For a laboratory stirred-tank fermentor (STF) with foaming system of 0.5M sulfite solution containing an anionic soft detergent, the performing of a foam-breaking apparatus with a rotating disk (FARD)fitted to the STF was evaluated. The gas holdup in a mechanical foam-control system (MFS), i.e., the stirred-tank fermentor with the rotating disk foambreaker, was confirmed to be larger than that in a nonfoaming system (NS), i.e., the STF with an antifoam agent added. The agitation power in the mechanical foam control system was found to be smaller compared with the agitation power in the nonfoaming system, due to the increased gas holdup. Comparison of the oxygen absorption coefficient between the mechanical foam control system and the nonfoaming system in terms of the specific power input also demonstrated the superiority of the mechanical foam control system, not only in oxygen transfer performance but also in power input economy.  相似文献

A simple experimental diffusion controlled fermentor (DCF), coupled with the use of a mathematical model based on mass balance, is proposed to measure the variation of the gas (CO(2) and O(2)) diffusion coefficients in solid state fermentation. The DCF was packed with an ion-exchange resin impregnated with a nutritive medium and inoculated with Aspergillus niger. The growth conditions in the DCF were very similar to those found in equipment operated with convective oxygen supply. The diffusion coefficient was shown to be very dependent on the biomass concentration within the solid state fermentor, and attained values of less than 5% of the molecular diffusion in air when the biomass in the fermentor reached 27 mg dry/g dry support.  相似文献

Methods for the selection of transfectoma cells that express large quantities of mouse-human chimeric antibodies have been develped. SP2/0 mouse myeloma cells were transfected with pSV2-gpt and pSV2-neo based immunoglobulin expression vectors. Double transfectants were selected using the xanthine-guanine phosphoribosyl transferase (gpt)and the neomycin (neo) selection marker genes. ELISA-based screening of transfectoma clones resulted in the isolation of IgG-producing transfectomas. Introduction of the kappa light-chain 3'-enhancer into the light-chain expression vector significantly increased immunoglobulin expression, but only when the enhancer was located at its physiological site, 9 kb downstream of the kappa constant region exon. With some of the transfectomas, final yields of up to 80 mg/L of chimeric IgG were obtained in conventional flask cultures using serum-free growth medium. A pilot-scale AcuSyst Maximizer hollow fiber cell culture system was used for the production of gram amounts of chimeric IgG. Results obtained with different transfectoma clones in conventional culture were not fully predictive for yields in the hollow fiber system. In contrast, differences in productivity between individual clones in the laboratory-scale Tecnomouse cell culture unit were comparable with those in the Maximizer system. Approaches to increasing the productivity of benzophenanthridine alkaloids in suspension cultures in Escherichia californica were made in an airlift fermentor under different culture conditions. Elicitation with yeast extract elicitor reduced the time required to obtain a certain amount of alkaloid production. In a two-phase airlift fermentor with compounded silicone fluid, total alkaloid concentration in silicone fluid was 153.1 mg/L and that in the aqueous cellular phase was 8.2 mg/L at day 21 from inoculation. The large accumulation capacity of silicone fluid made it possible to store correspondingly large amounts of total alkaloid and increased the alkaloid production. Act day 21 from inoculation, the volumetric alkaloid productivity and the netproduction in a two-phase airlift fermentor were 1.4 and 1.5 times higher than those of normal airlift fermentor operation. This performance was furthermore enhanced by elicitation. Elicitation in two-phase airlift fermentor operation increased the volumetric productivity and the new production 3.3- and 3.5-fold compared to those of normal airlift fermentor operation. 研究了中试厌氧氨氧化(Anaerobic ammonium oxidation,Anammox)反应器的启动性能。结果表明,以硝化反硝化污泥、短程硝化污泥、厌氧絮体污泥和厌氧颗粒污泥混合接种,经过255d的运行,可在常温下(5oC~27oC)成功启动中试Anammox反应器,反应器的基质氮去除速率可达1.30kg/(m3·d)。厌氧氨氧化是致碱反应,厌氧氨氧化成为反应器内的主导反应后,进水pH宜控制在厌氧氨氧化适宜范围的偏低水平(6.8左右)。亚硝酸盐既是Anammox菌的基质,也是抑制剂,控制进水亚硝酸盐浓度(13~36mg/L)有助于厌氧氨氧化反应。菌种是生物反应器的功能之源,向中试装置投加少量厌氧氨氧化污泥(投加比2%),可大大加速中试Anammox反应器的启动进程。  相似文献

