Prenatal hypoxia (PH) is a common pregnancy complication, harmful to brain development. This study investigated whether and how PH affected Wnt pathway in the brain. Pregnant rats were exposed to hypoxia (10.5% O2) or normoxia (21% O2; Control). Foetal brain weight and body weight were decreased in the PH group, the ratio of brain weight to body weight was increased significantly. Prenatal hypoxia increased mRNA expression of Wnt3a, Wnt7a, Wnt7b and Fzd4, but not Lrp6. Activated β‐catenin protein and Fosl1 expression were also significantly up‐regulated. Increased Hif1a expression was found in the PH group associated with the higher Wnt signalling. Among 5 members of the Sfrp family, Sfrp4 was down‐regulated. In the methylation‐regulating genes, higher mRNA expressions of Dnmt1 and Dnmt3b were found in the PH group. Sodium bisulphite and sequencing revealed hyper‐methylation in the promoter region of Sfrp4 gene in the foetal brain, accounting for its decreased expression and contributing to the activation of the Wnt‐Catenin signalling. The study of PC12 cells treated with 5‐aza further approved that decreased methylation could result in the higher Sfrp4 expression. In the offspring hippocampus, protein levels of Hif1a and mRNA expression of Sfrp4 were unchanged, whereas Wnt signal pathway was inhibited. The data demonstrated that PH activated the Wnt pathway in the foetal brain, related to the hyper‐methylation of Sfrp4 as well as Hif1a signalling. Activated Wnt signalling might play acute protective roles to the foetal brain in response to hypoxia, also would result in disadvantageous influence on the offspring in long‐term.  相似文献

Physiological responses of the euryhaline red drum, Sciaenops ocellatus, to chloride salt addition, low salinity, and high sulfate concentration were measured. Survival was increased by addition of calcium chloride (CaCl2) or magnesium chloride (MgCl2) to dilute artificial seawater (0.2 ppt salinity). Although survival and routine metabolic rates were greater in MgCl2 treatments, growth and feed efficiency were greater in CaCl2 treatments. Marginal metabolic scope increased when CaCl2 or MgCl2 were added to dilute artificial seawater. There was a strong positive linear relationship (p=0.0001, r=0.91) between fish survival and salinity of artificial seawater dilutions over the salinity range 0.1 to 3.0 ppt. Monovalent ion concentrations in red drum plasma varied; whereas, divalent ion concentrations were relatively constant. Survival and growth were not affected by high sulfate concentrations (2000 mg l-1) in 3.0 ppt artificial seawater supplemented with either sodium sulfate or magnesium sulfate. Routine metabolic rate and marginal metabolic scope of red drum exposed to high sulfate concentrations were slightly, but not significantly, lower than those of red drum in 3 ppt artificial seawater.  相似文献

在建立S~2=a'(-lnP_0)~b'模型的基础上,结合Gerrard&Chiang模型(m=a(-InP_0)~β)和扩散系数C,本文提出一个种群空间格局研究的零频率方法.根据种群聚集临界零频率,可对空间格局连续统作出定量描述,按参数(b'-β)可将生物种群划分为“聚集度非零频率制约型”、“聚集度逆零频率制约型”和“聚集度零频率制约型”3种类型.通过13种生物种群资料的实例验证,表明新模型能客现地反映出种群空间格局的本质特征,应用于低密度种群已显示出它的独到之处.此外,文中还推导出了确定理论抽样数的公式和简易序贯抽样决策模型.  相似文献

Miscanthus x ogiformis Honda 'Giganteus' shoot cultures were stored in vitro on proliferation or rooting medium for up to 27 weeks at temperatures of 8, 12, 16, or 20 °C and photosynthetic photon flux densities of 5, 10, or 20 μmol m−2 s−1. Plants survived storage much better on rooting medium than on proliferation medium. Plants stored on rooting medium for 1 week survived well when survival was assessed immediately after storage or after 14 days of acclimatization, but had the lowest survival 28 days after transplantation. With increasing storage period on rooting medium increasing survival was found 28 days after transplantation. This was probably a result of the development of rhizomes and/or roots during storage. Best survival was observed at 20 μmol m−2 s−1 and a temperature of 8-16 °C. Increasing the temperature to 25 °C during the last week of storage improved survival considerably. Root formation was slow at 8 °C, but after 27 weeks of storage the rooting percentage was the same at all storage temperatures. An increasing number of shoots per plant 28 days after transplantation was found with increasing PPFD during storage.Miscanthus shoot cultures can be stored in vitro for at least 27 weeks with limited losses when stored on rooting medium at 20 μmol m−2 s−1, a temperature of 16 °C, and given a 1-week end-of-storage treatment of 25 °C. Balance trials were performed to investigate the effects of experimental Eimeria bovis coccidiosis on the metabolism of water, sodium and potassium in calves. Non-infected pair-fed controls and controls fed according to plan were included in the study to allow differentiation between the effects due to infection and due to changes in feed intake. Primary infection with 5 × 104 (group A) or 1 × 105 (group B) oocysts caused mild diarrhoea in three out of four group A calves and mild to severe haemorrhagic diarrhoea in all five group B calves. Losses of sodium and potassium via faeces tended to increase in the infected calves during patency and apparent digestibility (AD) of these minerals was comparably low. In the urine of the infected calves the Na/K-ratio decreased due to a reduced urinary excretion of sodium. The retention (RT) of sodium was particularly high in the calves that had received the higher oocyst dose. Potassium RT did not underlie significant changes during the course of coccidiosis. In the infected calves the plasma level of sodium was reduced transiently while the level of potassium remained fairly stable. Infections with the higher oocyst dose caused a distinct reduction of fluid excretion via urine which compensated for the increased faecal water losses during severe diarrhoea. Reinfection of the group A calves with 1 × 105 oocysts did not cause any significant metabolic impairment. The results of this study indicate that although acute sublethal bovine coccidiosis alters electrolyte and water metabolism the overall balance of electrolytes and water is largely maintained by physiologic adaptation.  相似文献

