The present study aimed to investigate the effect of supplemental genistein (an isoflavonoid) on performance, lymphoid organs’ development, and cellular and humoral immune responses in broiler chicks. A total of 675-day-old male broiler chicks (Ross 308) were randomly assigned to the five replicate pens (15 chicks each) of nine experimental diets. Dietary treatments included a negative (not-supplemented) control diet, two positive control groups (virginiamycin or zinc-bacitracin, 20 mg/kg), and diets containing 10, 20, 40, 80, 160 and 320 mg/kg of genistein. The cutaneous basophil hypersensivity (CBH) test was measured at day 10 of age after toe web injection with phytohemagglutinin-P. In addition, sera samples were collected after different antigen inoculations to investigate antibody responses. At day 28 of age, three randomly selected birds from each pen were euthanized to evaluate the relative weights of lymphoid organs. Results showed that dietary supplementation of both antibiotics increased (P<0.01) feed intake during 1 to 42 days of age. Furthermore, daily weight gain was influenced (P<0.01) by dietary treatments throughout the trial, so that the birds fed on antibiotics and 20 to 80 mg/kg genistein diets revealed the greater weight gains compared with other experimental groups. The best (P<0.05) feed conversion ratio assigned to the birds fed on diets containing antibiotics and moderate levels (40 to 80 mg/kg) of genistein. Although the relative weights of thymus (P<0.05) and bursa of Fabricius (P<0.01) were greater in birds fed on genistein-supplemented diets compared with antibiotics-supplemented birds, the spleen weight was not affected by experimental diets. Similarly, CBH response and antibody titers against Newcastle and infectious bronchitis disease viruses were markedly (P<0.05) greater in chicks fed on diets supplemented with 20 to 80 mg/kg of genistein. Interestingly, the higher dosages of genistein suppressed CBH and antibody responses to the levels seen by control and antibiotics chicks. Dietary inclusion of genistein increased (P<0.05) lymphocytes and subsequently reduced (P<0.01) heterophil to lymphocyte ratio. The present findings indicate that dietary genistein supplementation at the levels of 20 to 80 mg/kg not only improves growth performance, but also could beneficially affect immunological responses in broiler chicks.  相似文献

The contents of plant vacuoles vary in different organs and with the health of the plant, but little is known of the cell-to-cell distribution of soluble organic compounds within plant tissues. Soluble fluorescent phenolic compounds can be immobilized in plant tissues using an anhydrous freeze-substitution and resin embedment process. The vacuolar fluorescence can be characterized in fluorescence photomicrographs for variations in color and intensity, or more quantitatively with spectra obtained using a microspectrofluorometer. This is demonstrated here in freeze-substituted roots and leaves of soybean. Excitation and emission spectra of individual vacuoles can be compared with spectra of pure compounds to form profiles of the varied phenolic contents of plant vacuoles. Such analyses will add an important anatomical dimension to the study of plant defense and stress responses.  相似文献

A new isoflavonoid phytoalexin isolated from the fungus-inoculated leaflets of Lotus hispidus (hairy birdsfoot trefoil) has been identified as 5,4′-dimethoxy-7,2′-dihydroxyisoflavan (5-methoxyvestitol). Three known isoflavans (demethylvestitol, vestitol and sativan) are also produced by L. hispidus. The synthesis of 5-, 6- and 8-methoxyvestitol is described. Preparation of the pterocarpan analogues of 6- and 8-methoxyvestitol has allowed the structures of two additional legume phytoalexins to be unequivocally confirmed.  相似文献

Success of a mycorrhizal symbiosis is influenced by the availability of phosphorus (P) in the soil. Maize ( Zea mays L. cv. Great Lakes 586) plants were grown under five different levels of soil P, either in the presence or absence of formononetin or the vesicular‐arbuscular mycorrhizal (VAM) fungus Glomus intraradices Schenck and Smith. We detected physiological differences in mycorrhizal roots very early in the development of symbiosis, before the onset of nutrient‐dependent responses. Under low P levels, VAM roots accumulated a greater shoot dry weight (13%), root P concentration (15%) and protein concentration (30%) than non-VAM roots, although root growth was not statistically significantly different. At higher P levels, mycorrhizal roots weighed less than non-VAM roots (10%) without a concomitant host alteration of growth or root P concentration. Mycorrhizal colonization decreased as soil P increased. Formononetin-treatment enhanced colonization of the root by G. intraradices and partially overcame inhibition of VAM colonization by high soil P concentrations. This is the first report that formononetin improves root colonization under high levels of soil P. Acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were closely related to the level of fungal colonization in corn roots. ACP activity in corn roots responded more to soil P availability than did ALP activity (38% more). These results suggest that ACP was involved in the increased uptake of P from the soil, while ALP may be linked to active phosphate assimilation or transport in mycorrhizal roots. Thus, soil P directly affected a number of enzymes essential in host-endophyte interplay, while formononetin enhanced fungal colonization.  相似文献

Systems of wild and cultivated relatives are good experimental systems to test chemical defense theory because they provide closely related varieties that differ in discrete traits. To determine the relationship between resistance and chemical defense diversity among wild, landrace and cultivar accessions of Phaseolus vulgaris, we measured resistance to fungal infection in laboratory and field experiments, quantified phytoalexin diversity, and assessed the effectiveness of phytoalexin mixtures extracted from live tissue. Results show a gradient of resistance to fungal infections between wild, landrace and cultivar varieties. In the laboratory, wild seedlings were more resistant (93% non-infected) than landrace seedlings (80% non-infected) and modern cultivar seedlings (68% non-infected). Under field conditions wild seedlings were more resistant (97% non-infected) than cultivar seedlings (71% non-infected). Wild seedlings presented the highest phytoalexin diversity (H=1.11), while those of the landrace presented an intermediate level (H=0.97) and cultivar seedlings presented the lowest diversity (H=0.93). No differences were found in total concentrations. The in vitro inhibitory properties on hyphal growth of the isoflavonoid mixtures produced by individual seedlings showed a similar trend. Our results are consistent with similar gradients in other species of Phaseolus beans and resistance to Colletotrichum sublineolum in sorghum.  相似文献

Considerable differences in flavonoid composition of the trunkwood characterize different specimens of Myroxylon balsamum (L.) Harms. Only calycosin among the 11 flavonoids found in M. peruiferum L.f., presently considered synonymous with M. balsamum, had previously been located in the latter species. Two of these flavonoids, 2′-hydroxy-7,3′,4′-trimethoxyisoflavanone and 2′-hydroxy-7,3′,4′-trimethoxyisoflavone are new natural products.  相似文献

Detached leaves of 25 annual and perennial Medicago species have been inoculated with the fungus, Helminthosporium carbonum, and examined for subsequent production of isoflavonoid phytoalexins. Hydroxylated pterocarpan (medicarpin) and isoflavan (vestitol, sativan and isosativan) phytoalexins were obtained from most of the species investigated. Although large quantities of two flavonoid derivatives, isoliquiritigenin and liquiritigenin, were isolated from leaves of M. lupulina, this species was apparently unable to produce isoflavonoid compounds. Traces of 7-hydroxy-2′,4′-dimethoxyisoflavanone (a substance not previously recorded in the tribe Trifolieae) were obtained (together with medicarpin, vestitol and sativan) from M. sativa cv Du Puits. Some taxonomic aspects of phytoalexin induction are discussed with reference to the generic location of certain controversial Medicago species including M. (Trigonella) radiata.  相似文献
