Near-ultraviolet (NUV) radiation killing of Escherichia coli K-12 can be enhanced by a sub-lethal concentration of hydrogen peroxide. This can be divided into a “RecA-dependent” and “RecA-independent” synergistic killing action. Stationary phase wild-type and 8 closely related repair-deficient mutants were examined for their NUV sensitivities in the presence and absence of H2O2. All exhibited the “RecA-independent” synergism; i.e., H2O2 enhanced NUV lethality when RecA repair was not operating. The “RecA-independent” synergism did not result from destruction of repair enzymes. Very few DNA—protein crosslinks could be detected following NUV plus H2O2 treatment. However, double-strand (DS) DNA breaks were produced, apparently by conversion of closely spaced single-strand (SS) breaks on opposite strands. The correlation between DS-break formation and lethality in wild-type and a polA mutant indicates that the RecA-independent synergistic killing results from the conversion of SS into lethal DS breaks.  相似文献

Summary Plant communities in the Mediterranean region have been analyzed with the emphasis on community processes regulating diversity trends. We want to know how several mechanisms that contribute to the maintenance or to the modification of species diversity, act: (i) in different communities, during an ecological succession (from old fields to forests), and (ii) in forest communities throughout the year. Some of these mechanisms were investigated through difference in stratification, life-form, phenology, dispersal agent, and microenvironmental gradient.The paper censists of two parts. The first one deals with the relationship between structural and biological diversity in conjunction with succession stages leading to a Quereus pubescens forest. It is concluded that diversity, however it is measured, fluctuates during the succession: it increases initially (15 years after abandonment) and also in later stages when trees appear in the fields; it decreases after 20 years when Brachypodium phoenicoides dominates and in the latest stages of the succession studied. These trends may be interpreted in relation to the evolution of life form and dispersal agents spectra during the succession.The second part concerns community-structure and biological diversity in relation to the organization of two Quercus pubescens forests. It appears that the species richness in the understory of the forests, may be explained by: (i) the tendency of the modal height of species foliage distribution to oceupy different positions, (ii) the quasicomplete asynchronism in the major phenophases of the dominant species, (iii) different horizontal patterns of species distribution along a light gradient.These schemes are discussed in relation to ecological hypotheses on diversity in relation to succession, and to community organization.Nomenelature follows: P. Effects of repeated carbon-dioxide anaesthesia during larval development on the number of moults, larval duration, and the body size of resulting adults were investigated. Concentration of carbon dioxide administered was 70%, and each period of anaesthesia lasted for 3 min. Anaesthesia administered twice a week until the 3rd instar increased the number of instars required to reach maturity. However, repeated anaesthesia after the 4th instar scarcely influenced the number of instars. On the other hand, repeated anesthesia prolonged larval duration whenever administered during the early or late instars. It is thus suggested that anaesthesia by carbon dioxide during larval development has two distinct effects: increasing the number of moults and prolonging larval duration. The former is stage specific and effective only during early instars, whereas the latter is effective during any particular instars until the last instar. The body size of resulting adults remained almost the same as for controls even after the repeated carbon-dioxide anaesthesia as many as 14 times. Some further observations concerning correlations between the number of moults, larval duration and adult body size are shown, and differences between the sexes regarding the number of instars and larval duration are also presented.  相似文献

