The role of water temperature and altitude of the river sites on distribution, prevalence and infection intensity of Tetracapsuloides bryosalmonae infected salmonids was investigated. The study is based on a 6 year survey of salmonids, mainly brown trout, Salmo trutta from 2000 to 2006 at 287 sampling sites across Switzerland. Almost 7000 fishes were tested for the presence of T. bryosalmonae by histological and immunohistochemical techniques. A good correlation between altitude and water temperature in Swiss rivers could be demonstrated. Therefore, the relation between altitude and sites tested for T. bryosalmonae infected fishes was investigated since water temperature values from these sites were not available. Percentage of sites with positive (infected) fishes was identical for altitudes between 200 and 800 m above sea level (m a.s.l.), while above 800 m a.s.l. only three sites with infected fishes were recorded. Site‐specific prevalence values ranging from 0 to 100% and infection intensities from 0 to 6 were found over the whole altitude range from 200 to 800 m a.s.l., and no correlation with altitude was found. Conditions at altitudes >800 m may not be favourable for the establishment of the infection in fishes. With rising temperatures due to climatic changes, however, the prevalence of T. bryosalmonae infected fishes in altitudes >800 m a.s.l. could increase.  相似文献

To understand the role of the superoxide-generating NADPH oxidase NOX1 in the vascular system, we have generated NOX1-deficient mice. NOX1-deficient mice had a moderately decreased basal blood pressure. In response to angiotensin II they showed an almost complete loss of the sustained blood pressure response, while the initial increase was conserved. NOX1-deficient mice showed a marked reduction in aortic media hypertrophy. Angiotensin II-induced smooth muscle cell proliferation was conserved, but there was a marked decrease in extracellular matrix accumulation. Our results establish a role for NOX1 in blood pressure regulation and vascular angiotensin II response.  相似文献

An expressed sequence tag (EST) of B cell translocation gene (BTG) 1 (gcbtg1) was obtained from a grass carp Ctenopharyngodon idellus intestinal complementary (c)DNA library and the full-length cDNA sequence was acquired by rapid amplification of cDNA ends (RACE) technology. The predicted Gcbtg1 protein contains the box A and box B motifs which characterized the BTG and transducer of ERBB2 (TOB) family. Multiple alignment analysis reveals that Gcbtg1 shares an overall identity of 65-94% with Gcbtg1s of other vertebrates. Real-time quantitative PCR analysis reveals that the highest expression level of gcbtg1 was detected in liver and the lowest in muscle. Western blotting analysis indicates that the immunological cross-reactivity occurs between C. idella and human Homo sapiens BTG1 protein. A 1008 bp 5'-flanking region sequence was cloned and analysed.  相似文献

The inhibitory effects of exogenous melatonin (MEL) on colon oncogenesis were investigated using an azoxymethane (AOM)/dextran sodium sulfate (DSS) rat model. Male F344 rats initiated with a single intraperitoneal injection of AOM (20 mg/kg bw) were promoted by 1% (w/v) DSS in drinking water for 7 days. They were then given 0.4, 2 or 10 ppm MEL in drinking water for 17 weeks. At week 20, the development of colonic adenocarcinoma was significantly inhibited by the administration with MEL dose-dependently. MEL exposure modulated the mitotic and apoptotic indices in the colonic adenocarcinomas that developed and lowered the immunohistochemical expression of nuclear factor kappa B, tumor necrosis factor α, interleukin-1β and STAT3 in the epithelial malignancies. These results may indicate the beneficial effects of MEL on colitis-related colon carcinogenesis and a potential application for inhibiting colorectal cancer development in the inflamed colon.  相似文献

