Organic anion transporting polypeptides (OATPs, gene symbol SLCO) mediate sodium-independent transport of endogenous compounds such as bile salts, hormones and their conjugates as well as toxins and drugs. OATP1B1 is the major OATP specifically expressed at the basolateral membrane of human hepatocytes and many clinically important drugs have been shown to be substrates of the transporter. According to the computer-based hydropathy analysis, a large intracellular loop 3 (IL3) is situated between transmembrane domain 6 and 7 of OATPs, in which a conserved NPxY motif is found. In the current study, HEK293 cells expressing the HA-tagged OATP1B1 was utilized to investigate the role of the NPxY motif for the function and expression of the transporter. Alanine replacement of N335 or P336 retained substantial uptake function; while simultaneous mutation of these residues resulted in a double mutant that lost almost all the transport activity. On the other hand, Y338A showed >80% reduction for estrone-3-sulfate uptake. Plasma membrane protein analysis revealed that N335/P336A completely lost its cell surface protein expression; while that of Y338A is dramatically reduced. Further investigation with pharmacological inhibitors and immunocytochemistry demonstrated that N335/336A is detained in the Golgi apparatus and Y338A exhibited accelerated protein degradation rate compared to that of the wild-type. Conservative replacement of Y338 with phenylalanine fully recovered uptake and expression of the transporter. In summary, a new role was observed for the NPxY motif located in the IL3 of OATP1B1, which may affect processing and stability of the transporter.  相似文献

A “double‐hydrophobic” elastin‐like triblock polypeptide GPG has been constructed by mimicking the localization of proline‐ and glycine‐rich hydrophobic domains of native elastin, a protein that provides elasticity and resilience to connective tissues. In this study, the effects of trifluoroethanol (TFE), an organic solvent that strongly affects secondary structures of polypeptides on self‐assembly of GPG in aqueous solutions were systematically studied. Beaded nanofiber formation of GPG , where nanoparticles are initially formed by coacervation of the polypeptides followed by their connection into one‐dimensional nanostructures, is accelerated by the addition of TFE at the concentrations up to 30% (v/v), whereas aggregates of nanoparticles are formed at 60% TFE. The concentration‐dependent assembly pattern discussed is based on the influence of TFE on the secondary structures of GPG . Well‐defined nanofibers whose diameter and secondary structures are controlled by TFE concentration may be ideal building blocks for constructing bioelastic materials in tissue engineering. © 2014 Wiley Periodicals, Inc. The stability between helical conformations of homopeptides of alanine, glycine, and aminoisobutyric acid has been studied by means of quantum-mechanical methods. The influence of peptide length on the relative stability between helical conformations has also been analyzed by means of systematic studies for peptides of size up to 11 residues. Finally, the influence of the solvent has been examined by using self-consistent reaction field methods. The results provide a detailed picture of the modulation exerted by these factors on the helical preferences of these peptides. © 1997 Wiley-Liss Inc.  相似文献

应用单向SDS-PAGE和双向IEF-SDS电泳技术对两个品种的普通小麦(T.aestivum L.)T型细胞质雄性不育系及其保持系的线粒体多肽进行了比较研究,结论如下:1.黄化苗期不育系和保持系线粒体多肽在单向SDS-PAGE和双向IEF-SDS电泳行为上无明显差别;2.在孕穗期幼穗线粒体多肽的单向SDS-PAGE图谱上,两个不育系都缺少28Kd多肽带纹,因而不育系和保持系间表现出明显的差异。双向IEF-SDS凝胶电泳证实28Kd带纹实际上是分子量相同而等电点分别为5.58和5.65的两个多肽;3.线粒体基因的表达是具有时空性质的;4.线粒体与T型细胞质雄性不育可能存在着某种特定的关系。 本文还就T型细胞质雄性不育的分子机制进行了探索性讨论。  相似文献

Vacuoles were isolated from primary leaves of barley (Hordeum vulgare L.) by mechanical breakage of protoplasts, and their polypeptide composition analyzed by two-dimensional gel electrophoresis. Vacuoplasts which consist of the vacuole, a portion of the plasmalemma and of the cytoplasma were prepared from protoplasts by ultracentrifugation. By comparing the vacuolar polypeptide pattern with polypeptide patterns of isolated chloroplasts and of vacuoplasts, vacuolar polypeptides could clearly be distinguished from polypeptides derived from cross-contaminating cell compartments. At least 14 polypeptides of apparent molecular mass between 12 and 76 kilodaltons and an isoelectric point between 4.5 and 7.6 could be attributed to the tonoplast fraction of the vacuole, and 35 polypeptides to the soluble fraction of the vacuole. Several lectins with different specificity were employed to characterize the degree and nature of glycosylation of vacuolar polypeptides. Concanavalin A bound to a large number of polypeptides. Electrophoretic analyses of non-reduced and reduced seed storage proteins from 34 cultivars of Capsicum annuum L. were performed on two gel systems. On the basis of clearly expressed difference in electrophoretic profiles of non-reduced proteins, the investigated cultivars were divided into two groups. The two observed phenotypes were genetically determined and related with intermediary subunits of 11S globulins and/or their oligomers.  相似文献

The sensitivity of the plasma membrane H+-ATPase in tobacco was investigated in vitro, both at the proton translocation level and the ATPase level, according to plant development and leaf location. Both activities are stimulated by auxin in all leaves, whatever the plant age and the leaf age. However, the sensitivity to auxin was heterogeneous with respect to plant development and leaf location. In parallel experiments using the same plasma membrane samples, polypepides patterns were investigated by two-dimensional gel electrophoresis and image analysis was used to quantify the relative abundance of 110 peptides. Systematic analysis of the two kinds of data identified 8 polypeptides, the abundance of which changed in a consistent way with the sensitivity, whatever the plant developmental state and leaf location. These unknown polypeptides are proposed as potential markers of the membrane response to auxin.  相似文献

When cells of Synechococcus PCC7942 were subjected to either iron or magnesium limitation, there was an appearance of specific proteins in the outer membrane (isolated as the cell wall fraction). Under iron limitation outer membrane polypeptides of M r 92000, 48000–50000 and 35000 appeared. Specific iron-limited outer membrane proteins (IRMPs) of M r 52000 and 36000 were also induced in iron-limited cultures of Synechocystis PCC6308. The three most abundant nonhistone polypeptides (molecular weights 75,000, 71,000 and 61,000) of the avian erythrocyte nucleus have previously been isolated in the nuclear envelope fraction. They have been separated by sodium dodecylsulfate-polyacrylamide gel electrophoresis and peptide-mapped after limited enzymatic digestion. Three enzymes–chymotrypsin, papain and Staphylococcus aureus protease–were used. Results obtained with each enzyme indicate strong similarities between the three nuclear envelope polypeptides. The amino acid compositions of the two most abundant polypeptides (P75 and P71) have been determined and found to be similar. Further, they readily yield large fragments upon brief alkaline hydrolysis. For both P75 and P71 the degree and the pattern of alkaline fragmentation are almost identical. A 61,000-dalton polypeptide which appears to be P61 is obtained from P75 and P71 by mild acid hydrolysis. These results establish the close chemical similarity of these predominant polypeptides in the erythrocyte nucleus and suggest that they serve related functions.  相似文献
