Redox and CO photolysis FTIR spectra of yeast cytochrome c oxidase WT and mutants are compared to those from bovine and P. denitrificans CcOs in order to establish common functional features. All display changes that can be assigned to their E242 (bovine numbering) equivalent and to weakly H-bonded water molecules. The additional redox-sensitive band reported at 1736?cm?1 in bovine CcO and previously assigned to D51 is absent from yeast CcO and couldn't be restored by introduction of a D residue at the equivalent position of the yeast protein. Redox spectra of yeast CcO also show much smaller changes in the amide I region, which may relate to structural differences in the region around D51 and the subunit I/II interface.  相似文献

The work reported herein concerns the assembly of N-stearoyl L -cysteine methyl ester [CH3(CH2)16COCysOMe, 1] on the surface of gold. This compound serves as a simple model of a related polypeptide, which has been designed to adopt a β-sheet architecture on metallic and oxide surfaces. We describe the preparation of monolayers of 1, and characterization of these layers via ellipsometry, vibrational spectroscopy and X-ray photoelectron spectroscopy. The results are most consistent with a disordered array of the alkyl chains, in which close packing is frustated by a mismatch in the cross-sectional areas of the cysteinyl ester head group and the stearoyl chains of the thiol. Despite the disorder, the alkyl chains form a hydrophobic surface layer, with an advancing contact angle for water comparable to that observed for octadecanethiol on gold © 1997 John Wiley & Sons, Ltd.  相似文献

We developed an automated quantification workflow for PRM‐enabled detection of D3‐Leu labeled apoA‐I in high‐density lipoprotein (HDL) isolated from humans. Subjects received a bolus injection of D3‐Leu and blood was drawn at eight time points over three days. HDL was isolated and separated into six size fractions for subsequent proteolysis and PRM analysis for the detection of D3‐Leu signal from ~0.03 to 0.6% enrichment. We implemented an intensity‐based quantification approach that takes advantage of high‐resolution/accurate mass PRM scans to identify the D3‐Leu 2HM3 ion from non‐specific peaks. Our workflow includes five modules for extracting the targeted PRM peak intensities (XPIs): Peak centroiding, noise removal, fragment ion matching using Δm/z windows, nine intensity quantification options, and validation and visualization outputs. We optimized the XPI workflow using in vitro synthesized and clinical samples of D0/D3‐Leu labeled apoA‐I. Three subjects’ apoA‐I enrichment curves in six HDL size fractions, and LCAT, apoA‐II and apoE from two size fractions were generated within a few hours. Our PRM strategy and automated quantification workflow will expedite the turnaround of HDL apoA‐I metabolism data in clinical studies that aim to understand and treat the mechanisms behind dyslipidemia.  相似文献

We set out to determine whether near infrared reflectance spectroscopy (NIRS) combined with principal component analysis–linear discriminant analysis (LDA) or, variable selection techniques employing successive projection algorithm or genetic algorithm (GA) could evaluate the bone repair in cranial critical‐size (5 mm) defect after stimulation with collagen sponge scaffold and/or infrared low‐level laser therapy directly on the local. Forty‐five Winstar rats were divided into nine groups of five each, namely: group H – healthy, n = 5 (without treatment and without cranial critical‐size defect), (GI positive control – n = 5, 21 days or n = 5, 30 days) without treatment and with cranial critical‐size defect; (GII‐n = 5, 21 days or n = 5, 30 days) cranial critical‐size defect filled with collagen sponge scaffold; (GIII–n = 5, 21 days or n = 5, 30 days) cranial critical‐size defect submitted to low‐level laser therapy; (GIV–n = 5, 21 days or n = 5, 30 days) cranial critical‐size defect submitted to combined collagen sponge scaffold + low‐level laser therapy treatment. In relation to the histological analysis, the collagen sponge scaffold + low‐level laser therapy treatment group (GIV) 30 days showed the best result with the presence of secondary bone, immature bone (osteoid) and newly formed connective tissue (periosteum). GA–LDA model also successfully classified control class of the others classes. Thus, the results provided by the good‐quality classification model revealed the feasibility of NIRS for application to evaluation of the wound healing in rat cranial defect, thanks to the short analysis time of a few seconds and nondestructive advantages of NIRS as an alternative approach for bone repair purposes. © 2017 American Institute of Chemical Engineers Biotechnol. 峰值骨密度是由遗传和环境因素及其相互作用共同决定的复杂性状。维生素D受体基因、雌激素α受体基因、白介素6基因、副甲状腺素基因、Ⅰ型胶原α2基因、骨钙素基因、α2巯基糖蛋白基因是与骨代谢相关的7个重要的候选基因。本研究旨在检测这7个候选基因之间的相互作用对中国女性峰值骨密度的影响。样本为中国上海的361个无关、健康的绝经前女性,均为汉族人,年龄为20—44岁。采用Hologic QDR2000+双能X射线扫描仪对腰椎与髋部的骨密度进行了检测。采用聚合酶链式反应-限制性片段长度多态分析方法对每个个体的以下8个多态性标记位点进行基因分型:维生素D受体基因的Apa Ⅰ位点,雌激素α受体基因的PvuⅡ和XbaⅠ位点,白介素6基因的BsrB Ⅰ位点,副甲状腺素基因的BstB Ⅰ位点,Ⅰ型胶原α2基因的Msp Ⅰ位点,骨钙素基因的Hind Ⅲ位点,娃巯基糖蛋白基因的SacⅠ位点。采用二元方差分析对基因相互作用与骨密度的关系进行研究。结果表明,白介素基因和雌激素α受体基因(PvuⅡ)的相互作用对髋部(P=0.019)、转子间区(P=0.016)和股骨颈(P=0.019)的骨密度有显著作用。在这3个部位,GGPp基因型携带者比GGpp基因型携带者的骨密度值分别高出18.0%、19.5%和14.8%。另外观察到醒巯基糖蛋白基因与自介素6基因的相互作用对股骨颈骨密度有显著影响(P=0.046)。GGSS基因型携带者的股骨颈骨密度值比GGSs基因型携带者高出18.8%。该项群体水平的统计分析表明:对于中国女性峰值骨密度的遗传决定,白介素基因和雌激素α受体基因、α2巯基糖蛋白基因的相互作用显著。  相似文献

