The effects of low-frequency magnetic fields (MFs) on early pregnancy were studied in CBA/S mice. The magnetic field was a 20 kHz, 15 μT sawtooth. Pregnant females were divided into four groups, two control groups and two exposed groups. One group was exposed to MFs continuously from day 1 postconception (pc) until day 5.5 pc, and the other group was exposed continuously until day 7 pc. All animals were sacrificed on day 19 pc, the day before partus, and their uterine contents were analyzed. No significant increase in the resorption (early fetal death) rate was found in the exposed animals compared to the sham controls. In the group exposed during days 1.0–5.5 pc, the body weight and length of the living fetuses were significantly decreased. Except on day 3 pc (progesterone) and day 13 pc (calcium) in the treated groups, there were no significant differences in progesterone and calcium levels in peripheral blood. Foliar phosphorus (P) and nitrogen (N) concentrations and nutrient resorption in the forest understory shrub Lonicera maackii (Rupr.) Maxim (Caprifoliaceae) were measured along contiguous topographic gradients in two southwestern Ohio forests during 1992–1994. Mean summer foliar P varied significantly among topographic positions (but not sites or years), with uplands having greatest P concentrations and bottomlands exhibiting the lowest. Unlike for P, the mean summer foliar N concentrations varied little among sites, topographic positions, and years. Mean absolute and proportional P resorption ranged from 0.48 mg/dm2 (33.7%) in slope positions to 0.80 mg/dm2 (53.1%) in bottomland positions. Repeated-measures analysis of variance (RMANOVA) for P resorption indicated significant topographic and year effects, a site × year and a site × year × topographic interaction. Mean absolute and proportional foliar N resorption ranged from 6.82 mg/dm2 (30.7%) in bottomlands to 8.41 mg/dm2 (37.3%) in slope positions. RMANOVA indicated a significant topographic effect for both absolute and proportional N resorption and a significant year effect for absolute N resorption. These significant year effects for P and N stemmed from lowest resorption of nutrients in 1993. The results for P resorption support the hypothesis that foliar resorption is greater in forested sites with lower P fertility. However, resorption rates for N did not support the hypothesis clearly, as slopes with intermediate N availability had greater N resorption rates than did N-rich bottomlands.  相似文献

Ebselen is a non-toxic seleno-organic drug with anti-inflammatory and antioxidant properties that is currently being examined in clinical trials to prevent and treat various diseases, including atherosclerosis, stroke, and cancer. However, no reports are available for verifying the pharmacological effects of ebselen on major metabolic bone diseases such as osteoporosis. In this study, we observed that ebselen suppressed the formation of tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells in an osteoblast/osteoclast co-culture by regulating the ratio of receptor activator of nuclear factor kappa-B ligand (RANKL)/osteoprotegerin secreted by osteoblasts. In addition, ebselen treatment in the early stage of osteoclast differentiation inhibited RANKL-dependent osteoclastogenesis by decreasing the phosphorylation of IκB, PI3K, and Akt in early signaling pathways and by subsequently inducing c-Fos and nuclear factor of activated T-cells c1. Further, ebselen induced apoptosis of osteoclasts in the late stage of osteoclast differentiation. In addition, ebselen treatment suppressed filamentous actin ring formation and bone resorption activity of mature osteoclasts. Reflecting these in vitro effects, administration of ebselen recovered bone loss and its µ-CT parameters in lipopolysaccharide-mediated mouse model. Histological analysis confirmed that ebselen prevented trabecular bone matrix degradation and osteoclast formation in the bone tissues. Finally, it was proved that the anti-osteoclastogenic action of ebselen is achieved through targeting N-methyl-D-aspartate (NMDA) receptor. These results indicate that ebselen is a potentially safe drug for treating metabolic bone diseases such as osteoporosis.  相似文献

