The static fluid mosaic model of biological membranes has been progressively complemented by a dynamic membrane model that includes phospholipid reordering in domains that are proposed to extend from nanometers to microns. Kinetic models for lipolytic enzymes have only been developed for homogeneous lipid phases. In this work, we develop a generalization of the well-known surface dilution kinetic theory to cases where, in a same lipid phase, both domain and nondomain phases coexist. Our model also allows understanding the changes in enzymatic activity due to a decrease of free substrate concentration when domains are induced by peptides. This lipid reordering and domain dynamics can affect the activity of lipolytic enzymes, and can provide a simple explanation for how basic peptides, with a strong direct interaction with acidic phospholipids (such as beta-amyloid peptide), may cause a complex modulation of the activities of many important enzymes in lipid signaling pathways.  相似文献

Presence of isolated beta or betagamma domains of streptokinase (SK) increased the catalytic activity of staphylokinase (SAK)-plasmin (Pm) complex up to 60%. In contrast, fusion of SK beta or betagamma domains with the C-terminal end of SAK drastically reduced the catalytic activity of the activator complex. The enhancement effect mediated by beta or betagamma domain on Pg activator activity of SAK-Pm complex was reduced greatly (45%) in the presence of isolated kringles of Pg, whereas, kringles did not change cofactor activity of SAK fusion proteins (carrying beta or betagamma domains) significantly. When catalytic activity of SAK-microPm (catalytic domain of Pm lacking kringle domains) complex was examined in the presence of isolated beta and betagamma domains, no enhancement effect on Pg activation was observed, whereas, enzyme complex formed between microplasmin and SAK fusion proteins (SAKbeta and SAKbetagamma) displayed 50-70% reduction in their catalytic activity. The present study, thus, suggests that the exogenously present beta and betagamma interact with Pg/Pm via kringle domains and elevate catalytic activity of SAK-Pm activator complex resulting in enhanced substrate Pg activation. Fusion of beta or betagamma domains with SAK might alter these intermolecular interactions resulting in attenuated functional activity of SAK.  相似文献

This study showed that four factors which stimulate transepithelial fluid secretion and inorganic ion transport across the main segment of the Malpighian tubules of Drosophila melanogaster also stimulate transepithelial secretion of the prototypical organic cation tetraethylammonium (TEA). TEA fluxes across the Malpighian tubules and gut were measured using a TEA-selective self-referencing (TEA-SeR) microelectrode. TEA flux across isolated Malpighian tubules was also measured using a TEA-selective microelectrode positioned in droplets of fluid secreted by tubules set up in a modified Ramsay assay. TEA flux was stimulated by the intracellular second messengers cAMP and cGMP, which increase the lumen-positive transepithelial potential (TEP), and also by tyramine and leucokinin-I (LK-I), which decrease TEP. The largest increase was measured in response to 1 micromol l-1 LK-I which increased transepithelial TEA flux by 72%. TEA flux in the lower tubule was stimulated slightly (13%) by 1 micromol l-1 tyramine but not by any of the other factors. TEA flux across the midgut was unaffected by cAMP, cGMP or tyramine. This is the first study to demonstrate the effects of insect diuretic factors and second messengers on excretion of organic cations.  相似文献

This paper analyses Charles Darwins bird collection and the ornithological knowledge he derived from it during the voyage of H.M.S. Beagle. Darwin collected 468 bird skins, 10 detached parts of the lesser rhea, and the nests and eggs of 16 different taxa as well as 14 whole birds and 4 parts of birds which he preserved in spirit. He labelled these specimens with a number tag only, cross-referring the number to a notebook entry. Partly because of his limited ornithological knowledge and partly because he was confronted at times with entirely unknown birds, Darwin was often unable to apply the correct generic designations and gave his South American specimens English and Spanish names from literature and the local tongues, as well as the scientific generic names of European birds. Back home, it was John Gould, the prominent ornithologist of the Zoological Society of London, who made sense of Darwins collection, among his many other scientific achievements correctly identifying the Galápagos finches as a group of closely related birds. Darwins bird collection did not receive much attention in the latter part of the 19th century. Most of the specimens had their original labels removed and replaced by ones of the custodian institution. Today, original Darwin specimens stemming from the Beagle voyage are to be found in at least eight different institutions, but almost half of the bird specimens Darwin collected on the Beagle voyage are not accounted for. The appendix to this paper lists for the first time all the birds which Darwin collected during the voyage. Darwins famous book On the origin of species hardly draws upon any ornithological examples from his voyage on the Beagle. Nevertheless, Darwin contributed much to ornithology. His collection contained 39 new species and subspecies of birds, mainly described by Gould, and some birds from populations now extinct, and he also made a few very good field observations, published in the sections of The Zoology of the Voyage of H.M.S. Beagle dedicated to birds.

