Extracellular levels of norepinephrine (NE) and glutamate (Glu) in the ventral bed nucleus of the stria terminalis (vBNST) of saline- and chronic morphine-treated rats, with or without withdrawal, were studied by means of the in vivo microdialysis technique in anesthetized rats. In addition, the tissue concentration of NE was studied at different rostrocaudal levels of the vBNST. Chronic morphine treatment significantly increased extracellular levels of NE, but not Glu, in vBNST. At 48 h after naloxone-induced morphine withdrawal there was a further significant increase in the extracellular levels of NE, but not Glu, in vBNST. The presence of UK 14304, an alpha(2)-adrenergic agonist, induced a significant decrease in NE extracellular levels in all experimental groups. In contrast, UK 14304 induced a significant decrease in Glu extracellular levels only in saline-treated rats. The results also show that the vBNST presents a rostrocaudal gradient of NE and contains 9.4% of total brain NE. The increase in NE extracellular levels in vBNST induced by chronic morphine treatment and the further increase in NE levels 48 h after naloxone-induced morphine withdrawal suggest that NE in vBNST may be involved in the pharmacological effects of chronic morphine and withdrawal.  相似文献

Eating and drinking are motivated behaviors that are made up of coordinated sets of neuroendocrine, autonomic, and behavioral motor events. Although the spinal cord, hindbrain, and hypothalamus contain the motor neurons and circuitry sufficient to maintain the reflex parts of these motor events, inputs from the telencephalon are required to furnish the behavioral components with a motivated (goal-directed) character. Each of these motor events derives from the complex interaction of a variety of sensory inputs with groups of neural networks whose components are distributed throughout the brain and collectively support motor expression and coordination. At a first approximation based on a variety of data, these networks can be divided into three groups: networks that stimulate, those that inhibit, and those that disinhibit motor functions. A fourth contributor is the circadian timing signal that originates in the hypothalamic suprachiasmatic nucleus and provides the temporal anchor for the expression of all behaviors. This article discusses the nature of these networks using neuroanatomical (tract-tracing and neuropeptide in situ hybridization), endocrine, and behavioral evidence from a variety of experimental models. A persistent problem when studying the control of food intake from a neural systems perspective has been the difficulty in separating those neuronal changes that result in hunger from those that are as a consequence of eating. To address this problem, dehydration-associated anorexia is presented as a particularly useful experimental model because it can be used to distinguish between neural mechanisms underlying anorexia and those changes that occur as a consequence of anorexia. The article concludes by highlighting the potential role of neuropeptidergic action in the operation of these networks, using forebrain neuropeptidergic innervation of the parabrachial nucleus as an example.  相似文献

A mathematical model has been developed for immobilized enzyme-catalyzed kinetic resolution of racemate in a fixed-bed reactor in which the enzyme-catalyzed reaction (the irreversible uni-uni competitive Michaelis-Menten kinetics is chosen as an example) was coupled with intraparticle diffusion, external mass transfer, and axial dispersion. The effects of mass-transfer limitations, competitive inhibition of substrates, deactivation on the enzyme effective enantioselectivity, and the optical purity and yield of the desired product are examined quantitatively over a wide range of parameters using the orthogonal collocation method. For a first-order reaction, an analytical solution is derived from the mathematical model for slab-, cylindrical-, and spherical-enzyme supports. Based on the analytical solution for the steady-state resolution process, a new concise formulation is presented to predict quantitatively the mass-transfer limitations on enzyme effective enantioselectivity and optical purity and yield of the desired product for a continuous steady-state kinetic resolution process in a fixed-bed reactor.  相似文献

The scaled down system developed in Part I of this series was further validated by using a 1-cm diameter column for method development studies for the separation of two model proteins, alcohol dehydrogenase and -glucosidase, from unclarified yeast homogenate by hydrophobic interaction expanded bed chromatography based on the STREAMLINE matrix. The efficacy of solids removal and establishment of optimal binding and separation condition by stepwise elution were investigated. Equilibration of the EBA column and loading at high salt strengths affected the subsequent recovery of the two target proteins. Although good resolution between the target proteins could be achieved, peak tailing was found to be a consistent problem. The optimised separation protocol was scaled up 25-fold to a column diameter of 5.0 cm. The results were in good agreement with the run conducted in the 1-cm column, indicating the potential of using the small columns as an viable approach for method scouting and development studies.  相似文献

The double-headed inhibitor of -amylase and Proteinase K was purified from wheat germ using Cu(II)-StreamlineTM-chelating resin. The endogenous -amylase could be inactivated by heating. Followed by this step, both packed bed and expanded bed gave similar activity yield of around 83% and fold purification around 23. In the case of expanded bed, it was not necessary to separate precipitated protein before the chromatography. The purified preparation gave a single band on SDS–PAGE and the estimated molecular weight of 21 kDa was in agreement with the reported value for the inhibitor designated as PKI-3 in the literature.  相似文献

