The distribution of cysteine desulfhydrase activity in microorganisms was studied with intact cells. The enzyme activity was found mainly in strains belonging to Enterobacteriaceae, especially to genus Aerobacter (Enterobacter). Aerobacter cloacae IFO 12009 showed markedly high activity.

l-Cysteine was essential as an inducer of the enzyme formation, of which 0.2% in the medium is appropriate.

Intact cells of bacteria containing high cysteine desulfhydrase activity, prepared from broth cultured for 19hr, catalyzed the synthesis of l-cysteine from pyruvate, ammonia and hydrogen sulfide.  相似文献

In order to clarify the substrate specificity of the α-L-mannosidase activity of naringinase (Sigma), the following disaccharides and phenol glycosides were freshly prepared: methyl 2-O-(α-L-mannopyranosyl)­β-D-glucoside (1), methyl 3-O-(α-L-mannopyranosyl)-α-D-glucoside (2), methyl 4-O-(α-L-mannopyranosyl)-α-D-glucoside (3), methyl 5-O-(α-L-mannopyranosyl)-β-D-glucoside (4), methyl 6-O-(α-L-mannopyranosyl)-α-D­glucoside (5), 6-O-(α-L-mannpyranosyl)-D-galactose (6), p-nitrophenyl α-L-mannoside (7), and 4-methyl umbelliferone α-L-mannoside (8).These compounds, except for 3 and 5, were hydrolyzed with naringinase.  相似文献

Abstract

This note documents unusual predation of southern royal albatrosses Diomedea epomophora on land by an individual New Zealand sea lion Phocarctos hookeri. During a survey of albatross nests on Campbell Island in the New Zealand sub‐antarctic in January 2005, 128 corpses of adult albatrosses were found at or near nests within a 230 ha area. Flattened vegetation and flipper prints near the nests suggested sea lion predation, and subsequently a male sea lion was found near a line of freshly killed birds. The predation rate was estimated to be at least seven birds per day over a number of weeks; similar skeletal remains from the previous year were observed in a neighbouring area. The impact of one threatened species on another can be a dilemma for conservation managers. In this case, apparently only one male sea lion was involved and, because of the ongoing impact to the albatross population, permission was obtained to cull that individual on 17 January 2005. No further incidents were observed over the following three seasons (2006–08).  相似文献

Mesenchymal stromal cells (MSC) can be expanded from different sources. We compared the influence of inflammation and TLR ligation on the phenotype and function of MSC derived from bone marrow (BM), adipose tissue (AT), and Wharton’s jelly (WJ). WJ-MSC were featured by a lack of TLR4 expression. While inflammation upregulated TLR3 in all three MSC types, TLR4 upregulation was observed only on BM-MSC. TLR ligation increased the production of inflammatory cytokines in BM- and AT-MSC but not in WJ-MSC and augmented anti-inflammatory cytokines in AT-MSC. Although inflammation increased in all MSC types the secretion of inflammatory cytokines, additional TLR triggering did not have further effect on WJ-MSC. The immunosuppressive potential of WJ-MSC on MLR was affected neither by inflammation nor by TLR triggering. This resistance was related to an overproduction of HGF. These data indicate that MSC source could be of importance while designing immunomodulating cell therapy in transplantation.  相似文献

Background aimsMesenchymal stromal cell (MSC) transplantation holds great promise for use in medical therapies. Several key features of MSCs, including efficient cell growth, generation of sufficient cell numbers and safety, as determined by teratoma formation, make MSCs an ideal candidate for clinical use. However, MSCs derived under standard culture conditions, co-cultured with animal by-products, are inappropriate for therapy because of the risks of graft rejection and animal virus transmission to humans. Alternative serum sources have been sought for stem cell production.MethodsWe demonstrate for the first time that human serum from umbilical cord blood (hUCS) is an effective co-culture reagent for MSC production from Wharton's jelly MSCs (WJMSCs). Ten umbilical cords were used to generate parallel cultures of WJMSC lines under medium supplemented with hUCS and embryonic stem cell-qualified fetal bovine serum. The WJMSC lines from each medium were analyzed and compared with regard to cell line derivation, proliferation ability and characteristic stability.ResultsThe phenotypic characteristics of WJMSC derived under either medium showed no differences. WJMSC lines derived under hUCS medium displayed comparable primary culture cell outgrowth, lineage differentiation capacity and cell recovery after cryopreservation compared with supplementation with embryonic stem cell-qualified fetal bovine serum medium. However, superior cell proliferation rates and retention of in vitro propagation (>22 passages) were observed in WJMSC cultures supplemented with hUCS. Additionally, more robust population doubling times were observed in hUCS-supplemented cultures.ConclusionsWe conclude that hUCS is an efficient and effective serum source for animal product–free WJMSC line production and can generate MSC lines that may be appropriate for therapeutic use.  相似文献

