Summary The structure of the T-DNA in Ri-transformed plants of Brassica napus, Nicotiana plumbaginifolia and Nicotiana tabacum was analysed. All the plants studied present a particular phenotype with wrinkled leaves. The T-DNA is composed of two parts: TL and TR. The size of the TL-DNA (19–20 kb) seems to be almost constant, except in N. tabacum where it is shorter. The TR-DNA can be absent, and its size varies from about 5–28 kb, with two predominant lengths. The smaller size does not include the region homologous to the tms genes of the pTi T-DNA. The copy number varies from one to four copies per plant genome. TL and TR-DNA are not always present in the same copy number, but in some cases are linked together. 相似文献

Understanding natural selection's effect on genetic variation is a major goal in biology, but the genome‐scale consequences of contemporary selection are not well known. In a release and recapture field experiment we transplanted stick insects to native and novel host plants and directly measured allele frequency changes within a generation at 186 576 genetic loci. We observed substantial, genome‐wide allele frequency changes during the experiment, most of which could be attributed to random mortality (genetic drift). However, we also documented that selection affected multiple genetic loci distributed across the genome, particularly in transplants to the novel host. Host‐associated selection affecting the genome acted on both a known colour‐pattern trait as well as other (unmeasured) phenotypes. We also found evidence that selection associated with elevation affected genome variation, although our experiment was not designed to test this. Our results illustrate how genomic data can identify previously underappreciated ecological sources and phenotypic targets of selection. 相似文献

2003和2004年于北京三海(西海、后海和前海)分别采集21、22个样品,利用蚕豆根尖微核技术对水体的污染状况进行了遗传毒性分析。结果表明,水体总体显示出不同程度的遗传毒性特征。2003、2004两年中,微核相对率〈1.5的样本分别占4.7619%、18.1818%,主要分布于前海和后海湖心区域;微核相对率为1.5~2.0的样本分别占28.5714%和13.6364%;微核相对率为2.0~3.5的样本分别占28.5714%和45.4545%;微核相对率〉3.5的样本分别占38.0952%和22.7273%。其中。在民居密集的西海区域,两年重度污染率均达到40%。结果表明,水体污染程度与人为因素具有显著相关。本文对试验方法亦进行了讨论。 相似文献

Invertebrate biomonitoring can reveal crucial information about the status of restoration projects; however, it is routinely underused because of the high level of taxonomic expertise and resources required. Invertebrate DNA metabarcoding has been used to characterize invertebrate biodiversity but its application in restoration remains untested. We use DNA metabarcoding, a new approach for restoration assessment, to explore the invertebrate composition from pitfall traps at two mine site restoration chronosequences in southwestern Australia. Invertebrates were profiled using two cytochrome oxidase subunit 1 assays to investigate invertebrate biodiversity. The data revealed differences between invertebrate communities at the two mines and between the different age plots of the chronosequences. Several characteristic taxa were identified for each age within the chronosequence, including springtails within the youngest sites (Order: Collembola) and millipedes within the oldest and reference sites (Order: Julida). This study facilitates development of a molecular “toolkit” for the monitoring of ecological restoration projects. We suggest that a metabarcoding approach shows promise in complementing current monitoring practices that rely on alpha taxonomy. 相似文献

采用微量稀释法测定了32种药物对临床分离猪源链球菌的体外最小抑菌浓度(MIC),以美国临床检验标准委员会(NCCL5)的临界浓度做为判断标准,判定了猪源链球菌对32种药物的耐药性。结果表明,临床分离的菌株以耐药菌为主,且96.6%的菌株呈多重耐药,41%菌株为链霉素高耐药菌株(MIC≥2mg/mL);对磺胺类药物(92.3%~98.3%)、氨基糖甙类药物(70.8%~78.5%)、四环素类(72.3%)、林可胺类(66.2%~64.6%)、大环内酯类(53.8%~67.7%)耐药性最为严重,对青霉素类(18.5%~53.8%)、头孢菌素类(18.5%~56.9%)、泰妙灵(21.5%)和喹诺酮类药物(36.9%~78.5%)耐药性次之,而所选菌株对氯霉素类药物氟苯尼考均敏感;检测了所有耐8内酰胺类抗生素菌株是否产β内酰胺酶,结果均为阴性。 相似文献

Effects of potassium on the photosynthetic recovery of Nostoc flagelliforme (Berk. & Curtis) Bornet & Flahault were investigated to determine its exact role during rehydration. Potassium enhanced recovery of the ability to reduce the primary quinone‐type acceptor (QA) and plastoquinone (PQ) pool and the area over the fluorescence rise curve was increased by 127%. The proportions of closed PSII reaction centers at phases J and I and the net rate of closure of PSII reaction centers were decreased by, respectively, 19%, 8%, and 23% with the addition of potassium, due to changes in the ability of PSII for multiple turnovers needed to reduce the PQ pool. Potassium significantly enhanced the probability of electron transfer beyond QA and the recovery of electron transport flux per PSII reaction center. Electron transport from water to methyl viologen for samples rehydrated in K+‐free BG11 medium was 54% of those with the addition of potassium. However, electron flow from water to p‐benzoquinone and from reduced 2,6‐dichlorophenol‐indophenol to methyl viologen showed little change with the addition of potassium. The fast phase and slow phase of millisecond delayed light emission and the ATP content for samples rehydrated in K+‐free BG11 medium were, respectively, 71.6%, 50.7%, and 77.1% of those with the addition of potassium. These suggested that potassium affected electron transfer from PQ to plastocyanin through the cytochrome b6f complex and the proton motive force across the thylakoid membranes, probably reflecting its role in charge balance during H+ transport by the cytochrome b6f complex. 相似文献
