental outcomes, with an emphasis on the period following land struggles and the recovery of peasant tenure rights in the mid-1980s. The analysis indicates that socionatural trajectories that optimize resource use and address the socioeconomic needs of the community include the maintenance of palm/pastures associations.  相似文献

A novel approach to utilizing whey permeate, the cultivation of mycelia of the edible mushroom Pleurotus ostreatus, is introduced. Response surface analysis (RSA) was successfully applied to determine the combination of substrate concentration, temperature and pH that would result in a maximal mycelial extension rate under solid state cultivation. The conditions to maximize the mycelial extension rate were predicted to be 44 g lactose l−1, pH 6.0 and 24.2 °C. Subsequent verification of these levels agreed with model predictions.  相似文献

For large-scale applications in biotechnology, cultivation of mammalian cells in suspension is an essential prerequisite. Typically, suspension cultures are grown in glass spinner flasks filled to less than 50% of the nominal volume. We propose a superior system for suspension cultures of mammalian cells based on orbital shaker technology. We found that "square-shaped" bottles (square bottles) provide an inexpensive but efficient means to grow HEK-293 EBNA and CHO-DG44 cells to high density. Cultures in agitated 1-L square bottles exceeded the performance of cultures in spinner flasks, reaching densities up to 7 x 10(6) cells/mL for HEK-293 EBNA cells and 5 x 10(6) cells/mL for CHO-DG44 cells in comparison to (2.5-4) x 10(6) cells/mL for cultures of the same cells grown in spinner flasks. For 1-L square bottles, optimal cell growth and viability were observed with a filling volume of 30-40% of the nominal volume and an agitation speed of 130 rpm at a rotational diameter of 2.5 cm. Transient reporter gene expression following gene delivery by calcium phosphate-DNA co-precipitation was the same or slightly better for HEK-293 EBNA cells grown in square bottles as compared to spinner flasks. Reductions in cost, simplified handling, and better performance in cell growth and viability make the agitated square bottle a new and very promising tool for the cultivation of mammalian cells in suspension.  相似文献

A novel method was developed for the quantitative analysis of the microbial metabolome using a mixture of fully uniformly (U) (13)C-labeled metabolites as internal standard (IS) in the metabolite extraction procedure the subsequent liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) analysis. This mixture of fully U (13)C-labeled metabolites was extracted from biomass of Saccharomyces cerevisiae cultivated in a fed-batch fermentation on fully U (13)C-labeled substrates. The obtained labeled cell extract contained, in principle, the whole yeast metabolome, allowing the quantification of any intracellular metabolite of interest in S. cerevisiae. We have applied the labeled cell extract as IS in the analysis of glycolytic and tricarboxylic acid (TCA) cycle intermediates in S. cerevisiae sampled in both steady-state and transient conditions following a glucose pulse. The use of labeled IS effectively reduced errors due to variations occurring in the analysis and sample processing. As a result, the linearity of calibration lines and the precision of measurements were significantly improved. Coextraction of the labeled cell extract with the samples also eliminates the need to perform elaborate recovery checks for each metabolite to be analyzed. In conclusion, the method presented leads to less workload, more robustness, and a higher precision in metabolome analysis.  相似文献

Guanosine-3',5'-tetraphosphate (ppGpp) and sigmaS, two regulators of the starvation response of Escherichia coli, have received increasing attention for monitoring cell physiological changes in production processes, although both are difficult to quantify. The kinetics of cAMP formation and degradation were not yet investigated in such processes, although the complex regulation of cAMP by synthesis, release, and degradation in connection with straightforward methods for analysis renders it a highly informative target. Therefore, we followed the cAMP concentration in various nonrecombinant and in four different recombinant glucose-limited fed-batch processes in different production scales. The intracellular cAMP concentration increases strongly at the end of the batch phase. Most cAMP is released to the cultivation medium. The rates of accumulation and degradation of extracellular cAMP are growth-rate-dependent and show a distinct maximum at a growth rate of about 0.35 h(-1). At very low growth rates, below 0.05 h(-1), extracellular cAMP is not produced but rather degraded, independent of whether this low growth rate is caused by glucose limitation or by the high metabolic load of recombinant protein production. In contrast to intracellular cAMP, which is highly unstable, analysis of extracellular cAMP is simpler and the kinetics of accumulation and degradation reflect well the physiological situation, including unlimited growth, limitation, and severe starvation of a production host.  相似文献

