17β-estradiol propionate was found to reduce the gap junctional communication in a concentration range similar to that of
testosterone propionate, in primary cultures of rat Sertoli cells and cardiac myocytes. Uncoupling was reversible on washing
out and occurred without concomitant rise in the intracellular calcium concentration.
Esterification was a prerequisite for the activity of extracellularly applied steroid compounds (for example, testosterone
was ineffective even at external concentrations up to 100 μm, whereas its intracellular application at 1 μm totally interrupted intercellular communication), but their uncoupling efficiency did not depend on the nature of the ester
chain nor on its position on the steroid nucleus. The derivatives of two other androgen hormones (derivatives of the androstane
nucleus) were also efficient as junctional uncouplers. Among five steroid molecules belonging to the pregnane family, only
one (pregnanediol diacetate) interrupted the junctional communication. Neither cholic acid nor cholesteryl acetate or ouabain
showed this effect. Altogether, no correlation with the presence or position of double bonds nor with the trans- or cis-fusion of the A and B rings could be recognized.
These results suggest that this reversible, nondeleterious uncoupling effect of steroids is independent of the shape of the
molecules and is more probably related to their size and liposolubility, that condition their insertion into the lipid bilayer.
Their incorporation into the membrane could disturb the activity of the membrane proteins by a physical mechanism.

Uptake rate of calcium, potassium, nitrate-N and phosphorus were measured in a second order Mediterranean temporary stream, in February and March 1992. This study analyzed a period of continuous surface flow between two hydrologic disturbance events (flood and drought) of an annual hydrological cycle (1991–92).The lowest values of uptake length were recorded for nitrate-N in February 92 and calcium in March 92. Nitrate had the highest uptake rate in both release performances, and potassium showed the lowest uptake rate values. The increase of calcium and nitrate uptake rate between February 92 and March 92 suggested a higher ecosystem efficiency in nutrient retention with a higher temperature and light intensity and slower water velocity, discharge and water depth. These results obtained were similar to those reported in permanent streams, indicating that in periods of continuous surface flow (without extreme hydrologic disturbance), abiotic factors can influence nutrient retention in temporary streams. 相似文献

Endothelin (ET-1) is found at elevated concentrations in the plasma of patients with heart failure and in animal models of cardiomyopathy. The peptide is a potent positive inotropic agent, the effects of which are mediated by increases in cytosolic Ca2+ in cardiomyocytes. The object of this study was to investigate at the cellular level, the actions of ET-1 on contractile function and on Ca2+ currents in heart-failed ventricular myocardium. Male New Zealand White rabbits (8 wks) were treated with twice weekly injections of epirubicin (4 mg/kg/wk, n=7) or with saline (n=7) for 6 wks, followed by a washout period of 2 wks. Ventricular cardiomyocytes were isolated from rabbit hearts using Langendorff perfusion with collagenase; contractile function was examined using a video microscopy method, and L-type Ca2+ currents were recorded using a whole-cell patch-clamp technique. ET-1 produced a concentration-dependent increase in contractile response (% increase from basal value) to a maximum at 1 nM ET-1 of 69 ± 11% (mean ± S.D.) in control cardiomyocytes and 33 ± 6% in heart-failed cells. However, there was no significant change in the EC50 obtained with ET-1 for healthy (0.31 ± 0.1 nM) and for failed cardiomyocytes (0.24 ± 0.1 nM). The effects of ET-1 on L-type Ca2+ channels were similar with a peak amplitude at 1 nM ET-1 of –3.26 ± 0.8 in control cardiomyocytes and –3.32 ± 0.9 nA in heart-failed cells. The attenuation of the contractile response to ET-1 in heart-failed cells may reflect a desensitization of ET receptors as a consequence of elevated circulating levels of ET and was not reflected by alteration of transmembrane Ca2+ conductance. In primary cultures of neonatal rat heart cells we found a linear correlation between the number of L-type calcium channel-specific dihydropyridine (DHP) binding sites and spontaneous beating frequency (v).Formation of glycoproteins in tissue culture was suppressed by different inhibitors of N-glycosylation. This inhibition alters to a different extent the binding of the DHP ligand (+)-[methyl-3H]PN 200-110 and v. The most severe but reversible effect occurs at 6 g/ml tunicamycin (Bmax 45% and v 6%, resp., of control), a slight increase in Bmax at 0.1–0.5 mM castanospermine and 0.05–2.5 mM deoxymannojirimycin. We investigated the effect of 10–8 M noradrenaline (NA) on [Ca2+], and electrical activity of single myocytes of guinea-pig ventricular myocardium loaded with Indo 1-AM. Membrane potential was recorded by means of the patch electrode and patch amplifier set to the current clamp mode. Cells were stimulated at a rate of 30/min by 3 ms pulses of the current injected through the recording electrode. Superfusion of NA resulted in slight shortening of action potentials (APs), increase in rate of rise and amplitude of the respective Ca2+ transients, and appearance of secondary Ca2+ transients of two kinds: 1. appearing before repolarisation of AP and decay of the preceding Ca2+ transient were completed and 2. appearing between the APs. We named them early after-transients (EAT) and delayed after-transients (DAT), respectively. Without any additional intervention EATS caused some prolongation of APs duration and DATs resulted in subthreshold delayed after-depolarisations (DADS). When sarcolemmal K+ conductance was decreased by tetraethylammonium (TEA) in the patch electrode or 20 M BaCl2 in the Tyrode solution, EATs initiated early after depolarizations (EADs) and DATs initiated suprathreshold DADs triggering full-sized APs. Superfusion of 30.0 mM Na+ (replaced with LiCl) resulted in reduction of AP duration by -70% and appearance of DATs. Also, the frequent multiple oscillations of Ca 2+ concentration were often observed. Neither DATs nor the oscillations had any affect on electrical activity of the cells. Their electrogenicity could not be increased by TEA or 20.0 M Ba2+. EATs and DATs and their respective EADs and DADs could not be initiated by NA or low Na+ superfusion in the cells pretreated with 2 × 10–7 M thapsigargin, a selective blocker of Ca2+-ATPase of sarcoplasmic reticulum (SR). We conclude that in contrast to the current hypothesis, EADs can be initiated by Ca2+ released early in the cardiac cycle from the overloaded SR, and that electrogenicity of both types of Ca2+ oscillations critically depends on the sarcolemmal K+ conductance. 相似文献

