1. Logging can strongly affect stream macroinvertebrate communities, but the direction and magnitude of these effects and their implications for trout abundance are frequently region‐specific and difficult to predict. 2. In first‐order streams in northern New England (U.S.A.) representing a chronosequence of logging history (<2 to >80 years since logging), we measured riparian forest conditions, stream macroinvertebrate community characteristics and brook trout (Salvelinus fontinalis) abundance. Principal component analysis was used to collapse forest data into two independent variables representing variation in logging history, riparian forest structure and canopy cover. We used these data to test whether logging history and associated forest conditions were significant predictors of macroinvertebrate abundance and functional feeding group composition, and whether brook trout abundance was related to logging‐associated variation in invertebrate communities. 3. Catchments with high PC1 scores (recently logged, high‐density stands with low mean tree diameter) and low PC2 scores (low canopy cover) had significantly higher total macroinvertebrate abundance, particularly with respect to chironomid larvae (low PC2 scores) and invertebrates in the grazer functional feeding group (high PC1 scores). In contrast, proportional representation of macroinvertebrates in the shredder functional feeding group increased with time since logging and canopy cover (high PC2 scores). Brook trout density and biomass was significantly greater in young, recently logged stands (high PC1 scores) and was positively related to overall macroinvertebrate abundance. In addition, three variables – trout density, invertebrate abundance and shredder abundance – successfully discriminated between streams that were less‐impacted versus more‐impacted by forestry. 4. These results indicate that timber harvest in northern New England headwater streams may shift shredder‐dominated macroinvertebrate communities supporting low trout abundance to a grazer/chironomid‐dominated macroinvertebrate community supporting higher trout abundance. However, while local effects on brook trout abundance may be positive, these benefits may be outweighed by negative effects of brook trout on co‐occurring species, as well as impairment of habitat quality downstream. Research testing the generality of these patterns will improve understanding of how aquatic ecosystems respond to anthropogenic and natural trajectories of forest change. 相似文献

Body composition analysis confirmed that egg development and reproductive senescence were delayed in silver female kokanee Oncorhynchus nerka (a small proportion of female kokanee exhibit extreme prespawning waiting and are called 'silvers' because of their colouration) relative to that of red female kokanee (most female kokanee exhibit this 'normal' phenology and are called 'reds'). Silver females were also smaller and younger than red females. A simple life-history model with two size thresholds is proposed to account for the observed body size and age differences. Demand-feeding behaviour was studied in Oncorhynchus mykiss held at 7–10°C in groups of 30 individuals, under a 16:8, 12:12 or 8 : 16 light : dark regime, using self-feeders connected to a computer. The daily food demand was variable between tanks from 1.5 to 5% of the body weight, the specific growth rate ranged between 1.4 and 2.6% of body weight increase per day, and the feed: gain ratio ranged between 0.84 and 3.17, regardless of the photoperiod length. More than 98% of the feeding demand occurred during the photophase, regardless of the photoperiod, with a main peak at dawn and an occasional peak at dusk. 相似文献

Environmental influences on cardiovascular variables in rainbow trout, Oncorhynchus mykiss (Walbaum)
Two groups of wild (lakedwelling and anadromous), and a group of hatchery-reared Oncorhynchus mykiss (Walbaum) (rainbow trout) were sampled in order to measure cardiac morphometrics, haemoglobin concentration, and the DNA and protein concentration in cardiac muscle. A combination of these variables was used to distinguish wild fish from domestic ones.
The wild fish had significantly higher levels of haemoglobin (for male fish, 10.10 and 10.07 g 100 ml−1 vs. 7.69 g d−1 ) and larger relative ventricle mass (females, 0.091 and 0.099% ofbody mass vs. 0.073%; males, 0.108 and 0.134% vs. 0.102%; immatures, 0.086 and 0.094% vs. 0, 072%, respectively) than the domestic fish. The anadromous and domestic fish had significantly higher amounts of compact tissue when compared with lake fish (females, 43 and 47% of ventricle mass vs. 34%, respectively). Ventricle size distinguished wild fish from domestic fish, except male anadromous and male domestic fish which were distinguished only by haemoglobin and compact tissue values. Immature fishes from all groups had lower total protein levels in the ventricle, lower compact tissue levels, and less haemoglobin. Points regarding the potential environmental influences in determining these cardiovascular trends are discussed. 相似文献