In the present study, we investigated the effect of hillslope gradient on vegetation recovery on abandoned land of shifting cultivation In Hainan Island, south China, by measuring community composition and structure of 25-year-old secondary forest fallows along a hillslope gradient (up-, middle-, and down-slope position). A total of 49 733 free-standing woody plant stems higher than 10 cm and belonging to 170 species, 112 genera, and 57 families was found in the three l-hm2 investigation plots. Stem density was highest in the down-slope stand and lowest in the up-slope stand. Species richness and the Shannon-Wiener Index were both highest in the middle-slope stand, and lower In the down-slope and up-slope stands. The recovery forest fallows on different hiUslope positions were all dominated by a few species. The five most abundant species accounted for 70.1%, 58.8%, and 72.9% of total stem densities in stands in the up-, middle-, and down-slope positions, respectively. The five species with the greatest basal areas accounted for 74.5%, 84.3%, and 74.7% of total stem basal area for the up-, middle-, and down-slope positions, respectively. The number of low-density species (stem abundance less than five) Increased from the up-slope position downward. Of the nine local common species among three different functional groups, the short-lived pioneer species dominated the up-slope position, but long-lived pioneer species dominated the middle- and down-slope positions. The climax species of primary tropical lowland rain forest was found in the downslope position. Both the mean diameter at breast height (DBH) and mean height of the trees Increased with decreasing hillslope gradient. The stem density and basal area in different size classes were significantly different in stands In different slope positions. Our results indicated that the rate of secondary succession varies, even over small spatial scales caused by the hlllslope gradient, in early vegetation recovery.  相似文献

The use of embryonic stem cell (ESC) derived cells has emerged as a potential alternative treatment for a number of degenerative diseases, including musculoskeletal diseases. Conventional ESC culturing methods use fetal bovine serum (FBS) as a major supplemental component of culture media, which is undesirable for clinical applications. These cultures are usually performed in small‐scale static vessels (gelatin‐coated dishes), which limit the number of cells that can be generated. It is essential to develop effective, reproducible protocols for efficient scalable production of ESC‐derived cells. Here we present serum‐free bioreactor protocols for (1) expansion and (2) differentiation of embryonic stem cells to osteoblasts. Cultivation of mESCs in serum‐free media, supplemented with 15% knockout serum replacement (KSR) resulted in a 27.1‐ and 48.6‐fold expansion in static culture and suspension respectively by day 5 of culture. Further induction to osteoblasts with a differentiation cocktail was verified by up‐regulation of osterix and osteocalcin. Mineralization was also enhanced, as indicated by an increase in the calcium deposition by osteogenic cells by day 28. These results will serve as the basis for developing protocols with human ESCs as a new treatment alternative for musculoskeletal diseases. Biotechnol. The potential of facultative photosynthetic bacteria as producers of photosynthetic pigments, vitamins, coenzymes and other valuable products has been recognized for decades. However, mass cultivation under photosynthetic conditions is generally inefficient due to the inevitable limitation of light supply when cell densities become very high. The previous development of a new cultivation process for maximal expression of photosynthetic genes under semi‐aerobic dark conditions in common bioreactors offers a new perspective for utilizing the facultative photosynthetic bacterium Rhodospirillum rubrum for large‐scale applications. Based on this cultivation system, the present study aimed in determining the maximal achievable cell density of R. rubrum in a bioreactor, thereby providing a major milestone on the way to industrial bioprocesses. As a starting point, we focus on aerobic growth due to higher growth rates and more facile process control under this condition, with the option to extend the process by an anaerobic production phase. Process design and optimization were supported by an unstructured computational process model, based on mixed‐substrate kinetics. Key parameters for growth and process control were determined in shake‐flask experiments or estimated by simulation studies. For fed‐batch cultivation, a computer‐controlled exponential feed algorithm in combination with a pH‐stat element was implemented. As a result, a maximal cell density of 59 g cell dry weight (CDW) L?1 was obtained, representing so far not attainable cell densities for photosynthetic bacteria. The applied exponential fed‐batch methodology therefore enters a range which is commonly employed for industrial applications with microbial cells. The biochemical analysis of high cell density cultures revealed metabolic imbalances, such as the accumulation and excretion of tetrapyrrole intermediates of the bacteriochlorophyll biosynthetic pathway.
