Myostatin (MSTN), or growth and differentiation factor 8 (GDF8), is a member of the transforming growth factor (TGF)-β superfamily. This family functions as a negative regulator of skeletal muscle development and growth in mammals. Single-nucleotide polymorphisms in exon 1 of the Bian chicken myostatin gene were detected by polymerase chain reaction-restriction fragment length polymorphism. A mutation (c.234G>A) in exon 1 was found. Female Bian chickens of genotypes AA and GA had significantly higher body weights than those of genotype GG (P < 0.05 or P < 0.01) from 6 to 18 weeks of age. These results suggested that the mutation c.234G>A in exon 1 could be used as a genetic marker for Bian chicken growth traits.  相似文献

The purpose of the current study was to investigate whether or not the FABP2 gene polymorphism modulated obesity indices, hemodynamic factor, blood lipid factor, and insulin resistance markers through 12-week aerobic exercise training in abdominal obesity group of Korean mid-life women. A total of 243 abdominally obese subjects of Korean mid-life women voluntarily participated in aerobic exercise training program for 12 weeks. Polymerase Chain Reaction with Restriction Fragment Length Polymorphism (PCR-RFLP) assay was used to assess the FABP2 genotype of the participants (117 of AA homozygotes, 100 of AT heterozygotes, 26 of TT homozygotes). Prior to the participation of the exercise training program, baseline obesity indices, hemodynamic factor, blood lipid factor, and insulin resistance markers were measured. All the measurements were replicated following the 12-week aerobic exercise training program, and then the following results were found. After 12-week aerobic exercise training program, wild type (Ala54Ala) and mutant type (Ala54Thr+Thr54Thr) significantly decreased weight (P > .001), BMI (P > .001), %bf (P > .001), waist circumference (P > .001), WHR (P > .001), muscle mass (wild type p < .022; mutant type P > .001), RHR (P > .001), viseceral adipose area (wild type p < .005; mutant type P > .001), subcutaneous area (P > .001), insulin (wild type p < .005; mutant type P > .001) and significantly increased VO2max (P > .001). And wild type significantly decresed NEFA (P > .05), glucose (P > .05), OGTT 120min glucose (P > .05) and significantly increased HDLC (p > .005). Mutant type significantly decreased SBP (P > .001), DBP (P > .01), TC (P > .01), LPL (P > .05), LDL (P > .001), HOMA index (P > .01). The result of the present study represents that regular aerobic exercise training may beneficially prevent obesity index, blood pressure, blood lipids and insulin resistance markers independent of FABP Ala54Thr wild type and mutant type.  相似文献

Uncovering biological diversity to more accurately understand diversity patterns, and ultimately the processes driving diversification, is important not only from an evolutionary perspective but also a conservation perspective. This is particularly pertinent in Africa's rivers in which overall diversity, as well as how it arose, is poorly understood in comparison with lacustrine environments. Here we investigate population divergence in the sexually dimorphic suckermouth catfish species Chiloglanis anoterus (Crass, 1960) from the African Highveld, in which we observe striking variability in exaggerated male caudal fins across its range. As this trait is likely to be indirect evidence for sexual selection by female choice, a mechanism that has been shown to increase species diversity in different taxa, we used an integrated approach to test if current diversity in this species is underestimated. Results based on phylogenetic inference, population genetics and geometric morphometrics indicate that the recognized species C. anoterus represents five distinct lineages that may be considered confirmed candidate species. We suggest that diversification in these highland catfish has been facilitated through geographical isolation in upper river catchments, and that sexual selection through female choice has probably driven variation in male caudal fin morphology. In contrast to the relatively large range size of the currently recognized species (C. anoterus), our findings highlight highly restricted ranges of the lineages identified here, indicating that these highland habitats may harbour higher levels of endemic diversity than previously thought.  相似文献

