Pretreatment of pigeon erythrocyte membrane vesicles with amino acids, ATP, GTP, Pi and some other simple cell constituents (singly and in combination) causes an increase in ATP-dependent Ca2+-uptake activity of vesicles upon subsequent incubation with 45Ca2+ after removal of the above agents from the ‘i’ face. Amino acids augment the stimulation by all stimulatory agents and are required for stimulation by Pi. The effects of amino acids, ATP, GTP and Pi all occur at physiological concentrations. Many if not all of the effects of the mixture of amino acids that occur naturally in the cells can be accounted for by the group transported by the ‘ASC’ transport system of Christensen (Christensen H.N. (1975) Biological Transport, 2nd edn., W.A. Benjamin, Inc., Reading, MA), but not by any single amino acid at its physiological concentration. The effects of ATP and GTP are not mimicked by their non-hydrolysable β, γ-imido analogues nor by the corresponding 3′, 5′-cyclic monophosphates. None of the effects described appears to involve calmodulin. We suggest that amino acid transport plays a role in metabolic regulation through effects on cell [Ca2+]. Analogous effects on cell [Ca2+] may be involved in the action of the many hormones which augment amino acid accumulation by the ‘A’ amino acid transport system.  相似文献

The regulation of nucleotide excision repair and base excision repair by normal and repair deficient human cells was determined. Synchronous cultures of WI-38 normal diploid fibroblasts and Xeroderma pigmentosum fibroblasts (complementation group D) (XP-D) were used to investigate whether DNA repair pathways were modulated during the cell cycle. Two criteria were used: (1) unscheduled DNA synthesis (UDS) in the presence of hydroxyurea (HU) after exposure to UV light or after exposure to N-acetoxy-acetylaminofluorene (N-AcO-AAF) to quantitate nucleotide excision repair or UDS after exposure to methylmethane sulfonate (MMS) to measure base excision repair; (2) repair replication into parental DNA in the absence of HU after exposure to UV light. Nucleotide excision repair after UV irradiation was induced in WI-38 fibroblasts during the cell cycle reaching a maximum in cultures exposed 14–15 h after cell stimulation. Similar results were observed after exposure to N-AcO-AAF. DNA repair was increased 2–4-fold after UV exposure and was increased 3-fold after N-AcO-AAF exposure. In either instance nucleotide excision repair was sequentially stimulated prior to the enhancement of base excision repair which was stimulated prior to the induction of DNA replication. In contrast XP-D failed to induce nucleotide excision repair after UV irradiation at any interval in the cell cycle. However, base excision repair and DNA replication were stimulated comparable to that enhancement observed in WI-38 cells. The distinctive induction of nucleotide excision repair and base excision repair prior to the onset of DNA replication suggests that separate DNA repair complexes may be formed during the eucaryotic cell cycle.  相似文献

Individual adult Schistosoma mansoni from strains selected for high or low infectivity to specific strains of the snail intermediate host, Biomphalaria glabrata, were subjected to enzyme electrophoresis on starch gels. Fourteen enzyme systems were analyzed in an attempt to find electrophoretic markers associated with genes for infectivity to snails. The S. mansoni strains were selected from different isolates from Puerto Rico in several strains of B. glabrata. Of an estimated 18 loci, 3 were polymorphic and the remainder monomorphic. For 1 of the 3 polymorphic enzyme loci, lactate dehydrogenase (Ldh, EC 1.1.1.27), phenotype frequencies were correlated with infectivity to snails. In schistosome strains of low infectivity, frequencies of the Ldh-N phenotype ranged between 0.56 and 0.69, while in strains of high infectivity, Ldh-N frequencies were typically 0.91 to 1.00. Whether the correlation is accidental or due to some form of association, such as chromosomal linkage, between the locus responsible for variation in lactate dehydrogenase and a gene for infectivity to snails remains to be determined.  相似文献

Fluorescence photobleaching recovery methods reveal marked changes in lateral mobilities of rabbit lymphocyte membrane components during the course of stimulation with succinyl concanavalin A (S Con A). The diffusion constant of S Con A receptors on T lymphocytes falls from 1.6×10?10 cm2/sec to 6.5×10?11 cm2/sec within 4 hr after stimulation, remains constant for 14 hr, and returns to its former value. The mobility of B cell receptors similarly falls from 1.4×10?10 cm2/sec to 5.5×10?11 cm2/sec but regains its unstimulated value much more slowly. In contrast, a fluorescent phospholipid analog shows constant mobilities of 1.9×10?8 cm2/sec and 1.5×10?8 cm2/sec in T and B cells, respectively, throughout the experiment.  相似文献