The effect of an induced salmonid parr-to-smolt metamorphosis (smoltification) on the optical quality of the ocular lens was studied. In two separate experiments, rainbow trout (Oncorhynchus mykiss) parr were fed thyroxine in their diet to induce the metamorphosis. Lenses were excised at regular samplings during the treatment period and optically scanned using a custom scanning laser monitor. Radioimmunoassay was used to measure serum titers of thyroxine and 3,5,3-triiodo-L-thyronine. It was found that lens optical quality was consistently negatively correlated with 3,5,3-triiodo-L-thyronine levels, but not with thyroxine levels. To test if thyroid hormones are directly responsible for the change in optical quality, rainbow trout lenses were cultured for 72 h in a medium containing 3,5,3-triiodo-L-thyronine, but no effect was observed. The significance of these findings in the contexts of the fishes visual capabilities and smolting physiology is discussed. 相似文献

Human activities play a major role in toxic and carcinogenic metal pollution of the environment. This study was undertaken
to evaluate the effects of copper and mercury at the 400-to 1000-μM concentration range on some biochemical markers of oxidative
stress, such as lipid peroxidation (LPO), glutathione-S-transferase (GST) activity, and reduced glutathione (GSH) content
in the rainbow trout gill homogenates with or without supplementation of manganese, selenium, and bovine serum albumin (BSA).
The integrity of DNA was also measured to assess metal ion toxicity. The results showed that the LPO and specific activity
of GST were elevated. This indicated that cell-protecting antioxidant mechanisms were overtaxed and could not prevent membrane
peroxidation. Following the addition of metals, the GSH content was also significantly reduced in a concentration-dependent
manner. Mercury was found to be more effective than copper. The application of antioxidants proved beneficial in inhibiting
LPO, reducing GST activity, and elevating the GSH levels in the gill samples. Manganese was more effective than selenium and
BSA. Surprisingly, when BSA (1.0%) was added to the gill homogenates treated with a 1000-μM concentration of metal ions, instead
of alleviating malondialdehyde (MDA) generation, a drastic elevation in the MDA levels, alleviation in GST activity, and a
further decrease in glutathione (GSH) levels were observed, which were most likely the result of pro-oxidant activity of BSA.
The results also indicated that mercury and copper functioned as genotoxic pollutants, which altered the DNA integrity by
inducing the single and double-stranded DNA breaks in the gill cell nuclei. Collectively, toxicity of metal ions is related
to the depletion of GSH content and inhibition of antioxidant enzyme GST, resulting in the propagation of LPO and DNA damage. 相似文献

In humans, leukocyte cell-derived chemotaxin 2 (LECT2) is a 16kDa chemotactic protein that consists of 133 amino acids and three intramolecular disulphide bonds. Although it was originally demonstrated to have a chemotactic function in vitro, recent data sustain a further multifunctional role of LECT2 that extends from cell growth, differentiation, damage/repair process and carcinogenesis to autoimmune diseases. The in vivo function of LECT2 protein still remains obscure. In order to study the phylogeny of LECT2, a full-length cDNA clone of LECT2 gene, 720 bp in size, was isolated in rainbow trout (Oncorhynchus mykiss). Its deduced amino acid sequence of 156 residues, presents 40, 45 and 61% overall identity to human, mouse and carp LECT2 proteins, respectively. In contrast to mammalian LECT2 protein, trout LECT2 protein reveals two potential N-glycosylation sites. Phylogenetic analysis shows that trout LECT2 is clustered with the known homologous proteins. Single nucleotide polymorphisms (SNPs) are a class of genetic markers that are well suited to a broad range of research and management applications. Although advances in genotyping chemistries and analysis methods continue to increase the potential advantages of using SNPs to address molecular ecological questions, the scarcity of available DNA sequence data for most species has limited marker development. As the number and diversity of species being targeted for large-scale sequencing has increased, so has the potential for using sequence from sister taxa for marker development in species of interest. We evaluated the use of Oncorhynchus mykiss and Salmo salar sequence data to identify SNPs in three other species (Oncorhynchus tshawytscha, Oncorhynchus nerka and Oncorhynchus keta). Primers designed based on O. mykiss and S. salar alignments were more successful than primers designed based on Oncorhynchus-only alignments for sequencing target species, presumably due to the much larger number of potential targets available from the former alignments and possibly greater sequence conservation in those targets. In sequencing approximately 89 kb we observed a frequency of 4.30 x 10(-3) SNPs per base pair. Approximately half (53/101) of the subsequently designed validation assays resulted in high-throughput SNP genotyping markers. We speculate that this relatively low conversion rate may reflect the duplicated nature of the salmon genome. Our results suggest that a large number of SNPs could be developed for Pacific salmon using sequence data from other species. While the costs of DNA sequencing are still significant, these must be compared to the costs of using other marker classes for a given application. 相似文献
