A comparative study of intermolecular interactions in crystals of two homologous low molecular weight proteins, gamma-II and gamma-IIIb crystallins, from calf eye lens was carried out. Crystal packings for these proteins are very different: intermolecular contact areas compose about 33% of the total accessible surface area of gamma-II as compared with 13% in gamma-III. Two key residues seem to be mainly responsible for the differences in protein association in the crystal medium. These are Ser 103 and Leu 155 in gamma-II, which are replaced by Met 103 and His 155 in gamma-IIb. A similar substitution of these residues is observed in different gene products of gamma-crystallins from a number of vertebrates. This is consistent with the existence of a genetically controlled mechanism for determining intermolecular association of gamma-crystallins in the native medium of the lens. 相似文献

Favourable mutations involving the two dehalogenases (DehI and DehII) of Pseudomonas putida PP3 and derivative strains containing the cloned gene for DehI (dehI) occurred in response to specific environmental conditions, namely: starvation conditions; the presence of dehalogenase substrates (halogenated alkanoic acids — HAAs) which were toxic to P. putida; and/or the presence of a potential growth substrate. Fluctuation tests showed that these mutations were environmentally directed by the presence of HAAs. the mutations were associated with complex DNA rearrangements involving the movement of dehI located on a transposon DEH. Some mutations resulted in switching off the expression of either one or both of the dehalogenases, events which were effective in protecting P. putida from toxic compounds in its growth environment. Other mutations partially restored P. putida's dehalogenating capability under conditions where toxic substrates were absent. Restoration of the capability to untilize HAAs was favoured when normal growth substrates were present in the environment. 相似文献

Mouse strains which develop tumors at a high incidence with characteristics very similar to human cancers have been derived over the last 8 years. The tumors are caused by defined genetic alterations in the mouse genome. Three areas of research have contributed to the derivation of these mouse strains: (1) Molecular analysis of human tumors has shown that distinct oncogenes and tumor suppressor genes are consistently involved in a high percentage of primary tumors. (2) Regulatory enhancer-promoter sequences have been identified which direct gene expression to specific target cells, preferentially mammary epithelial cells. (3) The introduction of recombinant DNA molecules into fertilized mouse eggs by microinjection and integration of the injected DNA into the genome of injected cells has given rise to mutant mouse strains with unique and defined genetic alterations. Studies with different promoter-oncogene combinations introduced into transgenic mouse strains have led to the following general conclusions: (1) Oncogenes expressed in mammary gland cells predispose transgenic mice to mammary tumors. (2) The oncogenic potential of individual oncogenes in mammary epithelial cells differs. (3) Oncogene expression initially often causes a preneoplastic state affecting growth and differentiation parameters of cells. (4) The expression of different oncogenes synergizes to reduce tumor latency. Synergism can also be observed with physiological growth signals like estrogen or growth hormone. The oncogenes with a role in mammary carcinomas which have been investigated in transgenic mice will be described here. The phenotypic consequences of oncogene expression and the implications for the multistep carcinogenesis model will be discussed. 相似文献

Point mutations in the gene of pyruvate oxidase from Lactobacillus plantarum, with proline residue 178 changed to serine, serine 188 to asparagine, and alanine 458 to valine, as well as a combination of the three single point mutations, lead to a significant functional stabilization of the protein. The enzyme is a tetrameric flavoprotein with tightly bound cofactors, FAD, TPP, and divalent metal ions. Thus, stabilization may be achieved either at the level of tertiary or quaternary interactions, or by enhanced cofactor binding. In order to discriminate between these alternatives, unfolding, dissociation, and cofactor binding of the mutant proteins were analyzed. The point mutations do not affect the secondary and tertiary structure, as determined by circular dichroism and protein fluorescence. Similarly, the amino acid substitutions neither modulate the enzymatic properties of the mutant proteins nor do they stabilize the structural stability of the apoenzymes. This holds true for both the local and the global structure with unfolding transitions around 2.5 M and 5 M urea, respectively. On the other hand, deactivation of the holoenzyme (by urea or temperature) is significantly decreased. The most important stabilizing effect is caused by the Ala-Val exchange in the C-terminal domain of the molecule. Its contribution is close to the value observed for the triple mutant, which exhibits maximum stability, with a shift in the thermal transition of ca. 10 degrees C. The effects of the point mutations on FAD binding and subunit association are interconnected. Because FAD binding is linked to oligomerization, the stability of the mutant apoenzyme-FAD complexes is increased. Accordingly, mutants with maximum apparent FAD binding exhibit maximum stability. Analysis of the quaternary structure of the mutant enzymes in the absence and in the presence of coenzymes gives clear evidence that both improved ligand binding and subunit interactions contribute to the observed thermal stabilization. 相似文献

This low level correlated with the lower abundance of cytoplasmic RNA in Minutes compared to control females. M(3)w females demonstrated reduced fecundity during the period of low yolk polypeptide content but gradually increased egg deposition as yolk polypeptide levels rose. Summary A theoretical investigation was made to ascertain the effects of random and non-random deviations, called errors, of phenotypic from genotypic values on population means and on the response to phenotypic recurrent selection. The study was motivated as a selection experiment for disease resistance where there was either variability in the inoculation or environment (the random errors) or where the inoculation was above or below the the optimum rate where genetic differences in resistance are maximized (the non-random errors). The study was limited to the genetics at a diallelic locus (alleles B and b) in an autotetraploid population in random mating equilibrium. The response to selection was measured as the covariance of selection and compared to the exact covariance which was the covariance of selection without errors in phenotype. The random errors were modeled by assuming that a given percentage () of the population was uniformly distributed among the five possible genotype classes independent of their true genotypes. This model was analyzed numerically for a theoretical population with the frequency of the B allele (p) ranging from 0.0 to 1.0 and assumed errors of=0.1 and 0.5 for the following six types of genic action of the B allele: additive, monoplex dominance, partial monoplex dominance, duplex dominance, partial duplex dominance, and recessive. The effect of random error was to consistently reduce the response to selection by a percentage independent of the type of genic action at the locus. The effect on the population mean was an upward bias when p was low and a downward bias when p approached unity. In the non-random error model below optimum inoculations altered the phenotypes by systematically including percentage of susceptible genotypes into one or more other genotype classes with more genetic resistance (a positive shift). With above optimum inoculations, some resistant genotypes are classed with the non-resistant genotypes (a negative shift). The effects on the covariance of selection were found by numerical analysis for the same types of genic action and's as investigated for random error. With a negative shift and a low p, the covariance of selection was always reduced, but for an increasing p the covariance approached and exceeded the exact covariance for all types of genic action except additive. With a positive shift and a low p, response to selection was greatly improved for three types of genic action: duplex dominance, partial duplex dominance, and recessive. The effect of a non-random error on population means was to greatly bias the means upwards for a low p and positive shift, but with increasing p the bias decreased. A relatively slight decrease in the mean occurred with a negative shift. This study indicated check varieties commonly used to monitor selection pressures in screening programs are very responsive to positive non-random shifts, but are relatively unresponsive to negative shifts. The fixation rates of selfing rate modifiers were found by stochastic simulation in an infinite site model, including effects of several deleterious alleles with variable effects, which were randomly distributed in the genome without assuming any pollen discounting. Previous results on the evolution of selfing obtained by more precise methods were in this study further validated, and it was concluded that the effect of genetic associations on the evolution of mating systems is small except in the case of full pollen discounting. Furthermore, attention was given to the uneven distribution of the genetic load in the population, and the accompanying large among-genome variation in fixation rates.