Aim We reconstructed the phylogeny of the lichen genus Nephroma (Peltigerales) to assess the relationships of species endemic to Macaronesia. We estimated dates of divergences to test the hypothesis that the species arose in Macaronesia (neo‐endemism) versus the oceanic archipelagos serving as refugia for formerly widespread taxa (palaeo‐endemism). Location Cosmopolitan with a special focus on the archipelagos of the Azores, Madeira and the Canary Islands. Methods DNA sequences were obtained from 18 species for three loci and analysed using maximum parsimony, maximum likelihood and Bayesian inferences. Divergence dates were estimated for the internal transcribed spacer (ITS)‐based phylogeny using a relaxed molecular clock. Reconstruction of the ancestral geographical range was conducted using the Bayesian 50% majority rule consensus tree under a parsimony method. Results The backbone phylogenetic tree was fully supported, with Nephroma plumbeum as sister to all other species. Four strongly supported clades were detected: the Nephroma helveticum, the N. bellum, the N. laevigatum and the N. parile clades. The latter two share a common ancestor and each includes a widespread Holarctic species (N. laevigatum and N. parile, respectively) and all species endemic to Macaronesia. The data suggest a neo‐endemic origin of Macaronesian taxa, a recent range expansion from Macaronesia of both widespread species, a range expansion limited to the Mediteranean Basin and south‐western Europe for another taxon, and a long dispersal event that resulted in a speciation event in the western parts of North America. Main conclusions The Macaronesian endemic species belong to two sister clades and originated from a most recent common ancestor (MRCA) shared with one widely distributed taxon, either N. parile or N. laevigatum. Estimates of the mean divergence dates suggest that the endemics originated in the archipelagos after the rise of the volcanic islands, along with the ancestor to the now widespread species, which probably expanded their range beyond Macaronesia via long‐distance dispersal. This study provides the first phylogenetic evidence of Macaronesian neo‐endemism in lichenized fungi and provides support for the hypothesis that oceanic islands may serve as a source for the colonization of continents. However, further data are needed to properly assess the alternative hypothesis, namely colonization from western North America.  相似文献

The eutherian placenta is considered to possess great plasticity, but it is not clear how this variation reflects adaptation to different ecological niches. Because South America was isolated for most of the Tertiary, it represents a natural laboratory to examine this question. We here describe placentation in three South American groups: Xenarthra have been part of the fauna from at least the mid‐Paleocene whereas caviomorph rodents and Neotropical primates are each derived from a single founder that reached South America in the Eocene and Oligocene, respectively. The common ancestor of Xenarthra had a villous, haemochorial placenta, from which the labyrinthine, endotheliochorial placenta of sloths later evolved. Placentation in Caviomorpha follows an extraordinary stable pattern, characterized by a haemomonochorial, labyrinthine and highly lobed structure with specialized growing areas. This pattern was present before arrival of these rodents in South America and enabled a successful radiation especially during the spread of grasslands. Neotropical primates have haemochorial, trabecular placentas with a specialized maternal blood supply; a pattern that contrasts with that of Old World monkeys and may have been present in the founder generation on arrival in South America. In conclusion, there is a dichotomy within Xenarthra but otherwise the ancient South American mammals do not show much variation in principal placental characters. Thus, the successful radiation of these three groups, and their adaptation to diverse ecological niches, did not require substantial alterations in placentation. J. Abstract

In this study, we assume that microwave radiation affects hydrogen bonding between dipolar water molecules and through that diffusion in water at constant temperature. The experimental study was performed on the setup of two identical reservoirs filled with pure water and 0.9% NaCl solution and connected by a thin tube. Alterations of NaCl concentration in the reservoir initially filled with pure water were measured using the resistance of the solution as an indicator. The applied 450?MHz continuous-wave microwave field had the maximal specific absorption rate of 0.4?W/kg on the connecting tube. The standard deviation of water temperature in the setup was 0.02?°C during an experiment. Our experimental data demonstrated that microwave exposure makes faster the process of diffusion in water. The time required for reduction of initial resistance of the solution by 10% was 1.7 times shorter with microwave. This result is consistent with the proposed mechanism of low-level microwave effect: microwave radiation, rotating dipolar water molecules, causes high-frequency alterations of hydrogen bonds between water molecules, thereby affects its viscosity and makes faster diffusion.  相似文献

Radiotherapy is one of the most important treatments for chest tumours. Although there are plenty of strategies to prevent damage to normal lung tissues, it cannot be avoided with the emergence of radiation‐induced lung injury. The purpose of this study was to investigate the potential radioprotective effects of glucosamine, which exerted anti‐inflammatory activity in joint inflammation. In this study, we found glucosamine relieved inflammatory response and structural damages in lung tissues after radiation via HE staining. Then, we detected the level of epithelial‐mesenchymal transition marker in vitro and in vivo, which we could clearly observe that glucosamine treatment inhibited epithelial‐mesenchymal transition. Besides, we found glucosamine could inhibit apoptosis and promote proliferation of normal lung epithelial cells in vitro caused by radiation. In conclusion, our data showed that glucosamine alleviated radiation‐induced lung injury via inhibiting epithelial‐mesenchymal transition, which indicated glucosamine could be a novel potential radioprotector for radiation‐induced lung injury.  相似文献
