为揭示柽柳(Tamarix chinensis)光合能力及耗水特征对地下水矿化度的响应规律, 以黄河三角洲建群种——柽柳3年生植株为研究对象, 在1.2 m的潜水水位下, 模拟设置淡水、微咸水、咸水和盐水4种不同的地下水矿化度, 测定柽柳叶片光合-光响应、蒸腾速率和树干液流的日变化。结果表明: 地下水矿化度通过影响土壤盐分可显著影响柽柳光合特性及耗水性能。随地下水矿化度升高, 柽柳叶片净光合速率(Pn)、最大Pn、蒸腾速率、气孔导度、表观量子效率和暗呼吸速率均先升高后降低, 而水分利用效率(WUE)持续降低。淡水、微咸水和盐水处理下, 柽柳Pn光响应平均值分别比咸水处理降低44.1%、15.1%和62.6%; 微咸水、咸水和盐水处理下, 柽柳WUE光响应平均值分别比淡水处理降低25.0%、29.2%和41.7%。随地下水矿化度升高, 柽柳叶片光饱和点先升高后降低, 而光补偿点持续升高, 光照生态幅变窄, 光能利用率变低。淡水和盐水处理下, 柽柳Pn下降分别是非气孔限制和气孔限制引起的。柽柳树干液流速率随地下水矿化度升高而先升高后降低, 咸水处理下树干液流速率日变幅最大, 日液流量最高。淡水、微咸水和盐水处理下日液流速率平均值分别比咸水处理降低61.8%、13.1%和41.9%。咸水矿化度下柽柳有较高的光合特性, 在蒸腾耗水较严重的情况下可实现高效生理用水, 适宜柽柳较好地生长。 相似文献

Due to the pronounced morphological variation and geographical distribution of Galápagos' Opuntia cacti, numerous hypotheses have been advanced regarding their radiation, diversification, and classification. The currently accepted classification is based on morphology and recognizes six species and fourteen varieties, but the plasticity of many of the characteristics renders any morphological taxonomy problematic. Our analysis of previously published morphological data agrees only partially with the current classification. We present the first molecular phylogeny of these plants. Multiple DNA sequences indicate little genetic distinction among the currently identified species, despite restricted gene flow and limited long distance dispersal within the archipelago. No clear relationship exists between morphological and genetic differences. 5-HT1A receptors are involved in the regulation of various behaviors and the mechanism of action of anxiolytics and antidepressants. It is rather difficult to study the expression of the 5-HT1A receptor gene in the brain because of the low concentration of its mRNA. A method developed for quantitating the level of 5-HT1A receptor gene expression in brain structures involves estimation of the copy number for contaminant genomic DNA, the cDNA of the glyceraldehyde 3-phosphate dehydrogenase (GAPDH) gene (a housekeeping gene), and the 5-HT1A receptor gene cDNA in a cDNA preparation. To estimate the GAPDH and 5-HT1A receptor cDNA copy numbers, the fluorescent intensity of the corresponding PCR products is calibrated using genomic DNA standards of known concentrations. The expression of the 5-HT1A receptor gene is corrected for the content of contaminant genomic DNA and presented as a 5-HT1A receptor cDNA copy number per 100 copies of the GAPDH cDNA. The method was used to demonstrate for the first time that expression of the 5-HT1A receptor gene is increased in the frontal cortex and the amygdala of mice knocked-out in the monoamine oxidase A gene. 相似文献

The controversy on how to measure fine root production of forests (P) most accurately continues. We applied four different approaches to determine annual rates of P in an old-growth temperate Fagus sylvatica–Quercus petraea stand: sequential soil coring with minimum–maximum calculation, sequential coring with compartmental flow calculation, the ingrowth core method, and a recently developed root chamber method for measuring the growth of individual fine roots in situ. The results of the four destructive approaches differed by an order of magnitude and, thus, are likely to introduce large errors in estimating P. The highest annual rates of P were obtained from the sequential coring approach with compartmental flow calculation, intermediate rates by sequential coring with minimum–maximum calculation, and low ones by both the root growth chamber and ingrowth core approaches. A carbon budget for the stand was set up based on a model of annual net carbon gain by the canopy and measurements on carbon sink strength (annual leaf, branch and stem growth). The budget implied that a maximum of 27% of the net carbon gain was available for allocation to fine root growth. When compared to the carbon budget data, the sequential coring/compartmental flow approach overestimated annual fine root production substantially; whereas the ingrowth core and root growth chamber approaches grossly underestimated P rates. With an overestimation of about 25% the sequential coring/minimum–maximum approach demonstrated the best agreement with the carbon budget data. It is concluded that the most reliable estimate of P in this temperate forest will be obtained by applying the sequential coring/minimum–maximum approach, conducted with a large number of replicate samples taken on a few dates per season, in conjunction with direct root growth observation by minirhizotrons. 相似文献

Stable oxygen isotope ratios of plant water (sap water) were observed at Spasskaya Pad experimental forest near Yakutsk, Russia in 1997–1999. The 18O of sap water in larch trees (Larix gmelinii) decreased soon after leaf unfolding every year, indicating that snowmelt water was used in the beginning of summer. During mid to late summer, a clear difference in the water source used by plants was observed between wet summers and severe drought summers. The 18O values of water in larch trees were high (–17.8 to –16.1) in August 1999 (wet summer), but low (–20.4 to –19.7) in August 1998 (drought summer). These results indicated that plants used rainwater during a wet summer, but meltwater from permafrost was used by plants during a drought summer. One important role of permafrost is to provide a direct source of water for plants in a severe drought summer; another role is to keep surplus water in the soil until the next summer. If this permafrost system is disturbed by future global warming, unique monotypic stands of deciduous larch trees in east Siberia might be seriously damaged in a severe drought summer. 相似文献

Evidence is presented to show that “Caucasian” genes (B, K, Lua, r, AK2, Pc, and Gm3,5,11) in hybrid North American Indian populations follow a Poisson distribution. A method of determining the maximum amount of admixture, given an observed count of Caucasian genes, is developed. Using allozymes and microsatellites we have analysed the genetic structure among European populations for several Polytrichum species to infer relevant factors, such as historical events or gene flow, that have shaped their genetic structure. As we observed low levels of genetic differentiation among populations, and no decreasing levels of genetic variation with increasing latitude within most of the examined species, no genetic evidence was obtained for a step-wise recolonization of Europe from southern refugia after the latest glacial period for P. commune , P. uliginosum , P. formosum and P. piliferum . The near absence of population substructuring within these species does indicate that extensive spore dispersal is the most important factor determining the genetic structure among European Polytrichum populations. Gene flow levels have apparently been sufficient to prevent genetic differentiation among populations caused by genetic drift, and to wipe out any genetic structure caused by the postglacial recolonization process. On the other hand, increased genetic differentiation of alpine P. formosum populations suggests that mountain ranges might restrict gene flow significantly among Polytrichum populations. In contrast to most examined Polytrichum species, P. juniperinum showed high levels of genetic differentiation and a profound genetic structure. Assuming that gene flow is not more restricted in P. juniperinum , these findings suggest that this species has recolonized Europe after the latest glacial period from two different refugia, one possibly being the British Isles. © 2003 The Linnean Society of London, Biological Journal of the Linnean Society 2003, 78, 203–213. 相似文献
