Possible effects of changes in net charge on protein hydrogen exchange rates were investigated by desalting hen egg-white lysozyme, which allowed its net charge to increase with decreasing pH in the acid region. Chloride ion-binding ratios, expressed as ratios of free to total Cl?, were measured with a chloride-specific electrode at pH 5 on a 2.4% solution of a five-time-desalted product. This ratio was used to show a 97% reduction of the 11% Cl? present in a commercial lysozyme preparation upon three passes of the enzyme through a column of ion-retardation resin. Net charges on the purified product were assigned from a combination of electrophoretic mobility and proton titration data gathered under minimal ionic strength conditions. The net charge on the desalted product increased by 1.64 units between pH 5.0 and 3.0. Hydrogendeuterium exchange studies on the purified lysozyme in D2O were obtained using the near-infrared region of a Cary 14R spectrophotometer. The rate-pD profile for k2, the rate constant for the intermediate class of exchanging hydrogens, showed a decrease in the apparent pD of minimum exchange rate of 0.3 units, when compared to that obtained earlier in 0.2 m added NaCl. However, the rate of exchange at pD minimum and the number of hydrogens in the class remained largely unaffected. A similar shift was observed for the rate-pD profile of the class 1 hydrogens. Thus, the effect of an increase in net positive charge is to shift the rate-pD profile to a lower pD. Moreover, the effect extended to the interior peptide hydrogens of this globular protein. Consequently, the exchange rates of all the observable hydrogens are altered by the net charge changes, and the effect appeared uniform. The shift can be accounted for quantitatively by applying electrostatic interaction terms to the acid and base catalytic constants characterizing the exchange process. The calculated electrostatic interaction factors in minimal salt and 0.2 m added NaCl were found to be 29 and 18% lower, respectively, than those obtained theoretically. Therefore, under conditions where changes in net charge may occur for a globular protein, the effect on hydrogen exchange rates can be estimated fairly well theoretically, especially at moderate ionic strengths. 相似文献

Apical ectodermal ridges (AERs) isolated from 3- to 4-day chick and quail embryos were prepared by means of trypsinization and microdissection and then were grafted to the dorsal or ventral side of a host chick wing bud. They induced supernumerary limb outgrowths from the host bud showing, respectively, a bidorsal or biventral organization, as determined by the patterns of feather germs. The grafted ridge cells persisted, as revealed by histological sections of supernumerary chick limb parts growing under the influence of quail AERs, whose cells are readily distinguished after application of the Feulgen reagent.These results show that the AER induces limb outgrowth regardless of whether it is associated with dorsal or ventral limb ectoderm and that its continued existence is not dependent on contributions of ectodermal cells from the opposed ectodermal faces of the limb bud. The AER is pictured as maintaining the subjacent mesoderm in a condition of developmental plasticity without specifying its differentiation with respect to the proximodistal axis. Sulfated glycosaminoglycan (GAG) synthesis by primary cultures of embryo, yolk sac, and trophoblast was compared with synthesis by the same tissues in utero. In general, the in vivo and in vitro results were in good agreement. As was the case in vivo, the three tissues synthesized chondroitin-4-sulfate and chondroitin-6-sulfate (but no dematan sulfate) at characteristic ratios.Cultured embryos are already capable of synthesizing chondroitin sulfates, primarily chondroitin-4-sulfate, before, or at, the 64-cell stage. During the attachment and initiation of outgrowth stages, blastocysts synthesize more chondroitin-6-sulfate than chondroitin-4-sulfate. Thereafter, progressively more chondroitin-4-sulfate is synthesized so that the 4:6 ratio increases, resembling that of trophoblast cells.Blastocyst-derived cell lines and teratoma cell cultures were also studied. One blastocyst-derived line, MB4, synthesized GAG with a pattern similar to that of yolk sac, which it resembles biochemically in other respects as well. The GAG profile of MB2, a parietal endoderm-like cell line resembled neither that of embryo, yolk sac, nor trophoblast cells. Recently developed differential staining techniques based on the incorporation of bromodeoxyuridine (BUdR) into DNA permits the unequivocal identification of metaphase cells which have replicated once, twice, and three or more times. This technique has the potential of being utilized in the examination of kinetics of dividing cell populations. This potential is examined in a phytohemagglutinin-stimulated lymphocyte system. Determinations of the effect of increasing concentrations of BUdR on the distribution of metaphase cells between different generation cycles reveals no inhibition of cellular kinetics below 35 μM. The ability to distinguish third generation metaphase cells from subsequent generations is examined through the determination of “labelled” centromeric regions. The ultrastructural features of a purified fraction of Na+,K+-adenosine triphosphatase (ATPase) isolated from dog kidney medulla were compared with those of the initial crude microsomal fraction in the purification sequence. Although both fractions consist of vesicular structures, the purified fraction is more homogeneous with respect to overall size and intramembrane protein particle size and distribution. Polyacrylamide gel electrophoresis profiles of both fractions reveal multiple proteins in the microsomal fraction but only two in the final purified fraction. The membranes of the pure fraction comprised one class of particles roughly 95–120 Å in diameter which represent the in vitro configuration of Na+,K+-ATPase. 相似文献