Two experiments were conducted to determine patterns of N change in tissues of autumn olive (Elaeagnus umbellata Thunb.) and black alder (Alnus glutinosa [L.] Gaertn.) during autumn in central Illinois, U.S.A. In the first study leaf nitrogen concentrations of autumn olive decreased 40% at an infertile minespoil site and 39% at a fertile prairie site throughout autumn whereas nitrogen concentrations in respective bark samples increased by 39% and 37%. Salt-extractable protein concentrations increased in bark and decreased in leaves over the sampling period. Free amino acid concentrations of autumn olive leaves decreased over the course of the experiment from peak concentrations in August. Asparagine, glutamic acid and proline were major constituents of the free amino acid pools in leaves. Total phosphorus concentrations of autumn olive leaves declined by 40–46% during autumn while bark concentrations of P did not significantly change.In the second experiment non-nodulated seedlings of alder receiving a low level of N-fertilization did not exhibit net resorption of leaf N during autumn whereas foliar N concentration of contrasting nonactinorhizal cottonwood plants (Populus deltoides Bartr. ex. Marsh) under the same fertilization regime decreased by 27% after the first frost. A gradual but significant decrease of 38% in foliar N concentration of nodulated alder seedlings grown under a low N-fertilization regime was associated with the cessation of nitrogenase activity during autumn in nodules. Compared with the low N fertilization regime, the higher level of N-fertilization resulted in smaller autumnal decreases of foliar N concentration in nodulated alder (17%) and in cottonwood (20%); but there was no decrease in foliar N concentration in non-nodulated alder. The higher level of N-fertilization promoted a greater accumulation of N in the roots than in the bark of both tree species after the first frost.Our results suggest that black alder lackingFrankia symbionts does not exhibit net leaf N resorption and that autumnal decreases in leaf N ofFrankia-nodulated black alder result primarily from declining foliar N import relative to export due to low temperature inhibition of N2 fixation. In contrast, autumn olive exhibited greater and more precipitous autumnal declines in foliar N concentration than those of alder, and the pattern of N decline was unaffected by site fertility.  相似文献

The number of ovarioles in female Musca vetustissima (potential fecundity/ovarian cycle) is positively correlated with fly size (headwidth) and potential fecundity does not vary between successive ovarian cycles. Field females of average headwidth (2 mm) have a mean potential fecundity of 25 eggs/cycle. Females are anautogenous, needing to ingest protein-rich material in order to mature their oocytes. Females that obtain insufficient protein material for maturation of full egg complements may cease ovarian development before or during early vitellogenesis, or resorb some of their oocytes and mature the remainder. Field females consistently matured in excess of 85% of their egg complements, indicating that female M. vetustissima have ready access to protein-rich material in the field.
Résumé Le nombre d'ovarioles des Musca vetustissima femelles (fécondité potentielle/cycle ovarien) est lié directement à la taille de la mouche (largeur de la tête); la fécondité potentielle ne varie pas entre les cycles ovariens successifs. Les femelles de la nature, d'une largeur de tête moyenne (2 mm) ont une fécondité potentielle moyenne de 25 oeufs par cycle. Les femelles anautogènes, ont besoin d'un repas riche en protéines pour produire des oeufs mûrs. Les femelles qui n'obtiennent pas la quantité suffisante de protéines pour permettre la maturation d'un lot complet d'oeufs peuvent interrompre leur développement avant ou pendant le début de la vitellogenèse, ou résorber certains ovocytes tout en conduisant à maturité le reste. Les femelles de la nature conduisent a maturité 85% de leurs lots d'oeufs, montrant ainsi que les femelles de M. vetustissima y ont un accès facile à des aliments riches en protéines.