Aim:  Lawsonia intracellularis is an obligate intracellular bacterium causing proliferative enteropathy in certain species of domestic, laboratory and captive animals. The aim of our study was to detect L. intracellularis in free-living rodents from pig farms in the Czech Republic.
Methods and Results:  Lawsonia intracellularis was demonstrated, using nested polymerase chain reaction, in the intestinal mucous membranes of 107 (36%) out of 296 small terrestrial mammals collected. In rodents, the bacterium was detected in 91 of 213 house mice, one of six brown rats, eight of 51 striped field mice, three of 12 yellow-necked mice and three of nine common voles. Moreover, one of one tested lesser white-toothed shrew (Insectivora) was positive. In 17 out of 69 (25%) rodents tested, serum IgG antibodies against L. intracellularis were detected using an indirect immunofluorescence antibody test. All seropositive animals were house mice.
Conclusions:  Free-living rodents may be serving as host or reservoir species and may be playing a role in the spread of L. intracellularis both within pig farms and in its transmission between farms and into the natural environment.
Significance and Impact of the Study:  This is the first detection of L. intracellularis in free-living small mammalian terrestrial species.  相似文献

Xao tam phan (Paramignya trimera) has been used for the treatment of cancer and cancer‐like aliments. Among different parts of the Ptrimera plant, leaf is considered as a residual part after harvesting of the root. This study aimed to determine the physiochemical properties and the antioxidant and anti‐proliferative capacities of Ptrimera leaf (PTL) using microwave drying for the preparation of dry sample; MeOH and microwave‐assisted extraction for the preparation of crude extract; and freeze‐drying for the preparation of powdered extract. The results showed that total phenolic, total flavonoid, proanthocyanidin, and saponin contents of PTL prepared by microwave drying at 450 W were 25.4 mg gallic acid equiv. (GAE), 86.3 mg rutin equiv. (RE), 5.6 mg catechin equiv. (CE), and 702.1 mg escin equiv. (EE) per gram dried sample, respectively. Gallic acid, protocatechuic acid, ellagic acid, rutin, and quercetin were identified in the PTL MeOH extract. Dried PTL displayed potent antioxidant activity, while the powdered PTL extract exhibited great anti‐proliferative capacity on various cancer cell lines including MiaPaCa‐2 (pancreas), HT29 (colon), A2780 (ovarian), H460 (lung), A431 (skin), Du145 (prostate), BE2‐C (neuroblastoma), MCF‐7 (breast), MCF‐10A (normal breast), and U87, SJ‐G2, and SMA (glioblastoma). Anti‐proliferative capacity on pancreatic cancer cells (MiaCaPa2, BxPc3, and CFPAC1) of PTL extract (200 μg/ml) was significantly higher (< 0.05) than those of ostruthin (20 μg/ml) and gemcitabine (50 nm ), and to be comparable to the powdered Ptrimera root extract and a saponin‐enriched extract from quillajia bark (a commercial product). The findings from this study allow us to conclude that the PTL is a rich source of phytochemicals that possess promising antioxidant and anti‐proliferative activities, therefore it shows potential as lead compounds for application in the nutraceutical, medicinal and pharmaceutical industries.  相似文献

The properties of some stem cells (SCs) that are most interesting in terms of their implications for medicine (embryonic, hematopoietic, and mesenchymal SCs) are considered. SCs are undifferentiated cells capable of both self-maintenance and differentiation into specialized cells. According to their origin, SCs are divided into embryonic and somatic ones. The former can be maintained in vitro for an infinitely long time and can differentiate into all cells of adult organisms. The latter have a limited capacity for differentiation and, probably, a limited proliferative potential. The plasticity of somatic SCs, i.e., their capacity for context-dependent differentiation into unrelated cell types, is of considerable therapeutic importance, although some researchers doubt this capacity. It is assumed that most types of SCs differentiate by the stepwise hierarchical maturation mechanism, one of the steps being rapidly proliferating progenitor cells. The use of SCs in medicine is currently at the stage of preclinical trials. Although embryonic SCs are promising for medicine, there are serious limitations of their use in therapy in the near future. However, the first clinical trials have demonstrated that the approaches involving autotransplantation of hematopoietic and mesenchymal SCs are effective for treating ischemia of extremities and the consequences of myocardial infarction. Obviously, the use of SCs in medicine promises dramatic progress in treating many severe diseases.  相似文献
