Ammonium acetate and BaCl2-triethanolamine were used to desorb Mg2+ from the root Donnan free space (DFS) of 23-d-old ryegrass (Lolium multiflorum Lam. cvs. Gulf and Wilo). Amounts of desorbed Mg2+ increased with the increase in Mg2+ activity of the nutrient solution. Slightly less Mg2+ was desorbed by Ba2+ than by NH4 +. Previously published data on short-term net Mg2+ uptake by intact 23-d-old ryegrass plants of the two cultivars were linearly related to the amount of exchangeable Mg+ desorbed from the root DFS (r2=0.90 and 0.81 for the desorption by NH4 + and Ba2+, respectively). A sward of Mg2+ ions attracted to the negative charges of the cell surface is suggested to represent a part of a pool of Mg2+ available for active transport through the plasmalemma.  相似文献

Graminaceous species can enhance iron (Fe) acquisition from sparingly soluble inorganic Fe(III) compounds by release of phytosiderophores (PS) which mobilize Fe(III) by chelation. In most graminaceous species Fe deficiency increases the rate of PS release from roots by a factor of 10–20, but in some species, for example sorghum, this increase is much less. The chemical nature of PS can differ between species and even cultivars.The various PS are similarly effective as the microbial siderophore Desferal (ferrioxamine B methane sulfonate) in mobilizing Fe(III) from a calcareous soil. Under the same conditions the synthetic chelator DTPA (diaethylenetriamine pentaacetic acid) is ineffective.The rate of Fe(III)PS uptake by roots of graminaceous species increases by a factor of about 5 under Fe deficiency. In contrast, uptake of Fe from both synthetic and microbial Fe(III) chelates is much lower and not affected by the Fe nutritional status of the plants. This indicates that in graminaceous species under Fe deficiency a specific uptake system for FePS is activated. In contrast, the specific uptake system for FePS is absent in dicots. In a given graminaceous species the uptake rates of the various FePS are similar, but vary between species by a factor of upto 3. In sorghum, despite the low rate of PS release, the rate of FePS uptake is particularly high.The results indicate that release of PS and subsequent uptake of FePS are under different genetic control. The high susceptibility of sorghum to Fe deficiency (lime-chlorosis) is most probably caused by low rates of PS release in the early seedling stage. Therefore in sorghum, and presumably other graminaceous species also, an increase in resistance to lime chlorosis could be best achieved by breeding for cultivars with high rates of PS release. In corresponding screening procedures attention should be paid to the effects of iron nutritional status and daytime on PS release as well as on rapid microbial degradation of PS.  相似文献

Rhizobia are soil bacteria which symbiotically infect legume roots and generate nodules in which they fix atmospheric nitrogen for the plant in exchange for photosynthetically fixed carbon. A crucial aspect of signal exchange between these symbionts is the secretion of phenolic compounds by the host root which induce nodulation gene expression in the bacteria. Stimulation of nod gene expression by host phenolics is required for nodule formation, is biochemically specific at 10-6 M, and is mediated by nodD. We and others have shown that rhizobia display chemotaxis to 10-9 M of the same phenolic compounds. Chemotaxis to inducer phenolics is selectively reduced or abolished by mutations in certain nod genes governing nodulation efficiency or host specificity. Conversely, mutations in rhizobia that affect general motility or chemotaxis have substantial effects on nodulation efficiency and competitiveness. These findings suggest that microbes entering the rhizosphere environment may utilize minor, non-nutrient components in root exudates as signals to guide their movement towards the root surface and elicit changes in gene expression appropriate to this environment.  相似文献

The effect of aluminium (Al) on root elongation was studied in solution culture and sand culture. Compared to solution culture, in sand culture a ten times higher Al supply was necessary to inhibit root elongation to a comparable degree. This was due to a much lower Al uptake into the 5 mm root tips in sand culture. Fe concentrations in root tips were also lower in sand culture. Ca concentrations were higher and less depressed by Al, whereas Mg and K concentrations were not affected by the culture substrate. Regressions of Al concentrations in root tips versus inhibition of root elongation by Al revealed root damage at lower Al concentrations in sand culture. The effect of culture substrate on Al tolerance was independent of N source and could also be shown in flowing solution culture with and without sand. The results indicate that mechanical impedance in sand culture decreased Al uptake. This may be due to enhanced exudation of organic complexors thus reducing activites of monomeric Al species.  相似文献

Most bacterial traits involved in colonization of plant roots are yet to be defined. Studies were initiated to identify genes in Pseudomonas which play significant roles in this process. The general approach is to use transposons to construct collections of insertion mutants, each of which is then screened for alterations in its interactions with the host plant. In one study a Tn5 derivative containing a constitutively expressed -galactosidase (lacZ) gene was used to generate a collection of insertion mutants which could be distinguished from the wild-type parent on X-gal plates. Each mutant was examined for its ability to colonize wheat seedlings in the presence of the wild-type parent. Mutants which gave wild-type:mutant ratio of 20:1 or greater were obtained. In a second study a Tn5 derivative which carries a promoterless lacZ gene located near one end of the transposon was constructed. Expression of the lacZ gene depends on the presence of an active promoter outside of the transposon in the correct orientation. Insertion mutants generated with this transposon were examined for changes in -galactosidase expression in the presence and absence of plant root exudate. A number of mutants which showed differential lacZ expression have been identified.  相似文献

