Few species in the genus Grateloupia have been investigated in detail with respect to the development of the auxiliary cell ampullae before or after diploidization. In this study, we document the vegetative and reproductive structures of two new species of Grateloupia, G. taiwanensis S.‐M. Lin et H.‐Y. Liang sp. nov. and G. orientalis S.‐M. Lin et H.‐Y. Liang sp. nov., plus a third species, G. ramosissima Okamura, from Taiwan. Two distinct patterns are reported for the development of the auxiliary cell ampullae: (1) ampullae consisting of three orders of unbranched filaments that branch after diploidization of the auxiliary cell and form a pericarp together with the surrounding secondary medullary filaments (G. taiwanensis type), and (2) ampullae composed of only two orders of unbranched filaments in which only a few cells are incorporated into a basal fusion cell after diploization of the auxiliary cell and the pericarp consists almost entirely of secondary medullary filaments (G. orientalis type). G. orientalis is positioned in a large clade based on rbcL gene sequence analysis that includes the type species of Grateloupia C. Agardh 1822 , Gfilicina. G. taiwanensis clusters with a clade that includes the generitype of Phyllymenia J. Agardh 1848 , Ph. belangeri from South Africa; that of Prionitis J. Agardh 1851 , Prlanceolata from Pacific North America; and that of Pachymeniopsis Y. Yamada ex Kawab. 1954, Palanceolata from Japan. A reexamination of the type species of the genera Grateloupia, Phyllymenia, Prionitis, and Pachymeniopsis is required to clarify the generic and interspecific relationships among the species presently placed in Grateloupia.  相似文献

A functional gene microarray was developed and used to investigate phytoplankton community composition and gene expression in the English Channel. Genes encoding the CO2‐fixation enzyme RUBISCO (rbcL) and the nitrate assimilation enzyme nitrate reductase (NR) representing several major groups of phytoplankton were included as oligonucleotide probes on the “phytoarray.” Five major groups of eukaryotic phytoplankton that possess the Type 1D rbcL gene were detected, both in terms of presence (DNA) and activity (rbcL gene expression). Changes in relative signal intensity among the Type 1D rbcL probes indicated a shift from diatom dominance in the spring bloom to dominance by haptophytes and flagellates later in the summer. Because of the limitations of a smaller database, NR probes detected fewer groups, but due to the greater diversity among known NR sequences, NR probes provided higher phylogenetic resolution than did rbcL probes and identified two uncultivated diatom phylotypes as the most abundant (DNA) and active (NR gene expression) in field samples. Unidentified chlorophytes and the diatom Phaeodactylum tricornutum Bohlin were detected at both the DNA and cDNA (gene expression) levels. The reproducibility of the array was evaluated in several ways, and future directions for further improvement of probe development and sensitivity are outlined. The phytoarray provides a relatively high‐resolution, high‐throughput approach to assessing phytoplankton community composition in marine environments.  相似文献

BACKGROUND AND AIMS: The amount of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39) synthesized in a leaf is closely correlated with N influx into the leaf throughout its lifetime. Rubisco synthesis and N influx are most active in the young leaf during expansion, but are very limited in the senescent leaf. However, it is not established whether Rubisco synthesis can be observed if N influx is increased, even in a very senescent leaf. This study first investigated changes in the relationships between rbcS and rbcL mRNA contents and Rubisco synthesis per unit of leaf mass with leaf senescence. Next, leaves were removed during late senescence, to examine whether Rubisco synthesis is re-stimulated in very senescent leaves by an increase in N influx. METHODS: Different N concentrations (1 and 4 mm) were supplied to Oryza sativa plants at the early (full expansion), middle and late stages (respectively 8 and 16 d after full expansion) of senescence of the eighth leaf. To enhance N influx into the eighth leaf 16 d after full expansion, all leaf blades on the main stem, except for the eighth leaf, and all tillers were removed and plants received 4 mm N (removal treatment). KEY RESULTS: Rubisco synthesis, rbcS and rbcL mRNAs and the translational efficiencies of rbcS and rbcL mRNAs decreased with leaf senescence irrespective of N treatments. However, in the removal treatment at the late stage, they increased more strongly with an increase in N influx than in intact plants. CONCLUSIONS: Although Rubisco synthesis and rbcS and rbcL mRNAs decrease with leaf senescence, leaves at the late stage of senescence have the potential actively to synthesize Rubisco with an increase in N influx.  相似文献

The only known population of Batrachospermum spermatiophorum Vis et Sheath, located in a small stream on east Maui, Hawaii, was sampled from November 2001 to October 2002 to investigate its phenology. Additionally, the taxonomic status of the alga was examined. Phylogenetic analyses of the large subunit of the ribulose‐1,5‐bisphosphate carboxylase/oxygenase (rbcL) gene for B. spermatiophorum confirm its placement within the Section Contorta of the genus Batrachospermum. Comparisons with rbcL gene sequences for all other available Batrachospermum species, including 10 other members of the Section Contorta, illustrate that B. spermatiophorum is distinct in its rbcL nucleotide sequence and should be maintained as a separate species. The phenology of B. spermatiophorum differed from most studies of Batrachospermum reproduction in that the gametophytes were present year round, and no macroscopic chantran‐sia stages were identified at any point during the present study. Maximum plant length was highest in the late fall and early winter months, while gametangial production peaked in February and March. Carposporophyte abundance rose sharply in July and August, and the percentage of carposporophytes producing carpospores was highest in late summer and early fall months. Although significant correlations were determined among stream conditions and among phenological characteristics, only pH was associated with several of the reproductive characters, which is unusual for studies of Batrachospermum phenology. Timing of sexual reproduction for all macroalgal community members in the stream site was not coordinated, with some reproducing year round and others more sporadically. The year‐round presence of Batrachospermum gametophytes may be a result of a combination of smaller seasonal changes in cues such as day length and water temperature in the tropical environment than occur in temperate locations, and lack of extreme conditions that might inhibit the persistence of the gametophyte.  相似文献