A calculation method has been developed to model the statistical transport of biological particles in bubble-driven flows, with special reference to the biokinetics of environmental excursions experienced by individual cells, aggregated cells, or immobilization beads in airlift bioreactors. Interim developments on modeling the transport of such particles in concentric tube devices are reported. The calculation is driven by user-prescribed global parameters for the bioreactor geometry, bulk air flow rate, and particle parameters (size and slip speed). The algorithm calls on empirical data correlations for void fraction, bulk liquid flow rate, and bubble sizes and slip speeds, optimally selected from a large bibliographic database. The Monte Carlo algorithm concentrates on simulating particle transport in the bubbly riser flows.The packaged family of correlations and calculations represents, in effect, an expert system augmented by a transport simulation suited to characterizing the biokinetic response of cells cultured in airlift bioreactors.  相似文献

Energy costs of biomass synthesis are relatively higher at low than at high specific growth rates () because of an increased protein content of the cell and increased costs of protein synthesis as such at low values. A comparison of aerobic, glucose limited cultures of Bacillus licheniformis in a chemostat and in a partial-recycling fermentor indicated that pulse-wise nutrient addition increased the maintenance energy demand (m). In the chemostat experiments, we also found a striking deviation from linearity between substrate consumption and , with large implications for the maintenance coefficient. The deviation is mainly due to a large shift in metabolic carbon flows at specific growth rates between 50 and 100% of max. At those growth rates, uncoupled growth occurs, presumably as a necessary condition for faster growth, since uncoupling results in a faster energysupply for biosynthetic purposes.The maintenance coefficient as determined by chemostat studies should be regarded as a compounds parameter, constituted of maintenance energy demands like ppGpp accumulation, variable costs of mRNA and protein accumulation, kinetic proofreading etc. and influenced by fermentor operation parameters like the substrate addition rate; moreover, both constancy of m and a linear relation between m and appear quite unlikely.  相似文献

An efficient and economical medium--Thermotoga maritima basal medium (TMB)--was designed for the cultivation of T. maritima under either liquid or solid conditions. When the broth was flushed with N2 or CO2 throughout cell growth in a 10-L fermentor (pH controlled to 6.5), the maximum cell density (OD600) on TMB containing 1% glucose rose to 2.0 or higher (1.63 x 10(9) cells mL(-1)). Sheath-less cells observed by electron microscopy were captured during growth in the fermentor. Using a two-layer plating method, isolated single-well colonies were consistently obtained within 24 h on the TMB in modified tissue culture flasks. The minimal inhibitory chloramphenicol concentrations for T. maritima on TMB agar were 5 microg mL(-1) after 24 h and 48 h, and 25 microg mL(-1) at 72 h.  相似文献

A process for production of mold lactase was developed. Tests were carried out in pilot and industrial scale with an Aspergillus niger strain selected after screening a number of molds.A computer coupled autoanalyzer system was used for monitoring enzyme formation in the pilot fermentor. Lactase production was investigated using different pH- and temperature-profiles. A. niger lactase has an acid pH optimum, a high temperature optimum and good stability. It does not require any metal ions. It is suitable for immobilization for hydrolysis of lactose in acid whey.Three-fold enhancement of lactase production was obtained by mutagenizing A. niger using NTG as mutagenic agent.The lactases produced by the mutants have the same pH and temperature optima and stability but the growth properties of the mutants were different from those of the original strain.Sufficient specific activity of the enzyme preparation for immobilization was obtained by purifying the enzyme by selective adsorption on Na-Ca-silicate.  相似文献