Douglas-fir, grown on strongly acid soil (pH-H2O 3.87), was fertilized with ammonium, nitrate or ammonium nitrate as N-source. Rhizosphere pH along the root axis was measured with microelectrodes. Pure ammonium supply resulted in acidification of the rhizosphere, almost along the entire root axis; only the extreme root tip was slightly alkaline compared with the bulk soil. With nitrate supply, the alkalization of the rhizosphere at the root tip was considerably stronger than with ammonium supply and the length of the alkalization zone greater, extending over the entire growth zone of the root. Acidification of the rhizosphere along the older parts of the root was less pronounced in the case of nitrate. It is concluded that nitrate nutrition enables the plant to protect its most essential root zone from the adverse effects of strong acidity by locally raising the rhizosphere pH.  相似文献

Nodules of cowpea plants (Vigna unguiculata (L.) Walp. cv. Vita 3 :Bradyrhizobium CB756) cultured for periods of 23 d with their root systems maintained in atmospheres containing a range of partial pressures of O2 (pO2; 1–80%, v/v, in N2) formed and exported ureides (allantoin and allantoic acid) as the major products of fixation at all pO2 tested. In sub-ambient pO2 (1 and 2.5%) nodules contained specific activities of uricase (urate: O2 oxidoreductase; EC 1.7.3.3) and allantoinase (allantoin hydrolyase; EC 3.5.2.5) as much as sevenfold higher than in those from air. On a cell basis, uninfected cells in nodules from 1% O2 contained around five times the level of uricase. Except for NAD: glutamate synthase (EC 1.4.1.14), which was reduced in sub-ambient O2, the activities of other enzymes of ureide synthesis were relatively unaffected by pO2. Short-term effects of pO2 on assimilation of fixed nitrogen were measured in nodules of air-grown plants exposed to subambient pO2 (1, 2.5 or 5%, v/v in N2) and15N2. Despite a fall in total15N2 fixation, ureide synthesis and export was maintained at a high level except in 1% O2 where formation was halved. The data indicate that in addition to the structural and diffusional adaptations of cowpea nodules which allow the balance between O2 supply and demand to be maintained over a wide range of pO2, nodules also show evidence of biochemical adaptations which maintain and enhance normal pathways for the assimilation of fixed nitrogen. Chlorella sp. strain 3.83, a symbiotic Chlorella isolated from the heliozoan Acanthocystis turfacea, excreted between 8% and 16% of assimilated 14CO2 as maltose in the light (15000 lx), with a pH optimum around 4.8. This percentage increased when the illuminance was lowered (36% at 1700 lx). Release of [14C]maltose continued in darkness and could be inhibited by the uncoupler carbonyl cyanide p-trifluoro-methoxyphenylhydrazone and by diethylstilbestrol. Net efflux of maltose was observed even at a concentration ratio of extracellular/intracellular maltose of 7.8. Exogenous [14C]maltose (5 mM) was taken up by the cells with a rate <2% of that of simultaneous maltose release, indicating a practically unidirectional transport. Nodules of cowpea (Vigna unguiculata (L.) Walp. cv. Vita 3:Bradyrhizobium CB 756) from 28-d-old plants cultured for 23 d with their root systems maintained in O2 levels from 1 to 80% (v/v, in N2) in the external gas phase showed a range of structural changes which have been interpreted in relation to an over- or under-supply of O2. A response to the partial pressure of O2 in the gas phase (pO2) was noted with respect to nodule size, lenticel development, the relative distributions of cortical and infected central tissue, the differentiation of cortex, especially the inner cortex, the frequency and size of infected and uninfected interstitial cells, the volume of extracellular spaces both in cortex and infected tissue, and in the frequency of bacteroids. As a consequence of these changes the surface area of inner cortex relative to the nitrogenase-containing units of fixing tissue (infected cells or bacteroids) was increased by as much as 20-fold. Effectiveness of bacteroid functioning increased from 0.10 ± 0.02 · 10-9 μmol acetylene reduced per bacteroid in air-grown nodules to 0.9 ± 0.16 · 10-9 (same units) per bacteroid in those cultured in 1% O2. Abstract. Kosteletzkya virginica (L.) Presl., a dicot halophyte native to brackish tidal marshes, was grown on nutrient solution containing 0. 85, 170 or 255 mol m 3 NaCl, and the effects of external salinity on root growth, ion and water levels, and lipid content were examined in successive harvests. Root growth paralleled shoot growth trends, with some enhancement observed at 85 mol m 3 NaCl and a reduction noted at the higher salinities. Root Na+ content increased with increasing external NaCl, but remained constant with time for each treatment. K+ content, although lower in salt-grown plants after 14 d salinization, subsequently increased to levels comparable to unsalinized plants. A strong K+ affinity was reflected in the increased K+/Na+ selectivity of salt-grown plants and by their low Na+/K+ ratios. Cl levels rose in salinized plants and values were double or more those for Na+, indicating the possibility of a sodium-excluding mechanism in roots. Root phospholipids and sterols, principal membrane constituents, were maintained or elevated and the free sterol/phospholipids ratio increased in salinized K. virginica plants, suggesting retention of overall membrane structure and decreased permeability. This response, considered in light of root calcium maintenance and high potassium levels, suggests that salinity-induced changes in membrane lipid composition may be important in preventing K+ leakage from cells.  相似文献
