Developmental and genotypic differences in the response of pea stem segments to auxin 总被引:1,自引:0,他引:1
The objective of this investigation was to examine the response to exogenous auxin (indole-3-acetic acid; IAA)of stem segments at two developmental stages. The standard auxin response of excised stem segments and intact plants consists of an initial growth response and a prolonged growth response. We found that this biphasic response does not occur in internodes at very early stages. Stem segments of light grown pea of various genotypes were cut when the fourth internode was at 6–13% of full expansion (early-expansion) or at 18–25% of full expansion (mid-expansion). Length measurements of excised segments were made after 48 hours of incubation on buffer with or without auxin. An angular position transducer linked to a computerized data collection system provided high-resolution measurement of growth of stacks of segments incubated in buffer over 20 hours. Early-expansion segments of all genotypes deviated from the standard auxin response, while mid-expansion segments responded in a manner consistent with previous reports. Early-expansion segments of tall, light-grown plants were unique in showing an auxin-induced inhibition of growth. The auxin-induced inhibition correlated with high endogenous auxin content, as determined by HPLC and GC/MS, across genotypes and between early-expansion and mid-expansion segments of tall plants. Measurement of ethylene evolved from stem segments in response to auxin, and treatment of segments with the ethylene action inhibitor, norbornadiene, showed the inhibition to be mediated in part by heightened ethylene sensitivity. Growth of early-expansion segments of dwarf and severe dwarf plants was stimulated by exogenous auxin, but the growth rate increase was delayed compared to that in mid-expansion segments. This is the first time that such a growth response, termed the delayed growth response has been emonstrated. Morphological and hybridization experiments were performed on Caloglossa leprieurii (Montagne) J. Agardh collected from Japan, Singapore and Australia in order to evaluate taxonomic characters of this species. Within C. leprieurii at least four mating groups were recognized from the Indo-Pacific region. These mating groups could be characterized by the blade width at the inter-node and the cell-row numbers on the opposite side derived from the first axial cell at the main axis, though these properties showed a certain variability even in the same plant under both field and culture conditions. The phylogenetic relationship and geographic distribution of the four mating groups are discussed. 相似文献

The three-dimensional structure ofDolichos biflorus seed lectin has been constructed using five legume lectins for which high resolution crystal structures were available. The validity of the resulting model has been thoroughly investigated. Final structure optimization was conducted for the lectin complexed with GalNAc, providing thereby the first three-dimensional structure of lectin/GalNAc complex. The role of theN-acetyl group was clearly evidenced by the occurrence of a strong hydrogen bond between the protein and the carbonyl oxygen of the carbohydrate and by hydrophobic interaction between the methyl group and aromatic amino acids. Since the lectin specificity is maximum for the Forssman disaccharide GalNAc(1–3)GalNAc-O-Me and the blood group A trisaccharide GalNAc(1–3)[Fuc(1–2)]Gal-O-Me, the complexes with these oligosaccharides have been also modelled. 相似文献

The carbohydrate binding properties of theDolichos biflorus seed lectin and DB58, a vegetative tissue lectin from this plant, were compared using two types of solid phase assays. Both lectins bind to hog blood group A + H substance covalently coupled to Sepharose 4B and this binding can be inhibited with free blood group A + H substance. However, the binding of the seed lectin is inhibited byD-GalNAc whereas DB58 binding was not inhbited by any monosaccharide tested, thus suggesting that its carbohydrate combining site may be more extensive than that of the seed lectin. The activities of these two lectins also differ from one another in ability to recognize blood group A + H substance adsorbed on to plastic and in the effects of salt and urea on their carbohydrate binding activities. Salt tolerance of pigeon pea (Cajanus cajan (L.) Millsp.) was determined at three growth stages following observations by a number of workers that degree of salt tolerance of different crops varies with their ontogeny. The salt tolerance of three accessions, Local arhar, ICPL-151 and ICPL-850014 of pigeon pea was assessed at the germination, seedling and adult stages. There was no positive correlation between tolerance at the early growth stages and at the adult stage since no clear difference in salt tolerance of the three accessions was observed at the germination and the seedling stages, whereas accessions differed considerably at the adult stage. Although increasing salt concentrations adversely affected the growth of all three accessions, ICPL-151 was superior to the other two accessions in fresh and dry biomass, yield and yield components when tested at the adult stage. The tolerant accession, ICPL 151, accumulated significantly lower Na+and CI in shoots. By contrast it was higher in shoot and root K+, K/Na ratios, K vs Na selectivity, soluble sugars, free amino acids and proline compared with the other two accessions. 相似文献

The shrews of the Sorex araneus group have undergone a spectacular chromosome evolution. The karyotype of Sorex granarius is generally considered ancestral to those of Sorex coronatus and S. araneus. However, a sequence of 777 base pairs of the cytochrome b gene of the mitochondrial DNA (mtDNA) produces a quite different picture: S. granarius is closely related to the populations of S. araneus from the Pyrenees and from the northwestern Alps, whereas S. coronatus and S. araneus from Italy and the southern Alps represent two well-separated lineages. It is suggested that mtDNA and chromosomal evolution are in this case largely independant processes. Whereas mtDNA haplotypes are closely linked to the geographical history of the populations, chromosomal mutations were probably transmitted from one population to another. Available data suggest that the impressive chromosome polymorphism of this group is quite a recent phenomenon. 相似文献

Summary This paper describes the food recruitment strategy of the antTetramorium bicarinatum, at both the individual and collective levels. The general organisation of recruitment used by this species during the exploitation of sucrose solutions shows similarities with group recruitment described for other species. However, our experiments demonstrate that inT. bicarinatum, the first trail laid by a recruiter during its return trip to the nest is more efficient than in other species using group recruitment, for exampleTetramorium impurum. Moreover, the efficiency of the first trail of aT. bicarinatum recruiter is comparable with that ofTapinoma erraticum, a species that uses mass recruitment. Despite the efficiency of the trail, choice experiments show that the recruited workers prefer to follow the leader rather than the first trail, suggesting the emission of a more attractive signal by the leader on its way back to the food. The function of the leader in this strategy is discussed in terms of collective decisions. 相似文献