On the basis of electron spin resonance results, the 1:1 Cu(II), Co(II), Co(II)-O2, and Ni(III) complexes of bleomycin(BLM) have been compared with the corresponding metal complexes of its biosynthetic intermediate(P-3A). The present study suggests that (1) P-3A is an useful ligand for the clarification of metal-binding sites of BLM; (2) the secondary amine, pyrimidine ring nitrogen, deprotonated peptide nitrogen of histidine residue, and histidine imidazole groups as planar ligand donors, and the α-amino group as axial donor, are substantially important for metal-coordination of BLM; and (3) the sugar and bithiazole portions of BLM probably contribute to stabilization of Co(II)-O2 adduct complex and axial sixth coordination of Cu(II) and Ni(III) complexes.  相似文献

Depending on the developmental stage, the gamma-glutamyltransferase (E.C. 2.3.2.2) exists in two different types in the liver and in the small intestine: a sialic acid-rich fetal type and a sialic acid-poor adult type. The fetal type could be detected in the undifferentiated cryptal cells, in the fetal small intestine and in the fetal liver, and the adult type in the differentiated villous cells and in the adult liver. The separation of both types was performed using ConA-sepharose, which does not bind the fetal type but the adult type. Binding was reached by neuraminidase treatment.  相似文献

Summary A quantitative method that optimizes the mapping of species diversity in phytogeographic studies is described. Diversity is computed on the basis of species number per unit area. The optimal size of unit area for which diversity is computed is held to be that which maximises the diversity difference between species-rich and species-poor regions. An example is given using Turkish Papaver. A very high correspondence is found between intuitive insights based on long study and the computer-generated diversity maps. Phytogeographic elements were also determined by computer after gridding Turkey at the scale discovered to be optimal for diversity and scoring the grid squares for presence-absence of each species. In this case, too, quite high correspondence was found between the computer and intuitive results.Nomenclature follows Cullen (1965).The authors express their thanks to Mr. K. The NADP+ specific glutamate dehydrogenase from wild-type Neurospora crassa forms a stable binary complex with NADPH. This can combine with L-glutamate, α-ketoglutarate or the substrate analogue D-glutamate to form ternary complexes which can be distinguished by their different fluorescence properties. The affinity of the enzyme for NADPH diminishes with increases in pH or ionic strength of the solution. Experimental data obtained using modified glutamate dehydrogenases from mutant strains of N. crassa suggest that the reduced-coenzyme binding sites observed fluorimetrically are the same as those observed by enzyme kinetics.  相似文献

In an attempt to increase tolerance to salinity stress in tobacco plants, the genes encoding the mutant form of glutamyl kinase (proB), pyrroline-5-carboxylate synthetase, and osmotin were cloned into three different shuttle vectors and were separately introduced into the tobacco plants. The transgenic lines were compared for their ability to produce shoots and grow in MS medium containing 320 mM NaCl; it was shown that the transgenic lines containing genetically handled osmotin gene are more resistant to salinity. The amount of chlorophyll a was used to show continuing growth of plant lines. The results showed that only the tobacco lines transformed with the modified osmotin gene exhibited greater tolerance to salinity. Unlike human serum albumin (HSA), dog serum albumin (DSA) does not possess the characteristics of the specific first binding site for Cu(II). In DSA, the important histidine residue in the third position, responsible for the Cu(II)-binding specificity in HSA, is replaced by a tyrosine residue. In order to study the influence of the tyrosine residue in the third position of DSA, a simple model of the NH2-terminal native sequence tripeptide of DSA, glycylglycyl-L-tyrosine-N-methylamide (GGTNMA) was synthesized and its Cu(II)-binding properties studied by analytical potentiometry, spectrophotometry, CD, and NMR spectroscopy. The species analysis indicated the existence of five mono-complexes at different protonation states: MHA, MA, MH-1A, MH-2A, MH-3A, and only one bis-complex MH-2A-2. The complexing ability of GGTNMA to Cu(II) was found to be weaker than that of the Cu(II) binding peptide models of HSA. The visible absorption spectra of Cu(II)-GGTNMA complexes are similar to those observed in the case of DSA-Cu(II) complexes. The weaker binding and the spectral properties of Cu(II)-GGTNMA complexes are consistent with less specific Cu(II)-binding properties of the peptide of this sequence similar to what was noted with DSA. CD results are in excellent agreement with species analysis and visible spectra where it is clearly evident that Cu(II) binds to GGTNMA starting from the alpha-NH2 group and step by step to deprotonated amide nitrogens as the pH is raised. The absence of any charge transfer band around 400 nm strongly indicates that Cu(II) does not bind to the phenolate group. Furthermore, NMR results are consistent with the noninvolvement of the tyrosine residue of GGTNMA in Cu(II) complexation. Thus, it is clear that the low Cu(II)-binding affinity of DSA is due to the genetic substitution of tyrosine for histidine at the NH2-terminal region of the protein.  相似文献
