Intermediate filament (IF) proteins exist in multiple structural forms within cells including mature IF, short filaments or 'squiggles', and non-filamentous precursors called particles. These forms are interconvertible and their relative abundance is IF type, cell type- and cell cycle stage-dependent. These structures are often associated with molecular motors, such as kinesin and dynein, and are therefore capable of translocating through the cytoplasm along microtubules. The assembly of mature IF from their precursor particles is also coupled to translation. These dynamic properties of IF provide mechanisms for regulating their reorganization and assembly in response to the functional requirements of cells. The recent findings that IF and their precursors are frequently associated with signaling molecules have revealed new functions for IF beyond their more traditional roles as mechanical integrators of cells and tissues.  相似文献

Mutations in the DIIS4-S5 linker and DIIS5 have identified hotspots of pyrethroid and DDT interaction with the Drosophila para sodium channel. Wild-type and mutant channels were expressed in Xenopus oocytes and subjected to voltage-clamp analysis. Substitutions L914I, M918T, L925I, T929I and C933A decreased deltamethrin potency, M918T, L925I and T929I decreased permethrin potency and T929I, L925I and I936V decreased fenfluthrin potency. DDT potency was unaffected by M918T, but abolished by T929I and reduced by L925I, L932F and I936V, suggesting that DIIS5 contains at least part of the DDT binding domain. The data support a computer model of pyrethroid and DDT binding.  相似文献

目的:观察2000年3月至2003年4月人工全膝关节置换术(TKR)患者围手术期进行系统康复训练的护理.方法:选择接受TKR的患者24例30个膝关节,入院后进行术前康复教育和肢体的康复训练(包括肌肉、关节训练及步态训练等),术后护理包括有针对性的心理护理及系统规范的康复训练.结果:本组术后随访6-12周,效果满意,优良率达95.8%,未发生并发症.结论:TKP患者围手术期间进行系统康复训练,可以减少膝关节并发症,增强膝关节活动范围,满足日常生活所需的关节活动,提高生活质量.  相似文献

In-gel digestion has been standardised using a poly(propylene) disposable. We designed a four-step rapid and simple in-gel digestion protocol which is carried out in one self-contained reaction tube avoiding keratin contamination. In order to quantify the efficiency of in-gel digestion, we developed a rapid on-column peptide acetylation protocol. Results show that trypsin in-gel uptake is increased and in-gel digestion is 90% complete within 15 min. We further show that spectrum quality, peptide yield and sequence coverage for mass spectrometric analysis are enhanced. We utilise 2-D PAGE separation of photosystem II from barley to demonstrate that the protocol facilitates identification of highly hydrophobic membrane proteins.  相似文献

摘要 目的:探讨听力障碍青少年治疗过程心理干预对患儿语言能力恢复和心境状态的影响。方法:选取我院2019年6月到2020年6月共收治的80例7~18岁听力障碍青少年作为研究对象,将患儿随机分为观察组与对照组,每组40例。对所有患儿应用人工耳蜗植入术进行治疗,给予对照组常规言语训练以及健康教育等干预。观察组患儿在常规干预基础上增加全治疗过程的心理干预。对比两组患儿的治疗效果,语言能力恢复,心境状态,ABC评分。结果:观察组患儿治疗的总有效率为92.50%,对照组患儿治疗的总有效率为75.00%,观察组高于对照组(P<0.05);两组患儿干预前的言语形成、听觉能力以及听觉感知对比无明显差异(P>0.05),两组患儿通过治疗与干预之后,言语形成、听觉能力以及听觉感知明显提升,且观察组评分高于对照组(P<0.05);两组患儿干预前的精神、控制、能力、忍受消极情感以及接受变化对比无明显差异(P>0.05),两组患儿通过治疗与干预之后,精神、控制、能力、忍受消极情感以及接受变化明显降低,且观察组评分低于对照组(P<0.05);两组干预前的兴奋、易激怒、刻板行为以及不恰当语言等评分比较无统计学意义(P>0.05),两组患儿干预后,各评分均下降,其中观察组小于对照组,差异有统计学意义(P<0.05)。结论:对听力障碍青少年在常规治疗与干预基础上增加心理干预,能够提升患儿的治疗效果,提升语言能力恢复情况,减少负面情绪,提升患儿的心境状态,值得临床应用推广。  相似文献

