Development of the mixed inflorescence in Zea diploperennis Iltis, Doebley & Guzman (Poaceae) Mixed inflorescences of diploperennial teosinte, which terminate the main branches of the plant, arise in the same fashion as tassel spikes. The apical meristem produces bracts in a decussate arrangement. A single axillary bud primordium is initiated in the axil of each bract. Growth of the bract is retarded as the bud enlarges and divides longitudinally into two separate spikelet primordia. The paired spikelets running in two ranks on either side of the inflorescence primordium produce the four-rowed condition typical of teosinte tasselS. In the transition region between male and female portions of the inflorescence, development of the pedicellate spikelet of each spikelet pair is arrested at an early ontogenetic stage. Continued growth of the sessile spikelet and associated rachis flaps destroy the remnants of the arrested spikelet in basal portions of the inflorescence. A similar abortion of the lower floret of the sessile spikelet results in a single pistillate floret per node at anthesis. These results provide further support for the hypothesis that a tassel-like mixed inflorescence of teosinte is ancestral to the maize ear.  相似文献

Pistil hyperplasia in rice spikelets as affected by heat stress   总被引:1,自引:0,他引:1  
Summary Sexual organogenesis in spikelets of rice (Oryza sativa L. cv Kinmaze) affected by heat stress was investigated using SEM and stereo-microscopy. Of the 243 spikelets dissected, 55.6% developed pistil hyperplasia, i.e., proliferated female organs or tissues, including multiple stigmata and/or ovaries, outgrowth of swollen parenchymatous tissues from inside the ovule, and differentiation of trichomes from ovary epidermis. Conversely, 7% of the spikelets exhibited stamen hypoplasia, represented by a decrease in the number of stamens, and only 3.7% of the spikelets showed hyperplasia, represented by an increase in the number of stamens. The morphological and structural development of the anther was disturbed, and microsporogenesis was inhibited. The shape of the anther was altered, though not perfectly, into the form of an ovary; the lemma and palea changed in form and ultimately resembled each other in shape and size. All of these changes in structure are more or less similar to those that usually result in rice spikelets subjected to cold and other environmental stresses. It was therefore concluded that rice plants show similar responses in sex differentiation in the spikelet under various environmental stresses.  相似文献

Recent studies have shown that molecular control of inner floral organ identity appears to be largely conserved between monocots and dicots, but little is known regarding the molecular mechanism underlying development of the monocot outer floral organ, a unique floral structure in grasses. In this study, we report the cloning of the rice EXTRA GLUME1 ( EG1 ) gene, a putative lipase gene that specifies empty-glume fate and floral meristem determinacy. In addition to affecting the identity and number of empty glumes, mutations in EG1 caused ectopic floral organs to be formed at each organ whorl or in extra ectopic whorls. Iterative glume-like structures or new floral organ primordia were formed in the presumptive region of the carpel, resulting in an indeterminate floral meristem. EG1 is expressed strongly in inflorescence primordia and weakly in developing floral primordia. We also found that the floral meristem and organ identity gene OsLHS1 showed altered expression with respect to both pattern and levels in the eg1 mutant, and is probably responsible for the pleiotropic floral defects in eg1 . As a putative class III lipase that functionally differs from any known plant lipase, EG1 reveals a novel pathway that regulates rice empty-glume fate and spikelet development.  相似文献

Background and Aims

High temperatures over 32–36 °C at anthesis induce spikelet sterility in rice. The use of a germplasm with an early-morning flowering (EMF) trait has been hypothesized as a way of avoiding this problem. In this study, the effect of the EMF trait on avoiding high temperature-induced sterility at anthesis by flowering at a cooler temperature in the early morning was evaluated.

Methods

The EMF trait was introgressed from wild rice (Oryza officinalis) into the rice cultivar ‘Koshihikari’ (O. sativa). First, spikelets of the EMF line and Koshihikari were subjected to rising temperatures during the daytime in the greenhouse to test for differences in spikelet sterility. Secondly, spikelets of both plants were exposed to 26, 34 and 38 °C at anthesis and to 38 °C beginning at least 1 h after flowering, in the growth chambers at 70 % relative humidity, to test for differences in tolerance to high temperatures.

Key Results

Spikelets of the EMF line started and completed flowering a few hours earlier than Koshihikari. In a greenhouse experiment, spikelets of Koshihikari opened after the air temperature reached 35 °C, but those of the EMF line could open at cooler temperatures. Under these conditions, spikelet sterility significantly increased in Koshihikari, but did not in the EMF line. The number of sterile spikelets increased as their flowering time was delayed in Koshihikari. Furthermore, the chamber experiments revealed that 60 % of the spikelets from both lines were sterile when exposed to 38 °C at anthesis, indicating that tolerance of high temperature was similar in both genotypes.

Conclusions

Reduced sterility in the EMF line subjected to rising temperatures at anthesis in the greenhouse was attributed to an earlier flowering time compared with Koshihikari. The EMF trait of wild rice is effective in mitigating anticipated yield loss due to global warming by escaping high-temperature stress at anthesis during the daytime.  相似文献