Haemolysis has been used as an initial selection criterion for the primary isolation of surfactant-producing bacteria. Only 37 of 492 strains of different origins had haemolytic activity. These 37 strains, together with 49 non-haemolytic ones chosen at random, were studied for surface activity. Only five strains, all of them haemolytic, tested positive. Haemolysis and biosurfactant-production are thus probably associated.  相似文献

When leaves of oilseed rape (cv. Cobra) were inoculated with conidial suspensions of Mycosphaerella capsellae (white leaf spot) and incubated in controlled environments, the lag period from inoculation to the appearance of the first lesions decreased, and the total number of lesions produced increased, as temperature increased from 5oC to 20oC, although differences between 15oC and 20oC were small. With incubation period estimated as the time from inoculation until 5%, 50% or 95% of the lesions were produced, there was a linear relationship between l/(incubation period in days) and temperature over the range 5oC to 20oC, from which values at intermediate temperatures could be estimated. Summed mean daily temperatures from inoculation to the production of 5% of the lesions were estimated as 115–130 degree-days in the controlled environment experiments at 5oC to 20oC. When pods or leaves of plants in oilseed rape crops (cv. Cobra or cv. Libravo) were inoculated with conidial suspensions of M. capsellae on five occasions from January to October, with variable temperatures during the incubation period, degree-days until the first appearance of lesions were in the range 115–230. The numbers of white leaf spot lesions cm-2 which developed on inoculated leaves differed greatly between nine oilseed rape cultivars, with most on cv. Tapidor and fewest on cv. Libravo, but the incubation period differed little between cultivars. Similarly, the number of lesions which developed differed between four M. capsellae isolates from different regions but the incubation period did not.  相似文献

Chickpea (Cicer arietinum L.) is extensively grown in areas where soils are deficient in zinc (Zn). To determine the response of chickpea to Zn nutrition and to diagnose Zn status in plant tissue, two glasshouse experiments were conducted using Zn-deficient siliceous sandy soil. In Experiment 1, two genotypes of desi chickpea (Dooen and Tyson) were grown at five Zn levels (0, 0.04, 0.2, 1.0 and 5.0 mg kg-1 of soil). After 4 weeks, no difference in growth and no visible symptoms of Zn deficiency were detected. After 6–8 weeks of growth, chlorosis of younger leaves and stipules occured in the Zn0 treatment, with shoot dry weight being only 24% of that recorded at the highest Zn level. Root growth increased from 0.52 g/plant when no Zn was applied to 1.04 g/plant in the treatment with 0.2 mg Zn kg-1 of soil; no response to further increase of Zn fertilization occurred. Zinc concentration in the whole shoot increased significantly with increased in Zn application. The critical Zn concentration in the shoot tissue, associated with 90% of maximum growth, was 20 mg kg-1 for both genotypes at flowering stage.In the second experiment, two genotypes of desi chickpea (Tyson and T-1587) were grown at three Zn levels (0, 0.5 and 2.5 mg kg-1 of soil) under two moisture regimes (field capacity 12% w/w, and water stress 4% w/w). Shoot growth was influenced by both Zn supply and water stress. The effect of water stress was severe in the 0.5 and 2.5 mg Zn treatments where shoot dry matter was reduced 52 and 46%, respectively. T-1587 was less sensitive to Zn deficiency and produced higher shoot dry weight than Tyson in the Zn0 treatment. Zinc concentration in shoots increased from 5 mg kg-1 when no Zn was applied to 40 mg kg-1 at the highest Zn level. The critical Zn concentration in shoots was 21 mg kg-1.The results of the two experiments showed that the critical concentration for Zn did not differ amongst the three cultivars used and was not affected by soil moisture. Similar studies should be undertaken with a wider number of genotypes to discover if a critical concentration of 20–21 mg kg-1 in the shoot can be used to diagose the Zn status of chickpea genotypes.  相似文献