Mitochondrial ultrastructural transformations have been examined in intact eggs and embryos from three sea urchins, Arbacia punctulata, Strongylocentrotus purpuratus, and Lytechinus pictus. Following fertilization, naturally occurring ultrastructural transformations (designated as condensed to orthodox) were observed to occur in mitochondria of all three families. The ratios of the soluble ADP and ATP pools were examined in eggs of S. purpuratus before and after fertilization in order to test whether the ultrastructural transformations reflect a decrease in relative size of the ADP pool following fertilization. Our data indicate that there is a decrease in the ADP:ATP ratio at fertilization. We report periodic movements during erection of Dictyostelium discoideum (Dd) sorocarps. Our observations lead to the working hypothesis that Dd sorocarp erection occurs by two superimposed processes: one periodic, with a modal period of 6 1/2 min, and one continuous. We tentatively identify the periodic process with cell movement into the apex of the Dd stalk, and the continuous process with cell vacuolation, together with stalk sheath extension. 相似文献

The biomass and production of the most important zooplankton species were followed for two years in three lakes of varying trophic status in the Lake Washington watershed. Cladocerans and copepods were of equal importance in the biomass of lakes Findley and Chester Morse (both oligotrophic), whereas, copepods were the main biomass component in Lake Sammamish (mesotrophic). Cladocerans dominated production in lakes Sammamish and Chester Morse, while in Findley Lake their productive role, like that of biomass, was equal to that of the copepods. Rotifers contributed a relatively small biomass and production.Data from this study supported Hillbricht-Ilkowska's postulate that the energy transfer efficiency between the primary and secondary trophic levels decreases with increasing trophic state. Energy transfer efficiencies for the lakes of this study expressed as a two year mean of the ratio-secondary: primary production, were as follows: Findley Lake-0.13; Chester Morse Lake-0.08; and Lake Sammanish-0.04.On the other hand, the hypothesis of Patalas that the secondary productivity: biomass ratio (P/B) tended to increase in proportion to the productivity of a lake, could not be supported. Lake Sammamish, the most productive of the lakes studied, had a P/B ratio of 0.03 while lakes Findley and Chester Morse had P/B ratios of 0.04. 相似文献

Pigment analyses by TLC were carried out during and after the peak of a Peridinium bloom in Lake Kinneret (Israel). During the experimental period considerable changes occurred in the absolute and relative amounts of the various pigments, especially carotene, diadinoxanthine, peridinin, chlorophyll a and chlorophyl c. The ratio between total Carotenoid to total chlorophyll content is also followed. The influence of suckling stimulation upon postpartum aggression was studied by removing the nipples (thelectomy) of female mice at various times during pregnancy and lactation. Prepartum thelectomy, regardless of whether it was performed prior to mating or shortly before parturition, in combination with the fostering of young, prevented the exhibition of aggression. The aggressive behavior of females thelectomized following either 2 or 5 days of suckling experience was similar to that of normal lactating females. However, only 25% of animals thelectomized following 24 hr of suckling experience exhibited aggressive behavior. The results demonstrate that suckling stimulation is important for the initiation of postpartum aggression but is not essential in animals that have had at least 48 hr of suckling experience. 相似文献
