This study was designed to examine the interaction of methacyline (METC) with human serum albumin (HSA) by multispectroscopy and a molecular modeling method under simulative physiological conditions. The quenching mechanism was suggested to be static quenching based on fluorescence and ultraviolet–visible (UV–Vis) spectroscopy. According to the Vant' Hoff equation, the values of enthalpy (?H) and entropy change (?S) were calculated to be ?95.29 kJ/mol and ?218.13 J/mol/K, indicating that the main driving force of the interaction between HSA and METC were hydrogen bonds and van der Waals's forces. By performing displacement measurements, the specific binding of METC in the vicinity of Sudlow's site I of HSA was clarified. An apparent distance of 3.05 nm between Trp214 and METC was obtained via the fluorescence resonance energy transfer (FRET) method. Furthermore, the binding details between METC and HSA were further confirmed by molecular docking studies, which revealed that METC was bound at subdomain IIA through multiple interactions, such as hydrophobic effect, polar forces, hydrogen bonding, etc. The results of three‐dimensional fluorescence and Fourier transform infrared (FTIR) spectroscopy showed that METC caused conformational and some microenvironmental changes in HSA and reduced the α‐helix significantly in the range of 52.3?40.4% in HSA secondary structure. Moreover, the coexistence of metal ions such as Ca2+, Al3+, Fe3+, Zn2+, Cu2+, Cr3+ and Cd2+ can decrease the binding constants of METC–HSA. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Floral biology, reproduction, pollinator specificity, and fruit set of Cirrhaea dependens were recorded in forest areas of Southeastern Brazil. Cirrhaea dependens is a lithophytic or epiphytic herb occurring very sparsely below dense canopies. Nearly all the flowers of a single plant open simultaneously before dawn. They are short-lasting and offer floral fragrances as rewards, which are collected by male euglossine bees. Observations carried out in mesophytic forests at Serra do Japi revealed that Euglossa VIRIDIS is their principal pollinator, with Eufriesea violacea a sporadic co-pollinator. Visitation started soon after flower opening, and attractiveness remained high for about 2 h, decreasing abruptly at sunrise. Flower anthesis with subsequent fragrance release seems to be correlated with bee attraction. Observations using chemical baits were carried out at Serra do Japi, and in the Atlantic Rain Forest of Picinguaba. Three euglossine species were captured with pollinaria of C. dependens in Picinguaba, whereas only E. violacea was attracted in Serra do Japi. In Picinguaba, C. dependens occurs sympatrically with C. saccata and C. loddigesii. Each species attracted different pollinators. The specificity and resulting reproductive isolation are due to the production of different odours by each orchid species. Cirrhaea dependens is self-compatible but pollinator-dependent. The reproductive success was low and appears to result from a combination of factors discussed here, such as the production of short-lived flowers, presence of floral mechanisms avoiding self-pollination, non-synchronization of flower phases among plants, and populations with few and sparsely distributed individuals.  相似文献

