Blood samples are commonly obtained in many experimental contexts to measure targets of interest, including hormones, immune factors, growth factors, proteins, and glucose, yet the composition of the blood is dynamically regulated and easily perturbed. One factor that can change the blood composition is the stress response triggered by the sampling procedure, which can contribute to variability in the measures of interest. Here we describe a procedure for blood sampling from the lateral tail vein in the rat. This procedure offers significant advantages over other more commonly used techniques. It permits rapid sampling with minimal pain or invasiveness, without anesthesia or analgesia. Additionally, it can be used to obtain large volume samples (upwards of 1 ml in some rats), and it may be used repeatedly across experimental days. By minimizing the stress response and pain resulting from blood sampling, measures can more accurately reflect the true basal state of the animal, with minimal influence from the sampling procedure itself.  相似文献

We assessed the effects of prescribed burning and cutting on mouflon (Ovis gmelini musimon × Ovis sp.) spring habitat using an experimental design (17.28 ha) of 2 burned, 2 cut, and 2 untreated plots within a homogeneous stand dominated by heather (Erica cinerea and Calluna vulgaris). Overall, we found a shift in treated plots from ligneous species to herbaceous species with high digestive and energetic values for mouflon. We also found a consistently higher number of mouflon feeding on these treated habitats compared to untreated plots. Such effects were still apparent 4 years after habitat modifications. Our approaches could be used by managers to improve and maintain the range of mouflon populations experiencing habitat loss (e.g., woody plant encroachment) and for which the condition of an animal has often a high economical value through trophy hunting. © 2011 The Wildlife Society.  相似文献

Highly repeated DNA sequences from man, five other primate species and rat were compared using five restriction endonucleases. Calculations of a similarity index based on the mobility of various bands indicate that man, chimpanzee and baboons are very similar. Vegetation management practices have been applied worldwide to enhance habitats for a variety of wildlife species. Big sagebrush (Artemisia tridentata spp.) communities, iconic to western North America, have been treated to restore herbaceous understories through chemical, mechanical, and prescribed burning practices thought to improve habitat conditions for greater sage‐grouse (Centrocercus urophasianus) and other species. Although the response of structural attributes of sagebrush communities to treatments is well understood, there is a need to identify how treatments influence wildlife population dynamics. We investigated the influence of vegetation treatments occurring in Wyoming, United States, from 1994 to 2012 on annual sage‐grouse population change using yearly male sage‐grouse lek counts. We investigated this response across 1, 3, 5, and 10‐year post‐treatment lags to evaluate how the amount of treated sagebrush communities and time since treatment influenced population change, while accounting for climate, wildfire, and anthropogenic factors. With the exception of chemical treatments exhibiting a positive association with sage‐grouse population change 11 years after implementation, population response to treatments was either neutral or negative for at least 11 years following treatments. Our work supports a growing body of research advocating against treating big sagebrush habitats for sage‐grouse, particularly in Wyoming big sagebrush (A. t. wyomingensis). Loss and fragmentation of sagebrush habitats has been identified as a significant threat for remaining sage‐grouse populations. Because sagebrush may take decades to recover following treatments, we recommend practitioners use caution when designing projects to alter remaining habitats, especially when focused on habitat requirements for one life stage and a single species.  相似文献

The biogeochemical and stoichiometric signature of vegetation fire may influence post‐fire ecosystem characteristics and the evolution of plant ‘fire traits’. Phosphorus (P), a potentially limiting nutrient in many fire‐prone environments, might be particularly important in this context; however, the effects of fire on P cycling often vary widely. We conducted a global‐scale meta‐analysis using data from 174 soil studies and 39 litter studies, and found that fire led to significantly higher concentrations of soil mineral P as well as significantly lower soil and litter carbon:P and nitrogen:P ratios. These results demonstrate that fire has a P‐rich signature in the soil–plant system that varies with vegetation type. Further, they suggest that burning can ease P limitation and decouple the biogeochemical cycling of P, carbon and nitrogen. These effects resemble a transient reversion to an earlier stage of ecosystem development, and likely underpin at least some of fire's impacts on ecosystems and organisms.  相似文献

Sleep disruption has been associated with increased risks for several major chronic diseases that develop over decades. Differences in sleep/wake timing between work and free days can result in the development of social jetlag (SJL), a chronic misalignment between a person’s preferred sleep/wake schedule and sleep/wake timing imposed by his/her work schedule. Only a few studies have examined the persistence of SJL or sleep disruption over time. This prospective investigation examined SJL and sleep characteristics over a 2-year period to evaluate whether SJL or poor sleep were chronic conditions during the study period. SJL and sleep measures (total sleep time [TST], sleep onset latency [SOL], wake after sleep onset [WASO]), and sleep efficiency [SE]), were derived from armband monitoring among 390 healthy men and women 21–35 years old. Participants wore the armband for periods of 4–10 days at 6-month intervals during the follow-up period (N = 1431 repeated observations).

The consistency of SJL or sleep disruption over time was analyzed using generalized linear mixed models (GLMMs) for repeated measures. Repeated measures latent class analysis (RMLCA) was then used to identify subgroups among the study participants with different sleep trajectories over time. Individuals in each latent group were compared using GLMMs to identify personal characteristics that differed among the latent groups.

Minor changes in mean SJL, chronotype, or TST were observed over time, whereas no statistically significant changes in SOL, WASO, or SE were observed during the study period. The RMLCA identified two groups of SJL that remained consistent throughout the study (low SJL, mean ± SE: 0.4 ± 0.04 h, 42% of the study population; and high SJL, 1.4 ± 0.03 h, 58%). Those in the SJL group with higher values tended to be employed and have an evening chronotype.

Similarly, two distinct subgroups were observed for SOL, WASO, and SE; one group with a pattern suggesting disrupted sleep over time, and another with a consistently normal sleep pattern. Analyses of TST identified three latent groups with relatively short (5.6 ± 1.0 h, 21%), intermediate (6.5 ± 1.0 h, 44%), and long (7.3 ± 1.0 h, 36%) sleep durations, all with temporally stable, linear trajectories. The results from this study suggest that sleep disturbances among young adults can persist over a 2 year period. Latent groups with poor sleep tended to be male, African American, lower income, and have an evening chronotype relative to those with more normal sleep characteristics. Characterizing the persistence of sleep disruption over time and its contributing factors could be important for understanding the role of poor sleep as a chronic disease risk factor.  相似文献