Activation of the O(2)(-) generating NADPH oxidase of phagocytes results from the assembly of the membrane-bound flavocytochrome b(558) with cytosolic proteins, p67(phox), p47(phox), and Rac. However, it has been recently reported that the arachidonic acid- and calcium-binding heterodimer S100A8/A9, abundant in neutrophil cytosol, influences the activation process. In a semi-recombinant system comprising neutrophil membranes, recombinant proteins, p67(phox), p47(phox), GTPgamma S-loaded Rac2, and arachidonic acid (AA), both the rate and the extent of the oxidase activation were increased by S100A8/A9, provided it was preloaded with AA. Binding of [(14)C]AA to S100A8/A9 was potentiated by recombinant cytosolic phox proteins and GTPgammaS, suggesting the formation of a complex, comprising oxidase activating proteins and S100A8/A9, with a greater affinity for AA. The rate constant of oxidase activation was not increased by AA-loaded S100A8/A9, whereas the maximal oxidase activity elicited was twice as high. AA-loaded S100A8/A9 increases oxidase activation probably by decreasing the deactivation rate.  相似文献

Nitrogen fixation, nitrate assimilation and primary production ((13)C/(15)N method) were investigated during one year and half in the northwestern Mediterranean Sea. Nitrogen fixation was detectable all over the year with rates ranged from 2 to 17 nmol N l(-1) d(-1)(d). Highest values being obtained during spring associated with the phytoplankton bloom. High rates (4-8 nmol N l(-1) d(-1)(d)) were also measured during summer, when primary productivity was very low. Then, diazotrophy process supplies significant new nitrogen during summer oligotrophic periods. This new nitrogen input can balance the annual nitrogen biogeochemical budget in the Mediterranean Sea and should explain the high nitrate/phosphate ratio observed in deep waters.  相似文献

The classic Chengjiang Lagerstätte (Lower Cambrian, Atdabanian stage: Yu''anshan Formation) Yunnan, southwestern China, has yielded, besides the exceptional and often controversial soft-bodied fossils, a fauna of primitive/early lingulid brachiopods. Diandongia pista (Rong 1974) is one of the commonest and most strongly mineralized of the phosphatic brachiopods from the Lagerstätte. The shells of this species have been found to commonly serve as a basibiont host. Epibionts comprise the coeval brachiopod Longtancunella chengjiangensis and the cone-shaped cnidarian-related Archotuba conoidalis, as well as rounded smaller-sized epizoans (lesser than 2 mm). A principle morphological analysis demonstrates that the ovoid and rounded organisms that often occur along the commissure of D. pista resemble small juvenile or immature brachiopods. Epibiont-bearing shells of D. pista with soft-tissue preservation demonstrate that the host brachiopods were overgrown while alive, and provide an argument for D. pista having a semi-infaunal life style with only the slim pedicle embedded in sediment. The epibiotic association sheds direct light on the ecology of Cambrian brachiopods in soft-substrate marine environments. The Chengjiang fossils demonstrate that the Early Cambrian brachiopods, as compared with recent lingulids, occupied different and a wider spectrum of ecological niches and tiers of space.  相似文献