Maize silage nutritive quality is routinely determined by near infrared reflectance spectroscopy (NIRS). However, little is known about the impact of sample preparation on the accuracy of the calibration to predict biological traits. A sample population of 48 maize silages representing a wide range of physiological maturities was used in a study to determine the impact of different sample preparation procedures (i.e., drying regimes; the presence or absence of residual moisture; the degree of particle comminution) on resultant NIR prediction statistics. All silages were scanned using a total of 12 combinations of sample pre-treatments. Each sample preparation combination was subjected to three multivariate regression techniques to give a total of 36 predictions per biological trait. Increased sample preparations procedure, relative to scanning the unprocessed whole plant (WP) material, always resulted in a numerical minimisation of model statistics. However, the ability of each of the treatments to significantly minimise the model statistics differed. Particle comminution was the most important factor, oven-drying regime was intermediate, and residual moisture presence was the least important. Models to predict various biological parameters of maize silage will be improved if material is subjected to a high degree of particle comminution (i.e., having been passed through a 1 mm screen) and developed on plant material previously dried at 60 °C. The extra effort in terms of time and cost required to remove sample residual moisture cannot be justified.  相似文献

Ultraviolet (UV) reflectance has been implicated in mate selection.Yet, in some bird species the plumage of young varies in UVreflectance already in the nest and long before mate choiceand sexual selection come into play. Most birds molt the juvenilebody plumage before reaching sexual maturity, and thus, someconspicuous traits of the juvenile body plumage may rather haveevolved by natural selection, possibly via predation or parentalpreference. This second hypothesis is largely untested and predictsa differential allocation of food between fledging and totalindependence, which is a time period of 2–3 weeks whereoffspring mortality is also highest. Here, we test the predictionthat parents use the individual variation in UV reflectanceamong fledglings for differential food allocation. We manipulatedUV reflectance of the plumage of fledgling great tits Parusmajor by treating chest and cheek feathers with a lotion thateither did or did not contain UV blockers and then recordedfood allocation by parents in an outdoor design simulating postfledgingconditions. The visible spectrum was minimally affected by thistreatment. Females were found to feed UV-reflecting offspringpreferentially, whereas males had no preference. It is the firstevidence showing that the UV reflectance of the feathers ofyoung birds has a signaling function in parent–offspringcommunication and suggests that the UV traits evolved via parentalpreference.  相似文献

通过田间小区试验,测定了3个春小麦品种(高原602、陇春8139和定西24)在不同生育期和不同种植密度的光谱反射率及对应叶面积指数(LAI)和地上生物量,分析了其光谱反射的一般特征和红边参数特征以及光谱变量与LAI和地上生物量的相关性.结果表明:在整个波段内,春小麦冠层光谱表现为高原602>陇春8139>定西24,其叶片光谱表现为定西24>陇春8139>高原602;春小麦冠层光谱在可见光波段和中红外波段成熟期明显大于孕穗期,而叶片光谱在近红外波段孕穗期明显大于成熟期;随着种植密度的提高,在近红外波段冠层和叶片的光谱反射率逐渐增加;冠层光谱的红边均具有"双峰"现象,从孕穗期到成熟期,冠层红边位置呈现"蓝移"现象;LAI和地上生物量与冠层光谱变量之间存在较好的相关性.  相似文献