Two wheat cultivars, Hesheng 2 with large grain yield potential, and Shannong 505 with small grain yield potential, were used for investigating the responses of postanthesis photosynthesis, dry matter accumulation and allocation, and grain growth to source/sink changes. At the initial grain filling stage, Hesheng 2 was sensitive to source reduction leading to an increase of net photosynthetic rate (PN) by 10 %; however, little effect of sink reduction was observed. In Shannong 505, PN was obviously decreased by sink reduction, and changed a little after source reduction. At the rapid grain filling stage, Hesheng 2 was sensitive to both source and sink reduction resulting in the increase or decrease of PN, respectively. However, the response of PN in Shannong 505 to source/sink changes was similar to that in previous stage. The dry matter (DM) accumulation after anthesis was affected by source/sink changes. In Hesheng 2, the decrease in DM was higher than that in Shannong 505 after the same source or sink reduction. Source reduction caused a decrease in the allocation of DM to the sheath and stem, and promoted the reserve photosynthates to be reallocated to grain. The effect of sink reduction was contrary. The grain mass of Hesheng 2 was more easily regulated by source/sink changes than that of Shannong 505. The effect source/sink changes on grain mass was in order upper > basal > middle spikelets on spike. As for a spikelet, the effect was found mainly in the grain mass at the positions 3 and 4 from base of the spikelet. A mutant of spikelet differentiation in rice called frizzle panicle (fzp) was discovered in the progeny of a cross between Oryza sativa ssp. indica cv. V20B and cv. Hua1B. The mutant exhibits normal plant morphology but has apparently fewer tillers. The most striking change in fzp is that its spikelet differentiation is completely blocked, with unlimited subsequent rachis branches generated from the positions where spikelets normally develop in wild-type plants. Genetic analysis suggests that fzp is controlled by a single recessive gene, which is temporarily named fzp (t). Based on its mutant phenotype, fzp (t) represents a key gene controlling spikelet differentiation. Some F2 mutant plants derived from various genetic background appeared as the "middle type", suggesting that the action of fzp (t) is influenced by the presence of redundant, modifier or interactive genes. By using simple sequence repeat (SSR) markers and bulked segregant analysis (BSA) method, fzp (t) gene was mapped in the terminal region of the long arm of chromosome 7, with RM172 and RM248 on one side, 3.2 cM and 6.4 cM from fzp (t), and RM18 and RM234 on the other side, 23.1 cM and 26.3 cM from fzp(t), respectively. These results will facilitate the positional cloning and function studies of the gene.  相似文献

用水稻幼穗为外植体进行组织培养,研究了影响其体细胞胚胎发生的有关因素,建立了高频率体细胞胚胎发生的培养程序。结果表明不同基因型之间体细胞胚胎发生率的差异达到61.2%;生长素2,4-D对诱导水稻体细胞胚起重要的调控作用,细胞分裂素BA有一定的协同促进作用。干燥处理可提高愈伤组织体细胞胚胎发生率,愈伤组织含水量在60%~80%范围的培养效果较好;外源DNA溶液浸泡发育早期的心状体细胞胚.其进一步发育时异常胚的频率显著增加.成苗率降低。  相似文献

Vernalization is a decisive physiological process for heading, flowering and graining of biennial plants. Variable duration of low-temperature treatment has effects on lateral morphogenesis, such as spike initiation, floral development and graining rate in winter wheat ( Triticum aestivum L.). The investigation data showed that the duration of vernalization treatment was a decisive factor for the initiation of spike relevant to the time of initiation; the longer the duration at low temperature, the earlier the spike initiation in winter wheat. In the process of the spike differentiation, relatively lower temperature and longer differential time benefited for spike differentiation. Under laboratory condition, a low-temperature treatment for 45 d was optial for flower differentiation and graining in winter wheat. It is novelly recognized that vernalization treatment is essential for development of both spikes and spikelets, besides for promoting initiation of differentiation in winter wheat.  相似文献

We report the first kinetic characterization of human liver cytosolic GTP-dependent phosphoenolpyruvate carboxykinase (GTP-PEPCK), which plays a major role in the development of type 2 diabetes in human. In this work two recombinant forms of the enzyme were studied. One form had a His10-tag and the other was His-tag-free, and with one exception, both exhibited similar kinetic properties. When Mn2+ was used as the sole divalent cation, the His10-tagged enzyme, but not the His-tag-free enzyme, was increasingly inhibited at Mn2+ concentrations greater than 0.7 mM. This inhibition did not pose any problem in kinetic analysis, for within the relevant Mn2+ concentration range the His-tagged human PEPCK behaved almost identically to the tag-free enzyme. This property will bring simplicity and speed to purifying and studying multiple structural variants of this important enzyme. Apparent Km values of tag-free enzyme for phosphoenolpyruvate, GDP and bicarbonate were 450, 79 and 20,600 μM, respectively, while those for oxaloacetate and GTP were 4 and 23 μM, respectively, emphasizing the enzyme's gluconeogenic character. Bicarbonate (> 100 mM) inhibited OAA-forming activity, which was a new observation with a GTP-PEPCK. The apparent Km for Mn2+ in the PEP-forming direction was 30-fold lower than that for the OAA-forming direction. Mn2+ and bicarbonate or CO2 might regulate the enzyme in vivo.  相似文献

以塔拉(Caesalpinia spinosa)种子为原料,研究了塔拉种子多糖的脱蛋白工艺及塔拉多糖的抗氧化性质。以多糖损失率和蛋白脱除率为评价指标,比较Sevage法、三氯乙酸法和木瓜蛋白酶法对塔拉多糖的脱蛋白效果。利用正交优化组合实验设计原理,采用四因素三水平的正交分析法,对木瓜蛋白酶法脱蛋白进行正交优化。结果表明:塔拉多糖最佳脱蛋白工艺条件为酶添加量0.15mL、酶解时间90min、酶解温度60℃、酶解pH=6,蛋白脱除率95.19%,多糖保留率75.02%。通过对塔拉多糖抗氧化性的研究,发现塔拉多糖总抗氧化性较好,对DPPH自由基有较强的清除作用。  相似文献
