In this histological study, we assessed the role of mesenchymal stem cells (MSCs) in the healing process that takes place during the subacute phase of myocardial infarction in dogs. Seven days after occlusion of the left anterior descending coronary artery, adult mongrel dogs received 100 × 106 4′-6-diamidino-2-phenylindole (DAPI)-labeled allogenic bone marrow–derived MSCs by the transendocardial (TE, n=6) and intracoronary (IC, n=4) routes; control dogs (n=6) received no infusion. The dogs were euthanized at 21 days after occlusion. Hearts were excised and sliced from apex to base into four transverse sections, which were divided into nine segments. Paraffin sections from each segment were stained with hematoxylin and eosin, trichrome, picrosirius red, and antibodies against several extracellular matrix components. Frozen sections were immunostained for host cardiac phenotypical markers and analyzed by epifluorescence and deconvolution fluorescence microscopy (DFM). We found less unresolved necrotic myocardium and more extracellular matrix deposition in MSC-treated dogs than in controls 2 weeks after cell delivery. By DFM, no DAPI+ MSC nuclei were observed within native cardiac cells. MSCs delivered during the subacute phase of acute myocardial infarction positively affect healing, apparently by mechanisms other than differentiation into mature native cardiac cells. Fibroblast apoptosis plays a crucial role in normal and pathological scar formation and therefore we studied whether the putative apoptosis-inducing factor curcumin affects fibroblast apoptosis and may function as a novel therapeutic. We show that 25-μM curcumin causes fibroblast apoptosis and that this could be inhibited by co-administration of antioxidants N -acetyl- l -cysteine (NAC), biliverdin or bilirubin, suggesting that reactive oxygen species (ROS) are involved. This is supported by our observation that 25-μM curcumin caused the generation of ROS, which could be completely blocked by addition of NAC or bilirubin. Since biliverdin and bilirubin are downstream products of heme degradation by heme oxygenase (HO), it has been suggested that HO-activity protects against curcumin-induced apoptosis. Interestingly, exposure to curcumin maximally induced HO-1 protein and HO-activity at 10–15 μM, whereas, at a concentration of >20-μM curcumin HO-1-expression and HO-activity was negligible. NAC-mediated inhibition of 25-μM curcumin-induced apoptosis was demonstrated to act in part via restored HO-1-induction, since the rescuing effect of NAC could be reduced by inhibiting HO-activity. Moreover pre-induction of HO-1 using 5-μM curcumin protected fibroblasts against 25-μM curcumin-induced apoptosis. On a functional level, fibroblast-mediated collagen gel contraction, an in vitro wound contraction model, was completely prevented by 25-μM curcumin, while this could be reversed by co-incubation with NAC, an effect that was also partially HO-mediated. In conclusion, curcumin treatment in high doses (>25 μM) may provide a novel way to modulate pathological scar formation through the induction of fibroblast apoptosis, while antioxidants, HO-activity and its effector molecules act as a possible fine-tuning regulator.  相似文献

The majority of specimens of the scallop Chlamys opercularis found in Strangford Lough, County Down, Ireland, are infected with a rare and exotic ciliate Licnophora auerbachii. The ciliate is found predominantly on the eyes of the scallop which (together with numerous tentacles) are borne by the middle marginal folds of the mantle. The ciliates are found attached to both the clear cornea covering the lens and also the surrounding pigmented iris, their large food vacuoles often containing black consistently sized granules, particles derived from the pigmented epithelial cells of the scallop eye. These cells are abraded from the surface of the iris and become detached due to the action of the basal disc of L. auerbachii during its clamping and locomotory activities on the eye surface. Scanning electron microscope observations show that the foot region of the ciliate exerts both a suction and abrasive action on the eye and leaves a distinct “footprint” on the eye surface in the form of a saucer-shaped depression. In many of the heavily infected scallops there was evidence of damage to the eyes with consequent loss of pigment and in some instances surface distortion of the eye itself.  相似文献

Abstract. Natural Pinus resinosa (red pine) stands in Newfoundland are restricted to 22 small, dry, nutrient-poor sites. A short wildfire cycle (15 - 30 yr) of both surface and crown fire regulates stand perimeters and is the main factor in regulating stand development. At the nucleus of current stands < 100 yr old, a few trees > 200 yr occur, usually showing multiple fire scars. Stem char heights confirmed an increased flammability of the stand and tree mortality for fires moving in the upslope position, as well as for mixed Pinus resinosa-Picea mariana stands. All P. resinosa stands are severely nutrient-deficient. Leaf concentrations of N, P and K were below or near the reported critical values. Nutrient concentrations were highest three months after a surface fire, but dropped considerably one year later. A gradual increase to near post-fire levels is achieved four years after fire. Foliar nutrient concentrations were positively correlated with average width of the annual rings. Aspects of the ericaceous understory dynamics and its relation to P. resinosa regeneration are also discussed.  相似文献