The purpose of this study was to determine the extent of aluminum (Al) accumulation in the human aorta and cerebral arteries.
The Al contents in the aortae and in the cerebral arteries from 23 human subjects was determined by inductively coupled plasma
atomic emission spectrophotometry (ICP-AES). The subjects' age range was 45–99-yr-old; 15 of the subjects were males and 8
were females. Al was detected in twelve aortae and in six cerebral arteries, when the entire specimen was analyzed. Two specimens
where Al was found in the cerebral arteries contained no Al in the aorta. No relationship to the subject's sex was found.
When related to age, two groups were established. Group L (45–75 yr old) and group H (>75 yr old), which exhibited aortal
Al concentrations of 33.3 and 72.7%, respectively. When the aortic wall was dissected into the tunica intima, media, and adventitia,
Al was found mainly in the tunica media. In the aorta, significant relationships were found between Al and phosphorus (P)
levels (r=0.801,p<0.01) and between Al and calcium (Ca) (r=0.661,p<0.05). We have concluded that Al accumulation is age-dependent and that it occurs both in the aorta and in the cerebral artery.
In the aorta, accumulation occurs mainly in the tunica media. Both P and Ca appear to enhance aortal Al accumulation. 相似文献

Three groups of rats were fed two types of synthetic diets for 52 d. The—A group was allowed free access to a vitamin A-deficient
diet and showed classical signs of vitamin A deficiency. The brain was the only organ in our experiment where no significant
weight difference was present among the three groups. In the brain, calcium concentration was significantly higher in the—A
group when compared with the PF (Pair-fed; allowed restricted amount of control diet) and +A groups (allowed free access to
control diet). In the tibia, calcium and magnesium concentrations were significantly lower in the—A group when compared with
other two groups. Excessive accumulation of calcium in brain and apparently similar unbalance in bone, mineral concentration
were observed in central nervous system (CNS) degenerative diseases. Our results suggest that abnormal metabolism of calcium
and magnesium in some tissues and excessive accumulation of calcium in brain may be responsible for the development of neurological

The element Cd is considered to have no biological function and is highly toxic to humans and animals. Toxic effects of this
metal upon cell membrane structure and function have been shown. On the other hand, Ca is an essential element in a wide variety
of cellular activities. The present study was initiated to research whether the interaction between Ca and Cd could affect
D-galactose absorption across the rabbit jejunum in vitro. In media with Ca2+, when CdCl2 was present at 0.5 or 1 mM, Cd was found to significantly reduce the sugar absorption. In Ca2+-free media, where CaCl2, was omitted and replaced isotonically with choline chloride, the sugar transport was not modified by Cd, but when CaCl2 was replaced isotonically with MgCl2, the inhibition is observed. Verapamil at 10−6
M (blocking mainly Ca2+ transport) did not modify the inhibitory effect of cadmium on D-galactose transport. When 10−6
M of A 23187 (Ca2+ specific ionophore) was added in media with/without Ca2+; CdCl2 produced no change in D-galactose transport. These results suggest that Ca and Cd could have affinity for the same chemical

Thylakoids and Photosystem II particles prepared from the cyanobacterium Synechococcus PCC 7942 washed with a HEPES/glycerol buffer exhibited low rates of light-induced oxygen evolution. Addition of either Ca2+ or Mg2+ to both thylakoids and Photosystem II particles increased oxygen evolution independently, maximal rates being obtained by addition of both ions. If either preparation was washed with NaCl, light induced O2 evolution was completely inhibited, but re-activated in the same manner by Ca2+ and Mg2+ but to a lower level. In the presence of Mg2+, the reactivation of O2 evolution by Ca2+ allowed sigmoid kinetics, implying co-operative binding. The results are interpreted as indicating that not only Ca2+, but also Mg2+, is essential for light-induced oxygen evolution in thylakoids and Photosystem II particles from Synechococcus PC 7942. The significance of the reactivation kinetics is discussed. Reactivation by Ca2+ was inhibited by antibodies to mammalian calmodulin, indicating that the binding site in Photosystem II may be analogous to that of this protein.Abbreviation HEPES