Marine palynological studies of Quaternary sediments usually focus on dinoflagellate and pollen assemblages for paleoceanographic and paleohydrographic interpretations of past events. This paper focuses on the use of palynofacies analysis for paleohydrological reconstructions of deltaic and deep sea environments to evaluate transport of organic matter to the ocean. These palynodebris data are used to interpret palaeoenvironmental and paleohydrographic changes in two marine cores from the continental shelf (core-1, 27 m water depth) and middle slope (core-2, 1030 m) offshore Egypt, south-eastern Mediterranean Sea, during the latest Pleistocene and the Holocene. The relative abundances of various types of sedimentary organic matter such as phytoclasts, zooclasts, amorphous organic matter and palynomorphs are related to paleohydrographic changes of the overlying water column. Based on the total palynodebris and organic carbon content, sediments of the inner continental shelf core are characteristic of a prodelta environment proximal to a fluvio-deltaic source and moderately distal oxic environments with enhanced structured organic matter preservation potential. In contrast, the palynodebris of the middle slope core show that the basal sediments (105-140 cm depth) indicate suboxic to dysoxic bottom water conditions, followed by anoxic-suboxic bottom water conditions for the interval from 30-85 cm, which represents the S1 Sapropel. The top sediments of core-2 (0-25 cm) were deposited under oxic bottom water conditions, suggesting good ventilation in the water column. A quantitative approach was also used for interpreting the Holocene sea-level changes, based on the correlation between phytoclast and organic matter abundances. Sedimentation rate in the continental shelf is varied, being relatively very low (6.7 cm/kyr) in the basal part and increased upward to be 20 cm/kyr (depth 115-120 cm). In the upper Gray clayey silt layer the sedimentation rate was high (about 40-45 cm/kyr) due to the high discharge from El Manzala Lagoon and Damietta Nile branch. In the middle slope depth the sedimentation rate was relatively low and uniform, around 14 cm/kyr.  相似文献

Questions: For wetland plants, dispersal by wind is often overlooked because dispersal by water is generally assumed to be the key dispersal process. This literature review addresses the role of seed dispersal by wind in wetlands. Why is wind dispersal relevant in wetlands? Which seeds are dispersed by wind and how far? And how can our understanding of wind dispersal be applied to wetland conservation and restoration? Methods: Literature review. Results and conclusions: Wind is a widely available seed dispersal vector in wetlands and can transport many seeds over long distances. Unlike water, wind can transport seeds in all directions and is therefore important for dispersal to upstream wetlands and to wetlands not connected by surface water flows. Wind dispersal transports seeds to a wider range of sites than water, and therefore reaches more sites but with lower seed densities. Many wetland plant species have adaptations to facilitate wind dispersal. Dispersal distances increase with decreasing falling velocity of seeds, increasing seed release height and selective release mechanisms. Depending on the adaptations, seeds may be dispersed by wind over many km or only a few m. The frequency of long‐distance wind dispersal events depends on these adaptations, the number of produced seeds, the structure of the surrounding vegetation, and the frequency of occurrence of suitable weather conditions. Humans reduce the frequency of successful long‐distance wind dispersal events in wetlands through wetland loss and fragmentation (which reduce the number and quality of seeds) and eutrophication (which changes the structure of the vegetation so that seed release into the wind flow becomes more difficult). This is yet another reason to focus on wetland conservation and restoration measures at increased population sizes, prevention of eutrophication, and the restoration of sites at short distances from seed sources.  相似文献

The molybdenum nitrogenase enzyme system, comprised of the MoFe protein and the Fe protein, catalyzes the reduction of atmospheric N(2) to NH(3). Interactions between these two proteins and between Fe protein and nucleotides (MgADP and MgATP) are crucial to catalysis. It is well established that salts are inhibitors of nitrogenase catalysis that target these interactions. However, the implications of salt effects are often overlooked. We have reexamined salt effects in light of a comprehensive framework for nitrogenase interactions to offer an in-depth analysis of the sources of salt inhibition and underlying apparent cooperativity. More importantly, we have identified patterns of salt activation of nitrogenase that correspond to at least two mechanisms. One of these mechanisms is that charge screening of MoFe protein-Fe protein interactions in the nitrogenase complex accelerates the rate of nitrogenase complex dissociation, which is the rate-limiting step of catalysis. This kind of salt activation operates under conditions of high catalytic activity and low salt concentrations that may resemble those found in vivo. While simple kinetic arguments are strong evidence for this kind of salt activation, further confirmation was sought by demonstrating that tight complexes that have previously displayed little or no activity due to the inability of Fe protein to dissociate from the complex are activated by the presence of salt. This occurs for the combination Azotobacter vinelandii MoFe protein with: (a) the L127Delta Fe protein; and (b) Clostridium pasteurianum Fe protein. The curvature of activation vs. salt implies a synergistic salt-protein interaction.  相似文献
