28s rDNA group-I introns: a powerful tool for identifying strains of Beauveria brongniartii 总被引:2,自引:0,他引:2
The nuclear ribosomal DNA of the entomopathogenic fungus Beauveria brongniartii is polymorphic in terms of both restriction site and length. Insertions of 350–450 bp long, identified as group-I introns, were detected in the 28 s rDNA. A panel of 47 strains of B. brongniartii , two B. bassiana and one Metarhizium anisopliae of various geographical and biological origins were found to contain 14 variant forms of intron differing in size and restriction pattern, at four different positions. Twelve types of ribosomal large subunit were defined on the basis of variant distribution and compared with strain clustering based on internal transcribed spacers analysis. There was a correlation between the characteristic introns and isolates collected from the sugar cane pest Hoplochelus marginalis . Primers for polymerase chain reaction amplification were chosen from these variants, and used to develop a specific method for detecting strains pathogenic towards Hoplochelus . 相似文献

Much empirical evidence suggests that there is an optimal body size for mammals and that this optimum is in the vicinity of l00g. This presumably reflects an underlying fitness function that is greatest at this mass. Here, I combine such a fitness function with an equilibrium model of competitive character displacement to assess the potential influence of a globally optimal body size in structuring local ecological communities. The model accurately predicts the range of body sizes and the average difference in size for species in communities of varying species richness. The model also predicts a uniform spacing of body sizes, rather than the gaps and clumps in the sizes of coexisting species observed in real communities. Alternative explanations for this phenomenon are discussed. The allometric relationships that result in a body size optimum subsume a large number of characteristics associated with the physiological, behavioral, demographic, and evolutionary dynamics of the species. Further integration of the underlying dynamics (e.g. individual energetics) of these relationships into all hierarchical levels of ecology will have to incorporate multiple interactive sites, spatial heterogeneity, and phylogenetic structure, but it has the potential to provide important discoveries into the means by which natural selection operates. 相似文献

To reveal how the matching models of the left ventricle and its afterload affect the pressure and flow in the aortic root, the differences between the measured pressure and flow waveforms and those determined by three kinds of matching model were compared. The results showed that, compared with the results by both matching models 1 and 2, the pressure and flow waveforms determined by matching model 3 established in this work were in the closest agreement with the corresponding experimental waveforms, therefore indicating that matching model 3 was a matching model that closely and rationally characterized the match between the left ventricle and the systemic artery. 相似文献

Congenital syphilis has been diagnosed very seldom in ancient populations. The case that we examined comes from San Jeronimo's Church (17th and 18th centuries AD; Mexico City). Coffin 43 contained an incomplete skeleton of an approximately 2-year-old infant. The pathological lesions of this skeleton include bilateral osteochondritis, diaphyseal osteomyelitis, and osteitis and/or periostitis on the long bones. The radiographic appearance depicts symmetrical osteomyelitic foci, particularly at the proximal extremity of both tibiae (Wimberger's sign). The skull exhibits hydroceph-aly and periosteal changes on the vault, and the unerupted upper incisors evince dental hypoplasia and other pathological alterations reminiscent of Hutchinson's incisors. 根据多年定位观察资料,包括从组成种的种子萌发、幼苗生长及成年物种的生长规律和物候节律对湿性常绿阔叶林动态和节律的研究,结果表明:林地种子贮量、可萌发种子量和萌发种数,雨季大于旱季。幼苗生长雨季快于旱季。表明雨季是种子萌发、幼苗生长的最佳季节。立木生长是种间竞争、自我调控、自疏的过程。成年物种各物候期长、不明显与温带阔叶林各物候期短、明显不同,而开花、结果、落果终年进行与西双版纳季节雨林物候节律相似。三者的不同在于季节雨林春季(2~3 月)落叶,常绿阔叶林冬季(11~12 月)落叶,温带阔叶林秋季(9~10 月)落叶。依据物种物候节律特点及其对气候环境的反应区分出3 种生态物候型:1.暖温生态物候型(占总种数(50)的82% );2.温性生态物候型(占12% );3.温凉生态物候型(占0.6% ) 相似文献

The dental awareness and needs of a population with Parkinson's disease were assessed by means of postal questionnaires sent to members of Parkinson's disease societies of Belfast and London. A total of 228 valid replies were received representing a response rate of 31%. Results showed that dental problems were reported often, xerostomia and burning sensation being at least twice that of the general population. Satisfaction with dental care was high although it was significantly reduced in those with loose dentures and burning sensation. The results of this study support a greater participation by dentists in the planning and provision of healthcare for sufferer of Parkinson's disease. 相似文献

Ouabain-blocked toad urinary bladders were maintained in Na+-free mucosal solutions, and a depolarizing solution of high K+ activity containing only 5 mM Na+ on the serosal side. Exposure to mucosal sodium (20 mM activity) evoked a transient amiloride-blockable inward current, which decayed to near zero within one hour. The apical sodium conductance increased in the initial phase of the current decay and decreased in the second phase. The conductance decrease required Ca2+ to be present on the serosal side and was more rapid when the mucosal Na+ activity was higher. At 20 mM mucosal Na+ and 3 mM serosal Ca2+ the initial (maximal) rate of inhibition amounted to 20% in 10 min. The conductance decrease could be accelerated by raising the serosal Ca2+ activity to 10 mM. The inhibition reversed on lowering the serosal Ca2+ to 3 μM and, in addition, the mucosal Na+ to zero. Exposure of the mucosal surface to the ionophore nystatin abolished the Ca2+ sensitivity of the transcellular conductance, showing that the Ca2+-sensitive conductance resides in the apical membrane. The data imply that in the K+-depolarized epithelia, cellular Ca2+, taken up from the serosal medium by means of a Na+-Ca2+ antiport, cause feedback inhibition by blockage of apical Na+ channels. However, the rate of inhibition is small, such that this regulatory mechanism will have little effect at 1 mM serosal Ca2+ and less than 20 mM cellular Na+. 相似文献

The effects of forskolin and cholera toxin on the regulation of cAMP release were studied in a neurotensin-secreting rat C-cell line. The interaction of these agents with norepinephrine, a potent neurotensin secretagogue, was also investigated. Forskolin stimulated cAMP release 10(2)-10(3) fold while it increased neurotensin release 2-3 fold. Cholera toxin caused a 10(2)-10(3) fold increase in cAMP release and had no effect on neurotensin release. We conclude that the 44-2 C-cells provide a new model for studying the regulation of the concomitant (via forskolin) or independent (via cholera toxin) secretion of cyclic AMP and/or neurotensin. 相似文献
