以狭果茶藨子果实为原料,多糖提取量为考察指标,通过单因素试验及响应面试验对多糖提取工艺进行优化,结果表明最佳多糖制备工艺条件为:提取温度40℃,时间30 min,料液比1∶30 g/mL,此时狭果茶藨子中多糖提取量达115.32 mg/g;流变学特性研究表明狭果茶藨子果实多糖溶液属于非牛顿流体,多糖溶液表现出剪切稀化的现象,当多糖质量浓度为1.0%时,其流变学特性与0.1%的羧甲基纤维素钠溶液相似,且过酸、过碱的环境均不会改变狭果茶藨子果实多糖溶液的流变学特性;抗氧化试验结果表明狭果茶藨子果实多糖具有潜在的抗氧化能力,对DPPH自由基的清除能力低于抗坏血酸,而对羟自由基的清除能力显著高于抗坏血酸。本研究结果可为狭果茶藨子果实多糖在食品领域的开发利用提供理论依据。  相似文献

Filamentous fungi are important organisms industrially and continue to attract research interest as microbiologists attempt to overcome the problems associated with their behavior in submerged culture. This review critically examines the literature describing these problems and where available suggests possible solutions to them. The influence of the chemical and physical environment on culture morphology, the process engineering challenges presented by different fungal morphologies, and the relationship between fungal morphology and metabolite production are all discussed.  相似文献

Circular dichroism of trimethylsilyl (TMS) derivatives of seven aldonic acid-γ-lactones were studied. As well as original sugar-γ-lactones the sign of the Cotton effect was determined by the C–2 configuration. The magnitude of CD peak was found to be related with the ring geometry. It leads to a conclusion that the observed molecular elipticities may be the sum of two effects; one effect of configuration at C–2 and the other effect of ring geometry. If these two effects are of the same sign, they overlap each other and give a strong CD peak. When the sign of these effects are opposite, they cancel each other and hence give a weak CD peak. Between the two effects, when C–2 substituent is OH or OTMS, the effect of configuration at C–2 is stronger than that of ring geometry.  相似文献

The evolution of stress in coagulating blood is described by aMaxwell-like constitutive model. The evolution is essentiallyaffected by conversion of fibrinogen to fibrin monomer, fibrinpolymerization and its crosslinking. The modifying effects of theprocess result from the active constituents of blood plasma,blood cell concentration and intensity of flow. Interrelationof stress evolution and the kinetics of polymerization species these effects in terms of the order of the kinetics ofpolymerization and degree of fibrin polymerization and values of the constitutive coefficients. Cell physiological processes require the regulation and coordination of both mechanical and dynamical properties of the actin cytoskeleton. Here we review recent advances in understanding the mechanical properties and stability of actin filaments and how these properties are manifested at larger (network) length scales. We discuss how forces can influence local biochemical interactions, resulting in the formation of mechanically sensitive dynamic steady states. Understanding the regulation of such force-activated chemistries and dynamic steady states reflects an important challenge for future work that will provide valuable insights as to how the actin cytoskeleton engenders mechanoresponsiveness of living cells.  相似文献

A theoretical rheological model for agar gels is proposed, based on the bead and spring model for linear flexible random coils and the model for crosslinked polymers. The model introduces the concept of a temperature dependence of the monomeric friction coefficient, ζ0, of the agar molecule. The model has a random coil-like behavior at high temperatures (close to 373 K), and contributions from a three-dimensional network at low temperatures (close to 273 K). A proposed temperature dependence of the net association rate allows the calculation of the fraction of associated molecules as a function of time and temperature. The proposed model predicts the gelation behavior of agar gels utilizing time–temperature data (cooling curves).  相似文献

A theoretical rheological model for agar gels, based on the bead spring model for linear flexible random coils and the model for crosslinked polymers, is successfully fitted to experimental gelation curves obtained over a wide range of cooling rates (0.5–20 °C min−1) and agar concentration (1–3 wt%). The theoretical gelation temperature, Tgelmodel increases with increasing agar concentration and decreasing cooling rate. The intrinsic net association rate increases significantly with increasing cooling rate. This increase is related to the higher probability of association of the non-associated agar molecules at higher cooling rates.  相似文献

Abstract

The β-glucans different from cellulose are the most abundant class of polysaccharides. They are found in microorganisms and higher plants as structural entities of cell wall, as cytoplasmic and vacuolar reserve materials, and as extracellular substances. Enzyme systems capable to hydrolyze β-glucans are produced by different microorganisms. The occurrence and nature of β-glucanases and their substrates are reviewed. The regulation of biosynthesis of these enzymes, their properties, substrate and product specificities, mode of action and molecular cloning are described. The participation of β-glucanases in the morphogenetic events of yeast cell is presented. The role and synergism of different types of 1,3-β-glucanases in microbial cell wall lysis and the potential application for isolation of intracellular materials like proteins, carbohydrates, enzymes and as an analytical tool are discussed in the light of current knowledge.  相似文献

目的:观察肠淋巴液引流对失血性休克大鼠红细胞流变性指标以及血液黏度的作用。方法:Wistar雄性大鼠均分为假休克组、休克组(复制失血性休克模型)、引流组(复制失血性休克模型,自低血压1 h引流休克肠淋巴液)。在低血压3 h或相应时间,经腹主动脉取血,检测红细胞参数、红细胞电泳、红细胞沉降率(ESR)以及血液黏度,计算红细胞聚集指数、红细胞变形指数。结果:与假休克组比较,休克组红细胞数量、红细胞比积(HCT)、血红蛋白(Hb)、平均红细胞血红蛋白浓度(MCHC)、红细胞电泳率与迁移率、红细胞变形指数、全血黏度、全血低切与高切相对黏度和还原黏度显著降低,休克组平均红细胞体积、红细胞电泳时间、ESR、血沉方程K值与校正K值、红细胞聚集性指数、血浆黏度显著升高;引流组MCHC、红细胞电泳率与迁移率、全血黏度、全血低切与高切还原黏度均显著降低,引流组红细胞体积分布宽度(RDW-SD)显著增加。同时,引流组HCT、RDW-SD、红细胞变形指数、全血黏度、全血低切与高切相对黏度显著高于休克组;ESR、血沉方程K值与校正K值、红细胞聚集性指数、血浆黏度显著低于休克组。结论:休克肠淋巴液引流可改善失血性休克大鼠红细胞流变行为,从而改善血液流变性。  相似文献