RFLP markers and predicted testcross performance of maize sister inbreds   总被引:1,自引:0,他引:1  
 Inbreds selfed from the same F2 or backcross population are referred to as sister inbreds. In some situations, maize (Zea mays L.) sister inbreds may not have testcross data available for best linear unbiased prediction (BLUP) of single-cross performance. This study evaluated the usefulness of BLUP and restriction fragment length polymorphism (RFLP)-based coefficients of coancestry ( f ) in predicting the testcross performance of sister inbreds. Parental contributions (p) were estimated from 70 RFLP loci for 15 inbreds that comprised three sister inbreds selfed from each of five F2 populations. Estimates of p were subsequently used to calculate RFLP-based f. Grain yield, moisture, and stalk lodging data were obtained for 2265 single crosses tested by Limagrain Genetics in multilocation trials from 1990 to 1995. Performance of the sister inbreds when crossed to several inbred testers was predicted from the performance of the tested single crosses and RFLP-based f. Correlations between predicted and observed performance, obtained with a delete-one cross-validation procedure, were erratic and mostly low for all three traits. Correspondence was poor between ranks for predicted and observed general combining ability of the sister inbreds. The results suggested that the proportion of the genome derived by a sister inbred from a given parental inbred does not solely determine its testcross performance. The failure of BLUP and RFLP-based f to consistently predict testcross performance indicated that actual field testing will continue to be necessary for preliminary evaluation of sister inbreds. Fish body shape is affected by the genetic makeup of an individual as well as environmental influences, such as diet, development, growth rate and nutrition. Fishes in the family Cichlidae exhibit tremendous morphological diversity in body shape and morphology related to feeding. Certain aspects of cichlid feeding morphology have been shown to be plastic in response to different diets but plasticity in body shape has not been examined previously. Plasticity affects ecological interactions, the direction and rate of evolution, and has ramifications for characters used in systematic studies. I examined the effect of different diets: chironomid larvae (bloodworms) and brine shrimp nauplii, on body shape in two species of the Neotropical cichlid genus Geophagus which differ in the size at which young begin feeding on external food sources. The fry of G. brasiliensis , a substrate spawner, begin to feed on external food sources earlier than the fry of G. steindachneri , a mouthbrooder. I hypothesized that the difference in size at first feeding could lead to a difference in the amount of plasticity inducible in the two species. The magnitudes of changes were mostly similar, although G. brasiliensis responded to the different diets with slightly greater changes in some of the head measurements. The pattern of changes in the two species were also similar, with fish fed ferine shrimp nauplii developing longer and shallower heads and shallower bodies and tails than fish fed chironomid larvae. I also examined the consequences of considering family and age as additional factors besides diet in G. steindachneri. Considering family or age as additional factors in the analyses did not change the conclusion that different diets induce differences, albeit small ones, in body shape. I argue that morphological plasticity is dependent on behavioural flexibility and that it may enhance evolutionary morphological diversification.  相似文献

Understanding the conditions for the stable coexistence of different alleles or species is a central topic in theoretical evolution and ecology. Different causes for stable polymorphism or species coexistence have already been identified but they can be grouped into a limited number of general processes. This article is devoted to the presentation and illustration of a new process, which we call 'habitat boundary polymorphism', and which relies on two key ingredients: habitat heterogeneity and distance-limited dispersal. Under direct competition and with fixed population densities, we show that this process allows for the equilibrium coexistence of more than n types in a n-habitat environment. Distance-limited dispersal indeed creates local maladaptation at habitat edges, which leaves room for the invasion of more generalist alleles or species. This mechanism provides a generic yet neglected process for the maintenance of polymorphism or species coexistence.  相似文献

ERCC4 plays an essential role in the nucleotide excision repair (NER) pathway, which is involved in the removal of a wide variety of DNA lesions. To determine whether the ERCC4 tagging SNPs (tSNPs) are associated with risk of gastric cancer, we conducted a hospital-based case-control study of 350 cases and 468 cancer-free controls. In the logistic regression (LR) analysis, we found a significantly decreased risk of gastric cancer associated with the rs744154 GC/CC genotypes [adjusted odds ratio (OR) = 0.56, 95% confidence interval (CI) = 0.42–0.75, false discovery rate (FDR) P = 0.003] compared with the wild-type GG genotype. Haplotype-based association study revealed that the CGC haplotype that containing the rs744154 C allele can decrease the risk of gastric cancer compared with the most common haplotype GGT (adjusted OR = 0.61, 95% CI = 0.46–0.81). Using the multifactor dimensionality reduction (MDR) analysis, we identified that the SNP rs744154 and smoking status were the best two predictive factors for gastric cancer with a testing accuracy of 55.76% and a perfect cross-validation consistency (CVC) of 10 (P = 0.001). Furthermore, the smokers with the rs744154 GC/CC genotypes showed a decreased risk of gastric cancer (adjusted OR = 0.55, 95% CI = 0.35–0.85) compared with the smokers with the GG genotype using multivariate LR analysis. The above findings consistently suggested that genetic variants in the ERCC4 gene may play a protective role in the etiology of gastric cancer, even in the smokers.  相似文献

Context: Cluster of differentiation 40 (CD40), and its ligand CD40L, are major co-stimulatory molecules whose interactions are important in both cellular and humoral immunity, and has been suggested to play a role in the pathogenesis of acute coronary syndrome.

Objective: The aim of this study was to examine the association of CD40 polymorphisms (-1?C>T (rs1883832) and 945G>T (rs4810485)) and myocardial infarction (MI), and to test the association of CD40 gene haplotypes with MI in Tunisians.

Materials and methods: Three hundred and fifty MI patients and 301 apparently healthy controls were included in the study. The polymorphisms of CD40 were analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).

Results: There were significant differences in the genotype and allele frequencies of CD40 gene -1?C>T (rs1883832) polymorphism between cases and controls. Stratifying according to gender, the association between the TT genotype and MI was statistically significant in males, only. Haplotype analysis revealed that the C-T and T-G haplotypes were associated with an increased risk of MI (p?=?0.012 and p?<?0.001, respectively).

Conclusions: Our work showed a significant association between the -1?C>T (rs1883832) polymorphism of the CD40 gene and MI in the Tunisians.  相似文献