Osteoclasts are large multinucleate cells unique in their capacity to resorb bone. These cells are exposed locally to high levels of ionised calcium during the process of resorption. We have therefore examined the effect of elevated extracellular calcium on the morphology and function of freshly disaggregated rat osteoclasts. Cell size and motility were quantitated by time-lapse video recording together with digitisation and computer-centred image analysis. In order to assess the resorptive capacity of isolated osteoclasts, we measured the total area of resorption of devitalised cortical bone by means of scanning electron microscopy and computer-based morphometry. The results show that elevation of the extracellular calcium concentration causes a dramatic reduction of cell size, accompanied by a marked diminution of enzyme release and abolition of bone resorption. We propose that ionised calcium might play an important role in the local regulation of osteoclastic bone resorption.  相似文献

The addition of physiological concentrations of zinc (25-200 (Μg/dL) to Dulbecco’s Modified Eagle’s Medium containing tibiae from 19-d chick embryos resulted in a concentration-dependent increase in tibial content of tartrate-resistant acid phosphatase (TRAP) and an increase in bone resorption, as measured by tibial calcium release. This increase in bone resorption was additive to the resorptive effect resulting from the addition of 10-9-10-7 M parathyroid hormone (PTH), but was not additive to similar effects produced by the addition of 10-9-10-7 M prostaglandin E2 (PGE2). An inhibitor of prostaglandin synthesis, flurbiprofen (10-6 M), did not influence the effect of zinc on bone resorption. However, the addition of 2,6-pyridinedicarboxylic acid (10-3 M, 2,6-PDCA), a chelator of zinc, did attenuate the effects of zinc, as did the addition of an inhibitor of DNA replication (hydroxyurea, 10-3 M). Hydroxyurea also attenuated the bone resorptive response to PGE2, but had no influence on the effects of PTH. These results indicate that physiological concentrations of zinc alter bone resorptive rates in vitro by a mechanism that is dependent on DNA replication.  相似文献

Mineral-containing bone particles (BPs) were implanted intramuscularly into rainbow trout (Oncorhynchus mykiss) to investigate the sequence of appearance of bone-resorbing cells. A fibrous substance first surrounded the implanted BPs and was gradually replaced by connective tissue containing capillaries. Two weeks after BP implantation, relatively small multinucleated cells (type-1 cells), whose cytoplasm stained deeply with hematoxylin, appeared along the surfaces of the BPs. At later stages (after 4–8 weeks), the majority of cells which appeared to be resorbing the BPs were multinucleated cells whose cytoplasm stained deeply with eosin (type-2 cells). Type-2 cells contained more nuclei than type-1 cells. Electron-microscopical observations revealed that type-2 cells had the characteristic features of osteoclasts: the presence of numerous mitochondria, vacuoles and granules, and a differentiation of the cell membrane and cytoplasm into a ruffled border and clear zone, respectively. A tartrate-resistant acid phosphatase activity, which is an established characteristic of osteoclasts in terrestrial vertebrates, but which had not previously been examined in teleosts, was demonstrated histochemically in the type-2 cells. Development of type-2 cells was closely correlated with the development of connective tissue. These findings suggest that the development of a capillary network around the implanted BPs enables circulating osteoclast-progenitors to reach the surface of the BPs.  相似文献

Sedges from genus Eleocharisdominate extensive wetlands in the sugar cane growing areas of the Caribbean. Correlative data suggest that macrophytes in these wetlands are phosphorus limited. To determine effects of increased P input that can be expected, e.g. from agricultural runoff, a common sugar cane fertilizer was applied to representative plots in four marl-based and four peat-based marshes. The plots were located in the proximity of patches of Typha domingensis, which has been reported to be able to outcompete Eleocharis under nutrient rich conditions. Responses to the fertilizer treatment were documented as changes in: Plant height, density, biomass, net primary production, nutrient resorption, decomposition, plant and soil nutrient concentrations, percent cover of cyanobacterial mats, and potential colonization by Typha. Additions of phosphorus significantly increased plant density and height and, consequently, the aboveground net primary production. Phosphorus resorption efficiency following senescence was independent of fertilizer addition in Eleocharis but decreased in Typha from the fertilized plots. Phosphorus resorption proficiency was lower in fertilized plots for both Typha and Eleocharis. Decomposition of litter and cellulose assays was significantly faster in fertilized plots. No spontaneous establishment of Typha occurred in the fertilized plots, but survival of transplanted Typha was higher in fertilized plots than in controls. Increased plant density in fertilized plots led to elimination of a key component of these ecosystems, the nitrogen fixing cyanobacterial mats.  相似文献