In order to determine the mechanical resistance of several forest tree species to rockfall, an inventory of the type of damage sustained in an active rockfall corridor was carried out in the French Alps. The diameter, spatial position and type of damage incurred were measured in 423 trees. Only 5% of trees had sustained damage above a height of 1.3 m and in damaged trees, 66% of broken or uprooted trees were conifers. Larger trees were more likely to be wounded or dead than smaller trees, although the size of the wounds was relatively smaller in larger trees. The species with the least proportion of damage through stem breakage, uprooting or wounding was European beech (Fagus sylvatica L.). Winching tests were carried out on two conifer species, Norway spruce (Picea abies L.) and Silver fir (Abies alba Mill.), as well as European beech, in order to verify the hypothesis that beech was highly resistant to rockfall and that conifers were more susceptible to uprooting or stem breakage. Nineteen trees were winched downhill and the force necessary to cause failure was measured. The energy (E fail) required to break or uproot a tree was then calculated. Most Silver fir trees failed in the stem and Norway spruce usually failed through uprooting. European beech was either uprooted or broke in the stem and was twice as resistant to failure as Silver fir, and three times more resistant than Norway spruce. E fail was strongly related to stem diameter in European beech only, and was significantly higher in this species compared to Norway spruce. Results suggest that European beech would be a better species to plant with regards to protection against rockfall. Nevertheless, all types of different abiotic stresses on any particular alpine site should be considered by the forest manager, as planting only broadleaf species may compromise the protecting capacity of the forest e.g. in the case of snow avalanches.  相似文献

The formation of glial scar and cystic cavities restricts axon regeneration after spinal cord injury. Chondroitin sulphate proteoglycans (CSPGs) are regarded as the prominent inhibitory molecules in the glial scar, and their inhibitory effects may be abolished in part by chondroitinase ABC (ChABC), which can digest CSPGs. CSPGs are secreted mostly by reactive astrocytes, which form dense scar tissues. The intermediate filament protein vimentin underpins the cytoskeleton of reactive astrocytes. Previously we have shown that retroviruses carrying full-length antisense vimentin cDNA reduce reactive gliosis. Here we administered both antisense vimentin cDNA and ChABC to hemisected rat spinal cords. Using RT-PCR, Western blotting and immunohistochemistry, we found that the combined treatment reduced the formation of glial scar and cystic cavities through degrading CSPGs molecules and inhibiting intermediate filament proteins. The modified intra- and extra-cellular architecture may alter the physical and biochemical characteristics of the scar, and the combined therapy might be used to inhibit glial scar formation.  相似文献

The aging central nervous system (CNS) of mammals displays progressive limited regenerative abilities. Recovery after loss of neurons is extremely restricted in the aged brain. Many research models fall short in recapitulating mammalian aging hallmarks or have an impractically long lifespan. We established a traumatic brain injury model in the African turquoise killifish (Nothobranchius furzeri), a regeneration‐competent vertebrate that evolved to naturally age extremely fast. Stab‐wound injury of the aged killifish dorsal telencephalon unveils an impaired and incomplete regeneration response when compared to young individuals. In the young adult killifish, brain regeneration is mainly supported by atypical non‐glial progenitors, yet their proliferation capacity clearly declines with age. We identified a high inflammatory response and glial scarring to also underlie the hampered generation of new neurons in aged fish. These primary results will pave the way to unravel the factor age in relation to neurorepair, and to improve therapeutic strategies to restore the injured and/or diseased aged mammalian CNS.  相似文献

In this paper we reconstructed flood events in a small mountain stream (6.6 km long, elevation 1100–1950 m a.s.l.) in the Dolina Waksmundzka Valley in the Tatra Mountains in the Western Carpathians. This reconstruction was based on cross-dated flood scars found in Norway spruce trees growing along stream banks. The scars were most likely formed by woody debris and stones transported during flood events. Reconstructed flood years were then compared with climatic records collected at the nearest meteorological station.Fifty-eight scars were cross-dated indicating 17 years with flood events in the period between 1928 and 2005. The large number of reconstructed flood events proves that the Potok Waksmundzki stream discharge can be highly variable. The high mid-summer rainfall (approximately 300 mm or more per month) peaks in June, July, August and this period coincides with some of the flood scar formation. The high winter and spring precipitation (December–May) does not seem to induce floods. The rate of snow melting seems to be more important. The highest number of scars (33%) was formed in dormant season of 1957/1958. In April and May 1958 there was an unusually large difference between mean monthly temperatures, the highest recorded in the twentieth century. This probably led to an abnormally rapid snow melt. No one single climatic factor can be held responsible for all flood events. Intensive mid-summer rainfall as well as rapid snow melting may induce floods in the Dolina Waksmundzka Valley. Cross-dated scars have enabled past flood events to be detected, which are otherwise invisible from climatic data alone.  相似文献