ABSTRACT For wildlife managers, determining inter-species differences in the behavioral responses of seabirds to visitation can allow greater efficacy of visitor guidelines. Two key management outcomes for such information include 1) tailoring visitor guidelines to protect the most sensitive species and 2) improving self-regulation during visits by identifying behaviors likely to indicate a change in the natural activity of visited species. On subantarctic Macquarie Island, Australia, I collected the behavioral responses of guarding king (Aptenodytes patagonicus), gentoo (Pygoscelis papua), and royal (Eudyptes schlegeli) penguins before, during, and after exposure to a standardized pedestrian visit, to compare species' behavioral responses to visitation. Gentoo penguins appeared more sensitive than royal or king penguins, exhibiting altered behavior for 5 minutes after the stimulus was removed; this pattern was not evident in kings or royals. Response behaviors useful for visitors to assess their impact on penguins include vigilance (repeated rapid head turning) in all 3 species, agonism in king and royal penguins (reaching and striking at conspecifics), and low threat-display (bill pointing) in gentoo penguins. This study is valuable for wildlife managers as it provides practical information in the application of on-ground visitor guidelines.  相似文献

The comb jelly Mertensia ovum, widely distributed in Arctic regions, has recently been discovered in the northern Baltic Sea. We show that M. ovum also exists in the central Baltic but that the population consists solely of small-sized larvae (less than 1.6 mm). Despite the absence of adults, eggs were abundant. Experiments revealed that the larvae were reproductively active. Egg production and anticipated mortality rates suggest a self-sustaining population. This is the first account of a ctenophore population entirely recruiting through larval reproduction (paedogenesis). We hypothesize that early reproduction is favoured over growth to compensate for high predation pressure.  相似文献

Urban open spaces such as parks can act as urban cool islands. For example, in Seoul, Korea, the many Lee Dynasty royal palaces and tombs act as urban cool islands that reduce the temperature of the park compared with the surrounding urban areas. In order to determine the effect of air temperature differences because of the urban royal tomb park, Seolleung Royal Tomb Park (SPK) and the nearby Seolleung subway station (SLS), air temperatures were recorded at 10 min intervals from November 9th, 2007 to November 8th, 2008. The data were analyzed and examined to illustrate air temperature-reducing effects during the day and over the year. Results of the investigation show that the average air temperature differences due to SPK in the nearby SLS was 1.76°C, and the maximum air temperature-reducing effects due to SPK occur in September, not during the winter. The air temperature-reducing effect due to SPK decreases rapidly from the park boundary, because of the urban morphology and land use.  相似文献

Organisms with external fertilization are often sperm limited, and in echinoids, larger eggs have a higher probability of fertilization than smaller eggs. This difference is thought to be a result of the more frequent sperm-egg collisions experienced by larger targets. Here we report how two components of egg target size, the egg cell and jelly coat, contributed to fertilization success in a selection experiment. We used a cross-sectional analysis of correlated characters to estimate the selection gradients on egg and jelly-coat size in five replicate male pairs of the sand dollar Dendraster excentricus. Results indicated that eggs with larger cells and jelly coats were preferentially fertilized under sperm limitation in the laboratory. The selection gradients were an average of 922% steeper for egg than for jelly-coat size. The standardized selection gradients for egg and jelly-coat size were similar. Our results suggest that fertilization selection can act on both egg-cell and jelly-coat size but that an increase in egg-cell volume is much more likely to increase fertilization success than an equal change in jelly-coat volume. The strengths of the selection gradients were inversely related to the correlation of egg traits across replicate egg clutches. This result suggests the importance of replication in studies of selection of correlated characters.  相似文献