Morphological observations and molecular analyses of the north‐western Pacific species of the red algal genus Grateloupia (Halymeniaceae) indicate the presence of an entity, which is somewhat similar in gross morphology to G. asiatica Kawaguchi et Wang but is distinguished from the latter species by some morphological features. These include: (i) a somewhat fleshy texture; (ii) wider and much thicker (4.5–10 mm wide and up to 1300 μm thick) axes, of which an inner cortex consists of more (6–9) cells; (iii) generally longer (up to 17 cm), marginal and surface proliferations that are clearly constricted (terete) at bases; and (iv) much elongated, oblong auxiliary cells. Phylogenetic analysis using the ribulose‐l,5‐bisphosphate carboxylase/ oxygenase (rbcL) gene of G. asiatica and the alga in question shows them to be distantly related and strongly supports the differentiation of these two entities at the species level. Judging from the literature, this entity is actually Grateloupia subpectinata Holmes, which has been placed into synonymy under G. asiatica [as G. filicina (Lamouroux) C. Agardh] or G. prolongata J. Agardh in previous reports, and therefore the Holmes name is reinstated.  相似文献

Abstract An invasive, cold‐tolerant strain of the tropical green alga Caulerpa taxifolia was introduced recently in the Mediterranean Sea and along the Californian coast. We screened 50 aquarium and open‐sea C. taxifolia specimens for the presence/absence of an intron located in the rbcL gene of chloroplast DNA. We also reanalysed a total of 229 sequences of the Internal Transcribed Spacer (ITS) of ribosomal DNA, combining previously published sequences from different studies with 68 new sequences to complement rbcL data. The introduced Mediterranean strain was found to be characterized by the absence of the rbcL intron and by the occurrence of a particular monomorphic ITS type. A PCR assay based on rbcL gene was developed to detect new introductions of the invasive strain of C. taxifolia. This rapid and inexpensive test could be useful to assist environment managers in the preservation of coastal marine ecosystems.  相似文献

rbcL (1310 bp) and matK (1014 bp), using 15 species representing the family. The study included analyses of Ticodendron (Ticodendraceae) and three species of Betulaceae as close relatives, and one species each of Juglandaceae and Myricaceae as outgroups. Analyses based on matK gene sequences, which provided a much better resolution than the analyses based on rbcL gene sequences alone, resulted in a single most parsimonious tree whose topology is almost identical with the strict consensus tree generated by the combined data set of rbcL and matK gene sequences. Results showed that Casuarinaceae are monophyletic, comprising four distinct genera, Allocasuarina, Casuarina, Ceuthostoma and Gymnostoma, which were not recognized until recently. Within the family, Gymnostoma is positioned at the most basal position and sister to the remainder. Within the remainder Ceuthostoma is sister to the Allocasuarina-Casuarina clade. Morphologically the basalmost position of Gymnostoma is supported by plesiomorphies such as exposed stomata in the shallow longitudinal furrows of the branchlets, a basic chromosome number x=8 and the gynoecium composed of two fertile, biovulate carpels. The three other genera, Allocasuarina, Casuarina, and Ceuthostoma, have invisible stomata in the deep longitudinal furrows of the branchlets, a higher basic chromosome number x=9 or 10–14 (unknown in Ceuthostoma), the gynoecium composed of one fertile and one sterile carpel with a single ovule (unknown in Ceuthostoma). The diversity of infructescence morphology found in the latter three genera suggests that they may have evolved in close association with the elaboration of fruit dispersal mechanisms.  DNA sequences of both 5′ and 3′ regions of the plastid ndhF gene were generated in order to study the position of Patrinia and Nardostachys, to check the potential paraphyletic nature of Patrinieae, and to evaluate the possible link between the tribe and Linnaeaceae. Parsimony analysis showed very strong support for Patrinia as sister to all members of Valerianaceae (including Nardostachys) and indicated the paraphyletic nature of the tribe Patrinieae. Additionally, trees were constructed from available rbcL data separately and supplemented with ndhF sequences. Topologies of these combined cladograms are in agreement with the ndhF phylogeny, suggesting that the traditionally circumscribed Patrinieae can no longer be recognized but must be considered as part of a basal grade in Valerianaceae. Parsimony analysis based on a morphological data set supported a monophyletic Patrinieae; combination with the molecular data showed a paraphyletic Patrinieae. The seven currently recognized species of Geranium endemic to the Hawaiian Islands are unusual in their shrubby or arborescent habit and unlobed, parallel-veined leaves rather than the palmately cleft or lobed leaves and herbaceous habit typical of the genus. Their placement within the genus and their biogeographic source have been obscured by this morphological distictiveness and the limited resolution of relationships on the basis of morphology in the very speciose subgenus Geranium. Phylogenetic analysis of rbcL gene sequences provides strong support for the monophyly of the Hawaiian group, and indicates that the Hawaiian clade is deeply nested within section Geranium rather than comprising a separate section. The continental relatives studied to date with the greatest similarity in sequence to the Hawaiian group are native to the Americas rather than Asia or the Pacific. The Hawaiian species are extremely similar to one another in rbcL sequence, while the tree topology obtained is consistent with a basal position for Geranium arboreum within the group.  相似文献