Anthropogenic nutrient enrichment is increasingly modifying community structure and ecosystem functioning in terrestrial and aquatic ecosystems. In marine ecosystems, the paradigm is that nutrient enrichment leads to a decline of seagrasses by stimulating epiphytic algal growth, which shades and overgrows seagrasses. This ignores the potential for herbivores, which graze upon epiphytic algae, to partially or wholly counter such nutrient effects. We conducted a field experiment to assess the role that the trochid gastropod Calthalotia fragum plays in reducing nutrient impacts on the seagrass, Posidonia australis, in an urbanized Australian estuary, Botany Bay, Sydney. In a field experiment, where nutrient loading and grazer density were orthogonally manipulated, nutrient enrichment failed to promote epiphyte biomass or diminish growth and primary productivity of P. australis. To the contrary, nutrient enrichment enhanced photosynthesis of the seagrass in plots where the grazer was present at higher density. Epiphytic growth was negatively affected by increased C. fragum density, while P. australis shoot growth was positively influenced. Thus, in this study system, grazing appears to play a much greater role in determining seagrass primary productivity and above‐ground growth than moderate nutrient loading, suggesting that the interaction between grazers and nutrients depends on the relative levels of each. Our study contributes to a growing body of literature suggesting that effects of nutrient loading on benthic assemblages are not universally negative, but are dependent on the biotic and abiotic setting.  相似文献

Soy protein concentrates and soy protein isolates act as ingredients in bakery, meat and dairy products, baby formulas, starting materials for spun textured vegetable products, and other nutritional supplements. In this study, the effectiveness of a liquid-solid circulating fluidized bed (LSCFB) ion exchanger is demonstrated for the recovery of soluble soy proteins from full fat and defatted soy flour. Under steady-state operating conditions, about 50% of the proteins could be recovered from the feed streams entering the ion exchanger. The LSCFB was shown to be a promising system for the recovery of soy protein from both defatted and full fat soy flour solutions. As the ion exchange process captures dissolved proteins, the system may offer a less damaging form of processing compared with the acid precipitation process where soy protein aggregates form and functionality is affected. In addition, the LSCFB allows simultaneous adsorption and desorption of the proteins allowing for a continuous operation. No prefiltration of feed containing suspended particles is required as well, because fluidization is used in place of packed bed technology to improve on current ion exchange processes.  相似文献

2010年4~5月,在甘肃省马鬃山地区调查研究了盘羊的卧息地生境选择,共测得盘羊利用样方81个,对照样方81个。通过比较表明,马鬃山地区的盘羊夏季卧息地主要选择高海拔、阳坡、中坡位、坡度大、红砂植被、碎石地、植被密度低、远离道路和居民点、接近水源和隐蔽物、隐蔽程度好的区域。逻辑斯蒂回归分析表明,海拔(OR值1.614,95%CI1.248~4.419)、坡向(95%CI1.682~3.745)、坡位(95%CI0.839~3.156)、坡度(OR值1.705,95%CI1.227~2.369)、植被类型(95%CI0.682~2.732)、距水源距离(OR值0.180,95%CI0.041~0.787)、距道路距离(OR值1.628,95%CI2.598~3.941)、隐蔽级(OR值0.911,95%CI0.857~0.967)是盘羊春季卧息地选择的主导因子。  相似文献

Bienzymatic production of laminaribiose from sucrose and glucose was combined with adsorption on zeolite BEA to introduce a first capture and purification step. Downstream processing including washing and desorption steps was characterized and optimized on a milliliter scale in batch mode. Results were then transferred to a packed bed system for enzymatic production and adsorption where the influence of adsorbent particle diameter on purity and productivity was evaluated. Finally, a continuous enzymatic production of laminaribiose was conducted over 10 days. The subsequent downstream processing of the loaded zeolites led to purities of over 0.5 gLaminaribiose gsugar?1 in the desorbate with a total productivity of 5.6 mgLaminaribiose Lenzyme bed?1 h?1 without the use of recycles.  相似文献

Nests built by wild chimpanzees (Pan troglodytes verus) were studied at the Fongoli research site in southeastern Senegal from January 2004-May 2004 to investigate the role of comfort in nest building behavior by relating measures of nest comfort and building effort. Nest comfort across zones of the nest surface were compared with construction effort for 25 nests. Several variables of nest comfort were assessed: (1) physical discomfort, (2) visible discomfort, and (3) softness. Physical discomfort was used as a representative measure of nest discomfort. Building effort was measured by (1) construction force, (2) complexity, and (3) added material. Spearman rank correlations compared Effort and Comfort measures for both whole nests and central versus edge zones. The results show that construction force and complexity do not influence comfort of the nest as a whole. Greater Construction force correlates with more nest edge discomfort, yet the central area shows no difference. More complex nests do result in a more comfortable central area in the nest. Nests built with greater force may result in more discomfort, whereas complexity may allow chimpanzees to maintain comfort in a central area for sleep. Chimpanzees may place additional leaves or twigs over hard branches, protruding from the nest surface after construction, to increase comfort of the central nest area. Functions of chimpanzee nest building are likely to be several, but these results suggest comfort is a factor in nest building behavior.  相似文献