  • Field studies integrating pollination investigations with an assessment of floral scent composition and thermogenesis in tropical aroids are rather few. Thus, this study aimed to investigate the pollination biology of nine species belonging to Schismatoglottis Calyptrata Complex Clade. The flowering mechanism, visiting insect activities, reproductive system, thermogenesis and floral scent composition were examined.
  • Anthesis for all species started at dawn and lasted 25–29 h. Colocasiomyia (Diptera, Drosophilidae) are considered the main pollinators for all the investigated species. Cycreon (Coleoptera, Hydrophilidae) are considered secondary pollinators as they are only present in seven of the nine host plants, despite the fact that they are the most effective pollen carrier, carrying up to 15 times more pollen grains than Colocasiomyia flies. However, the number of Colocasiomyia individuals was six times higher than Cycreon beetles. Chaloenus (Chrysomelidae, Galeuricinae) appeared to be an inadvertent pollinator. Atheta (Coleoptera, Staphylinidae) is considered a floral visitor in most investigated species of the Calyptrata Complex Clade in Sarawak, but a possible pollinator in S. muluensis. Chironomidae midges and pteromalid wasps are considered visitors in S. calyptrata.
  • Thermogenesis in a biphasic pattern was observed in inflorescences of S. adducta, S. calyptrata, S. giamensis, S. pseudoniahensis and S. roh. The first peak occurred during pistillate anthesis; the second peak during staminate anthesis. Inflorescences of all investigated species of Calyptrata Complex Clade emitted four types of ester compound, with methyl ester‐3‐methyl‐3‐butenoic acid as a single major VOC (volatile organic compound). The appendix, pistillate zone, staminate zone and spathe emitted all these compounds.
  • A mixed fly–beetle pollination system is considered an ancestral trait in the Calyptrata Complex Clade, persisting in Sarawak taxa, whereas the marked reduction of interpistillar staminodes in taxa from Peninsular Malaysia and especially, Ambon, Indonesia, is probably linked to a shift in these taxa to a fly‐pollinated system.

研究了华凤仙(Impatiens chinensis L.)的花部特征和传粉系统,结果表明华凤仙单花期为2.3 d,雄性期是雌性期的6.8倍。花蜜量和花蜜可溶性糖含量分别为0.56 μL和28.2%。开花当天柱头即具有可授性,花粉活力在花朵刚开放时最高(95%),至花朵凋谢之前仍保持较高的活力(55%)。花粉胚珠比(5 730.3±2 941.1)和套袋实验的结果表明华凤仙属于自交亲和但以异交为主的繁育系统,传粉过程需要传粉者。蜜蜂、熊蜂、蝶类和几种芦蜂是华凤仙的主要访花者,最有效的传粉者为三条熊蜂。考氏无垫蜂作为盗蜜者,可能是除正常访花者之外,对华凤仙的有性繁殖影响最大的生物因素。  相似文献

BACKGROUND AND AIMS: The reproductive biology of Syngonanthus mucugensis and S. curralensis (Eriocaulaceae) was studied in areas of 'campo rupestre' vegetation in the Chapada Diamantina, north-eastern Brazil. These species are herbaceous and the individuals have a grouped distribution. Their leaves are united in a rosette, and their inflorescence is monoecious, of the capitulum type. The staminate and pistillate rings mature in a centripetal manner on the capitulum. METHODS: A field study was conducted, including observations concerning the morphology and biology of the flowers, fruit development, insect visits and anemophily, in both S. mucugensis and S. curralensis. Experimental pollinations were also carried out to study the mating systems of S. mucugensis. KEY RESULTS: Both species flower from June to August. The staminate cycle lasts approx. 7 d, and the pistillate cycle from 3 to 4 d, with no temporal overlap between them on the same capitulum. The pollen viability of S. mucugensis was 88.6%, and 92.5% for S. curralensis. The inflorescences of both species demonstrated ultraviolet absorbance, and a sweet odour was detected during both the staminate and pistillate phases. No nectar production was ever noted, although nectaries are present. Both species were visited by numerous groups of insects, with the Diptera being the principal pollinators, especially the species of Syrphidae and Bombyliidae. There were secondary pollinators among species of Coleoptera and Hymenoptera. There was no evidence of wind pollination. Syngonanthus mucugensis is a self-compatible species, and forms fruits by agamospermy at low frequencies. CONCLUSIONS: This is apparently the first report for pollination biology and mating systems of Eriocaulaceae. Conversely to that stated by some authors, entomophily, mainly effected by species of Diptera but also by species of Coleoptera and Hymenoptera, is probably the only pollination system in these species. In spite of the monoecious inflorescences without overlap of the staminate and pistillate phases, geitonogamy may occur in S. mucugensis, as the species is self-compatible and different capitula in the same plant at different phases is common.  相似文献