采伐是调整林分结构的重要手段。不同林层的树木对采伐强度有着不同的响应方式。但以往考察采伐对树木生长的影响时多采用定性或简单定量的方法(如按树高等距)划分森林的垂直层次, 这就忽略了同一林层内不同树种间和不同发育阶段树木生长的差异。该研究在吉林蛟河天然阔叶红松(Pinus koraiensis)林内建立轻度(胸高断面积采伐强度17.3%)、中度(34.7%)、重度(51.9%)采伐以及对照(不采伐)样地, 跟踪调查采伐后自然恢复2、4、7年保留木的生长动态。根据不同树种每一个体所处的林层位置和生长发育阶段, 将保留木划分为3个组别: 林冠层树种的成熟个体(I)、林冠层树种的未成熟个体(II)以及林下层树种的全部个体(III), 比较不同恢复时期各组别树木的生长对于采伐强度的响应差异。结果表明, 第II组树木的平均胸径相对生长速率(0.033 cm·cm-1·a-1)显著高于第I (0.016 cm·cm-1·a-1)和III组(0.018 cm·cm-1·a-1)。总体来看, 采伐促进了大多数林冠层优势树种(第I、II组)的生长, 尤其是第II组树木的相对生长速率随采伐强度的增加而增加, 但第I组树木的相对生长速率只在重度采伐样地显著高于对照样地。然而林冠层少见种的生长速率并未受到采伐活动的显著影响。值得注意的是, 第I和II组树木生长对于采伐的响应都存在一定的时间滞后, 伐后短期内(2年)采伐样地与对照样地的生长速率没有显著差异, 而采伐对树木生长的促进效果在伐后2-4年才开始出现, 并在随后的监测期内持续存在。各组别树木的相对生长速率均随初始胸径的增大而降低, 且这种负相关关系的斜率随采伐强度增加逐渐增大, 表明随着采伐强度增加, 较小的树木个体从减弱的竞争中获益更多, 呈现出更加明显的生长释放现象。  相似文献

植物凋落物碳输入显著影响陆地生态系统土壤CO2排放和有机碳(SOC)形成,然而,针对不同质地土壤添加不同化学结构外源碳去向依然不清楚。本研究将13C标记的葡萄糖、淀粉和纤维素添加至红壤和风沙土,比较2种质地土壤添加不同化学结构外源碳在土壤释放的CO2、SOC、可溶性有机碳(DOC)和微生物生物量碳(MBC)库的净累积量、回收率及贡献比例上的差异。结果表明: 添加外源有机碳显著提高了CO2、SOC、DOC和MBC的δ13C值,且随着外源有机碳化学结构复杂性的增加,CO2的δ13C峰值依次延迟出现;外源有机碳种类、土壤类型和培养时间均显著改变外源碳去向及其在各碳库的贡献比例;在风沙土中,外源有机碳更多被矿化为CO2,且CO2库的外源碳净累积量和回收率大小依次为葡萄糖>淀粉>纤维素;红壤添加外源碳转变为SOC的累积量和回收率显著高于风沙土,且红壤SOC库的外源碳净累积量和回收率大小顺序也为葡萄糖>淀粉>纤维素。可见,外源有机碳化学结构和土壤质地共同调控外源碳去向及累积贡献。  相似文献

This study investigated stress softening recovery in intact, separated muscle and mucosa-submucosa esophageal tubes in streptozotocin-induced diabetic rats. Fifteen Wistar rats were made diabetic (DM group) by intraperitoneal injection of 50 mg kg−1 streptozotocin and another 11 rats served as Sham group by injection of saline. All rats survived for 8-weeks. Three series of inflation-deflation loadings at luminal pressure levels of 0.5, 1.0 and 2.0 kPa were carried out on different esophageal tubes. Five distension cycles on each pressure level were done in Ca++-free Krebs solution before and after KCl activation in Ca++-containing Krebs solution. The wall stiffness and stored energy recovery were compared between two groups. The stiffness was biggest in the DM group for the intact tube at pressure 0.5 kPa (P < 0.01) and for the muscle tube at all pressure levels (P < 0.05). Energy recovery induced by stress softening and stiffness loss recovery were significantly smaller in the DM group than in the Sham group for the intact esophagus and separated tubes at all pressure levels (P < 0.05, P < 0.01). In conclusion, the reversible stress softening and passive stiffness recovery were altered in STZ-induced diabetic rats. This study fills a gap in the knowledge about diabetes-induced esophageal remodeling.  相似文献

A new Early Triassic marine fauna is described from an exotic block (olistolith) from the Ad Daffah conglomerate in eastern Oman (Batain), which provides new insights into the ecology and diversity during the early aftermath of the Permian–Triassic Boundary mass extinction. Based on conodont quantitative biochronology, we assign a middle Griesbachian age to the upper part of this boulder. It was derived from an offshore seamount and yielded both nektonic and benthic faunas, including conodonts, ammonoids, gastropods and crinoid ossicles in mass abundance. This demonstrates that despite the stratigraphically near extinction at the Permian–Triassic Boundary, Crinoidea produced enough biomass to form crinoidal limestone as early as middle Griesbachian time. Baudicrinus, previously placed in Dadocrinidae, is now placed in Holocrinidae; therefore, Dadocrinidae are absent in the Early Triassic, and Holocrinidae remains the most basal crown‐group articulates, originating during the middle Griesbachian in the Tethyan Realm. Abundant gastropods assigned to Naticopsis reached a shell size larger than 20 mm and provide another example against any generalized Lilliput effect during the Griesbachian. Whereas the benthic biomass was as high as to allow the resumption of small carbonate factories, the taxonomic diversity of the benthos remained low compared to post‐Early Triassic times. This slow benthic taxonomic recovery is here attributed to low competition within impoverished post‐extinction faunas.  相似文献