It has been suggested that plants that are good colonizers will generally have either an ability to self‐fertilize or a generalist pollination system. This prediction is based on the idea that these reproductive traits should confer resistance to Allee effects in founder populations and was tested using Gomphocarpus physocarpus (Asclepiadoideae: Apocynaceae), a species native to South Africa that is invasive in other parts of the world. We found no significant relationships between the size of G. physocarpus populations and various measures of pollination success (pollen deposition, pollen removal and pollen transfer efficiency) and fruit set. A breeding system experiment showed that plants in a South African population are genetically self‐incompatible and thus obligate outcrossers. Outcrossing is further enhanced by mechanical reconfiguration of removed pollinaria before the pollinia can be deposited. Self‐pollination is reduced when such reconfiguration exceeds the average duration of pollinator visits to a plant. Observations suggest that a wide variety of wasp species in the genera Belonogaster and Polistes (Vespidae) are the primary pollinators. We conclude that efficient pollination of plants in small founding populations, resulting from their generalist wasp‐pollination system, contributes in part to the colonizing success of G. physocarpus. The presence of similar wasps in other parts of the world has evidently facilitated the expansion of the range of this milkweed.  相似文献

应用荧光显微法和石蜡切片解剖法对‘晚大新高’梨授粉受精过程进行了系统观察研究。结果表明:‘晚大新高’梨自花授粉不结实;异花最佳授粉品种为‘黄花’,其次为‘翠冠’和‘丰水’。与选用‘黄花’为异花授粉品种相比,自花和异花的授粉受精过程存在明显差异,自花花粉在授粉后2h开始萌发,8h花粉管生长至离柱头约1/3处停止生长,顶端膨大呈球形,表现出自交不亲和性;异花花粉在授粉后1h开始部分萌发,8h花粉管生长至花柱中部,24h到达花柱基部并进入子房,48h进入胚囊,72h完成双受精过程。  相似文献

Anthecological observations of an orchidBletilla striata were carried out in Kobe, Japan. The purplish pink flowers were visited by 26 species of insects belonging to Hymenoptera, Diptera and Lepidoptera, although the flower did not offer food rewards of any kind. Candidates for pollination were seven species of aculeate Hymenoptera with a body size that well fitted the space in between the column and labellum. The most effective candidate was the male of the longhorn beeTetralonia nipponensis, judging from its abundance and legitimate intrafloral behavior. The female ofT. nipponensis and the other six species also contributed to promote out-crossing, but to a lesser extent than the maleT. nipponensis, since they occasionally exhibited illegitimate intrafloral behaviors for pollination and also, their flower visitation rate was lower.  相似文献

The pollination of Caesalpinia gilliesii (Fabaceae) , a legume species with long, brush-type flowers, was studied in wild populations in Argentina. A total of 341 hawkmoths (Sphingidae), belonging to 10 species, with tongues ranging from 1.5 to 13.5 cm in length, were captured by means of light traps. Hawkmoths could be classified in four significantly different tongue-length classes or guilds: short (15.35 ± 1.11 mm), medium (30.23 ± 2.24 mm), long (55.85 ± 4.98 mm) and very long (95.61 ± 13.63 mm). A total of 133 of the hawkmoths, belonging to seven species, carried pollen of Cae. gilliesii , mainly on their venter. Successful contact with anthers while drinking nectar depended upon hawkmoth tongue length. The relative frequency of pollen-carriage and the mean tongue length of each species were positively and significantly correlated. Considering a null hypothesis of nonspecialization, in which hawkmoths carrying pollen are distributed at random among individuals of the four guilds, long-tongued hawkmoths carried Cae. gilliesii pollen more frequently than would be expected by chance. In contrast, short- and medium-tongued guilds carried pollen less frequently than is predicted by random expectation. Individuals with short tongues accessed nectar from below without touching the anthers, whereas very long-tongued hawkmoths, with tongues longer than the length range of the flower reproductive organs, can probably reach nectar without touching the fertile organs. Results show that a pterotribic pattern of pollen deposition on the body of long-tongued hawkmoths is operating in this species.
